    Notizen: Abstract: GTP and GDP decreased the saturable binding of [3H]baclofen or [3H]γ-aminobutyric acid ([3H]GABA) to GABAB but not GABAA receptors whereas GMP displayed negligible activity. This effect was specific to guanyl nucleotides and was not mimicked by high concentrations of ATP. The inhibition of ligand binding was the result of a diminished receptor affinity with no change in receptor number. The use of a complete physiological saline solution rather than Tris buffer plus Ca2+ or Mg2+ increased the potency of GTP at the GABAB receptor. The results are discussed in relation to the effects of GABA and GTP on adenylate cyclase activity in the brain.

    Notizen: Following bronchial provocation tests with inhalent allergens, late asthmatic reactions (LAR) frequently follow immediate asthma reactions (IAR). This study of atopic asthmatic children demonstrated that patients could show IARs to one allergen, ryegrass extract, but isolated LARs without any preceding immediate response when challenged with an unrelated allergen extract of D. pteronyssinus. The patients’ degree of skirt sensitivity to the concentration of extract inhaled appeared one factor which determined whether an isolated LAR or IAR followed allergen inhalation,

    Notizen: The movement of chloride ion and the movement and degradation of fluometuron in two soils of contrasting texture and structure was predicted with a simulation model which included terms for the degradation of fluometuron as a function of temperature and moisture. The simulated distributions of soil water, chloride ion and fluometuron were compared with the observations reported in a previous paper (Hance, Embling, Hill, Graham-Boyce & Nicholls, 1981), Soil waler contents and fluometuron distribution were simulated reasonably accurately. Movement of chloride to below 30 cm in both soils and degradation of fluometuron in the WRO sandy loam soil were not well simulated. The discrepancies between observed and predicted water contents and chloride distribution show the importance of measuring dynamic as well as static water properties. Therefore refined simulation models may need to account for pore continuity, soil particle movement and mobile and immobile categories of water.

    Notizen: Immunoglobulin E (IgE), directed against components of Acacia (wattle) pollen, has been detected by radioallergosorbent tests (RAST) in the sera of some children and adults who develop allergic symptoms in the presence of flowering Acacia trees in Australia. All these subjects also had high levels of IgE directed against Lolium perenne (rye grass) pollen. Inhibition by RAST showed that most of the IgE molecules which bound to Acacia pollen components also bound to L. perenne pollen extracts, and to Glycoprotein I, the major allergen of L. perenne pollen. In these assays, the allergens have been immobilized on polyvinyl chloride microtitre trays: the sensitivity of this approach is compared to that of commercial RAST kits.

    Notizen: Eight children with severe, generalized, childhood eczema, uncontrolled by standard dermatological treatment, underwent a period of dietary antigen-avoidance, using an elemental based diet. For the group, there was statistically significant improvement in eczema during treatment with the elemental diet, and relapse of eczema followed within 6 weeks of returning to a normal diet. The results suggest dietary factors are important in childhood eczema.

    Notizen: Lymphocyte stimulation, as determined by incorporation of thymidine, to rye grass extract in twenty-three children with bronchial reactivity to rye grass and to house dust mite, did not differ significantly from four children with reactivity to house dust mite alone, or from nine children with asthma but without a bronchial response to these allergens. Sixteen children underwent hyposensitization with rye grass extract or treatment with placebo. There was no consistent effect of hyposensitization on the lymphocyte stimulation indices to rye grass. A decrease in lymphocyte responsiveness occurred to rye grass and to house dust mite after the grass pollen season but was not statistically significant. Analysis of changes in lymphocyte responsiveness to both house dust mite and rye grass of the children most highly sensitized to rye allergen, showed that the lymphocyte responsiveness to rye grass fell during the pollen season (P〈0.05) but this effect was not seen with house dust mite. The study suggests that a decrease in lymphocyte responsiveness to rye grass allergen in children with large amounts of anti-rye IgE antibodies is antigen specific and may be seen following seasonal exposure.

    Notizen: The specificity of immunoglobulin E for different cereal grain proteins was investigated using sera from twenty paedialric patients with asthma and/or eczema. Close correlations were observed between radioallergosorbent test values for grain extracts of wheat, rye and barley, and, to a lesser extent, oats. Of the different wheat flour fractions tested, the globulins and glutenins consistently bound higher levels of IgE than the gliadins and albumins. This is in contrast both with bakers' asthma (an allergy to inhaled flour where the albumins are important allergens) and with coeliac disease (in which gliadin is the most toxic fraction). Partial digestion of the flour proteins largely removed their ability to bind IgE, An analytical technique of identifying allergens after gel isoelectric focusing demonstrated that many different flour proteins were involved.

    Notizen: The levels of IgE antibodies to inhaled and dietary antigens and total serum IgG, IgA, IgM and IgE were studied in two highly selected populations of children who had either severe asthma and no history of eczema, or severe asthma and generalized eczema. These populations were age-matched with a healthy control population. The results showed that the eczematous children had significantly higher levels of IgE antibodies to common environmental antigens than the non-eczematous patients with severe asthma. Both patient groups had significantly higher IgE antigen specific antibody levels than the control population to these antigens. Minor differences in total IgG, IgA and IgM levels were noted between the groups. The variations in total IgE levels between the groups showed the same pattern as for specific IgE antibodies.
