    Notizen: A microcomputer-based facility is described that permits the data required for three-dimensional reconstructions to be collected quickly and inexpensively from serial sections. The facility consists of a microcomputer, a digitizer tablet, a graphics terminal, a printer, a plotter, and telephone coupler. Images of serial sections are superimposed on the digitizer tablet. Contours of interest on each section are digitized and the coordinates are stored on “floppy” disks. The problems of putting successive sections in correct register and of taking into account magnification factors are discussed briefly. Use of the facility for high-resolution applications is also considered.

    Notizen: Plasmodium berghei-infected murine red cells possess protein kinase activity that is associated with the isolated parasites. Schizonts contain significantly higher levels of this protein kinase than the more immature forms, suggesting a relationship between this enzyme activity and parasite development. Partially purified protein kinase has a Km for ATP of ∼30 μM, whereas the Km for GTP is ∼300 μM and the substrate preference is phosvitin 〉 casein 〉 〉 histone 〉 protamine. The Mg2+ optimum is 10-20 mM, and the protein kinase activity is stimulated by the polyamines spermine and spermidine. The flavone, quercetin, inhibits the protein kinase activity in a competitive manner with' respect to ATP (Ki ∼3 μM), and P chabaudi also has a very similarly regulated protein kinase. Protein kinases from both species are very similar to the type I casein kinase.

    Notizen: Four detergents have been compared for identification of the Plasmodium knowlesi variant antigen on infected erythrocytes by immunoprecipitation analysis. Erythrocytes infected with late trophozoite and schizont forms of cloned asexual parasites were labeled by lactoperoxidase-catalyzed radioiodination and extracted either with the anionic detergents sodium dodecyl sulfate (SDS) or cholate, the neutral detergent Triton X-100, or the zwitterion 3-[(3-cholamidopropyl)di-methylammonio]-1-propane sulfonate (CHAPS). After addition of Triton X-100 to SDS and cholate extracts, parallel immunoprecipitations of the four extracts were performed using rhesus monkey antisera of defined agglutinability. Identical results were obtained with clone Pkl(A+ ), which has 125I-variant antigens of Mr 210,000 and 190,000, and with clone Pkl(B+)l+, which hasvariant antigens of Mr 200,000-205,000. SDS yielded maximal levels of immunoprecipitated 125I-variant antigens. Variant-specific immunoprecipitation was detected in some experiments with Triton X-100 and cholic acid but with significantly lower recovery than with SDS. CHAPS extraction did not yield the variant antigens on immunoprecipitation. The variant antigens could also be identified in Triton X-100-insoluble material by subsequent extraction with SDS, indicating that failure to recover these proteins in the Triton X-100-soluble fraction is due to failure of this detergent to extract the variant antigens rather than to degradation during extraction. We suggest that the 125I-variant antigens either have a structure that renders them intrinsically insoluble in Triton X-100, cholate, or CHAPS, or that they are associated in some way with host cell membrane components that also resist solubilization by these detergents.

    Notizen: To determine the role of thrombin high-affinity receptor occupancy and enzymic activity in thrombin initiation of cell proliferation, we have utilized thrombin derivatives which separate these functions. We previously showed that enzymically active γ-thrombin stimulates ion fluxes without binding to high-affinity sites, whereas proteolytically inhibited DIP-α-thrombin which binds to high-affinity receptors does not. Since neither derivative initiates DNA synthesis by itself, this suggested that two separate sequences of events might be necessary for a complete initiation signal. We now report that the combination of DIP-α-thrombin and γ-thrombin initiate DNA synthesis and cell proliferation to levels approaching the maximal initiation by native α-thrombin. This combinatory effect is dose-dependent for both γ-thrombin and DIP-α-thrombin in the same concentration range as α-thrombin alone. Thus, these same concentrations of α-thrombin alone may be required to initiate each sequence of events. The combinatory stimulation could be achieved even if the derivatives were added individually up to 8 hr apart. Moreover, preincubation with either derivative shortened the lag period for initiation of DNA synthesis by native α-thrombin. These results indicate that both receptor occupancy and enzymic activity are necessary for thrombin initiation of cell proliferation and that each action initiates a sequence of early events which moves the cell forward toward entry into a proliferative cycle.

    Notizen: The available descriptions of the development of sympathetic innervation of the chick heart conflict with the known sympathetic innervation of the adult chicken heart. The adult heart is innervated by bilateral sympathetic cardiac nerves originating from the first thoracic sympathetic ganglia. These nerves travel lateral and anterior to the lung and join the vagi just before entering the pericardium along the great vessels. Using catecholamine histofluorescence techniques and silver preparations, we have observed the development of the sympathetic cardiac nerves. The sympathetic cardiac nerves arise from the first thoracic sympathetic ganglia on the 7th day of incubation. They grow lateral and then ventral to the developing lungs to join the vagi, and are found in the bulbar region of the heart and atrium on the 10th day of incubation. Fluorescent cells without processes mark the course of the sympathetic cardiac nerves and are present in the bulbar region on the 10th day and thereafter. Sympathetic ganglion cells lose their fluorescence between day 8 and day 16 of incubation. This is presumably due to dilution of the transmitter in the rapidly increasing volume of cytoplasm in the sprouting neurons. Small intensely fluorescent (SIF) and adrenal medullary cells do not undergo a diminution of fluorescence during this period. SIF cells appear well differentiated at 16 days.

    Notizen: Yolk sacs from dogs at 40, 50, and 60 days of gestation were examined by electron microscopy. Free ribosomes, mitochondria, and rough endoplasmic reticulum (rER) are more prominent in both endoderm and mesothelium at 40 and 50 days than at 60 days, suggesting a greater synthetic capacity at the earlier stages. Smooth endoplasmic reticulum (sER) and glycogen are also present in greater amounts in the endoderm in the earlier stages. In the mesothelium, however, low amounts of sER and glycogen are consistently present. Certain possibilities relative to the nature of the synthetic activities in these two tissues are discussed. Large amounts of smooth-surfaced vesicles were observed along the basal edges of the 60-day mesothelium; they are indicative of transport processes occurring at this time. As gestation proceeds, in both endoderm and mesothelium, the Golgi complex remains well developed, there are more numerous lysosomelike bodies, and bundles of intermediate filaments either increase or become more diffused. In some endoderm cells at 60 days, large vacuoles and dense glycogen deposits were noted. These observations indicate that degenerative processes are gradually occurring in the endoderm and mesothelium as parturition draws near. Erythropoiesis occurs in the mesenchyme at 40 and 50 days. At 40 days also, segments of endothelium were seen within blood islands, indicating that the endothelial lining of some yolk sac vessels differentiates from cells located in the interior of such islands.

    Notizen: Frog skin contains three distinct types of exocrine glands: granular (poison), mucous, and seromucous. The granular gland forms a syncytial secretory compartment within the acinus, which is surrounded by smooth muscle cells. The mucous and seromucous glands are easily identifiable as distinct glands. The serous and mucous secretory cells are arranged in a semilunar configuration opposite the ductal end and are filled with granules. Within the acinus, located at the ductal pole of the gland, are distinct groups of cells with few or no granules in the cytoplasm. In both the mucous and seromucous gland there is a cell type with abundant mitochondria; the one in the mucous gland is located in the region adjacent to the secretory cells. The duct of these glands is two-layered, with the individual cells appearing morphologically similar to the layers of the skin epithelium as the duct traverses the skin. The duct appears to be patent throughout its length. The morphological heterogeneity and distinct distribution of the cell types within the gland acinus may be indicative of a functional heterogeneity that allows the production of distinctly different types of secretion from the same gland type, depending on the type of stimulus.

    Notizen: The env gene of Rous sarcoma virus codes for two glycoproteins which are located on the surface of infectious virions. Subcloning of these coding sequences in the place of the late region of SV40 DNA has allowed the expression of a normally glycosylated, functionally active glycoprotein complex on the surface of monkey cells. Through the use of site-directed mutagenesis, the role of specific amino acids in the signal peptide, signal peptidase cleavage site, and membrane anchor region have been investigated. Amino-terminal mutations have shown that deletion of the signal peptidase cleavage site along with one or two amino acids of the hydrophobic signal peptide results in the synthesis of an unglycosylated. uncleaved, and presumably cytoplasmically located precursor. Nevertheless, changing the signal peptidase cleavage site from ala/asp to ala/asn does not block the translocation of the glycoprotein across the membrane or the action of the peptidase. At the other end of the molecule, carboxy-terminal mutations have shown that the deletion of the hydrophobic membrane anchor region is not sufficient for the secretion of the truncated glycoprotein. Interpretations of these results based on recent models for protein transport and secretion are discussed.
