    Description / Table of Contents: Zusammenfassung Zur Bewertung der Laboratoriumsschnelldiagnose der anaeroben pyogenen Infektion haben wir die Ergebnisse von Gram-Färbungen ultravioletter Fluoreszenz und Gaschromatographie direkt an Eiter ausgeführt, mit denen der Anaerobierkultur verglichen. Die Fluoreszenz ging am schnellsten, aber es gab viele negative Ergebnisse, wennBacteroides melaninogenicus nicht vorlag. Gaschromatographie erwies sich als schnell und empfindlich, aber es gab einige falsch negative Resultate, häufig beiBacteroides fragilis-Monoinfektion, und auch einige falsch positive. Die Gram-Färbung stellt ebenfalls ein Schnellverfahren dar, sie allein erwies sich jedoch nur dann als hilfreich, wenn zahlreiche Erreger von gemischter oder charakteristischer Morphologie vorlagen. Die drei Methoden zusammen lieferten fast ausnahmslos eine verläßliche und schnelle vorläufige Diagnose der anaeroben pyogenen Infektion.
    Notes: Summary In order to assess the rapid laboratory diagnosis of anaerobic pyogenic infection, we compared the results of Gram stains, ultra-violet fluorescence and gas chromatography, all performed directly on pus, with those of anaerobic culture. Fluorescence was most rapid but there were many negatives unlessBacteroides melaninogenicus was present. Gas chromatography was rapid and sensitive but there were some false negatives, often in pureBacteroides fragilis infection, and a few false positives. Gram-staining was also rapid, but only helpful on its own when there were large numbers of organisms of mixed or characteristic morphology. The three methods together almost always provided a reliable and rapid presumptive diagnosis of anaerobic pyogenic infection.

    Notes: Abstract The turnover of myelin and of myelin protein fractions has been measured in the central nervous system of rats who were placed on a hyperphenylalaninaemia-inducing diet (3%L-phenylalanine and 0.12%p-chlorophenylalanine added to the normal laboratory chow) when they were 25 days of age. A considerably increased turnover of the fast component of myelin and of myelin protein fractions was observed, which was not found in weight-matched controls or in controls fed the normal laboratory chow supplemented with 0.12%p-chlorophenylalanine. The increased turnover is therefore due to the hyperphenylalaninaemic condition and not due to the slow-down in growth or the presence ofp-chlorophenylalanine. Furthermore, an inhibition of myelin synthesis due to the hyperphenylalaninaemic condition has been observed. Since these effects on myelin metabolism can be demonstrated to occur even when the brain has matured considerably, prudence should be exercised in considering the termination of the dietary treatment of patients with phenylketonuria.

    Notes: Summary The kinetic behaviour of heavy meromyosin (HMM) and subfragment-1 (S-1) has been compared for a range of pH and temperature for normal and SH-1 modified proteins. The rate of step 4 was measured by decay of tryptophan fluorescence, proton release and phosphate release measured as an increase in conductance. The temperature dependence curves fork 4 andk 5 cross over at 11–13° C for Mn2+-ATPase, 3–5° C for SH-1 modified Mg2+-ATPase and below 0° C for normal Mg2+-ATPase. A change of the rate-determining step fromk 4 tok 5 accounts for the nonlinear Arrhenius plots of Mn2+-ATPase and SH-1 modified Mg2+-ATPase. Modification also reduced the rate of the hydrolytic stepk 3 measured by the maximum rate of the phosphate early burst. The two fluorescence transitions (steps 2 and 3) are easily resolved for the modified enzyme andk 3 measured by fluorescence was equal to the rate of hydrolysis obtained from phosphate measurements.

    Notes: Summary 1. Adult male and female freshwater crayfish, acclimated to 15°C, were observed to migrate from hypoxic water into air at a mean $$P_{O_2 } $$ of 42 mm Hg. 2. The rates of recovery after initial disturbance of heart rate (f card), ventilation rate (f resp), oxygen consumption $$\dot M_{O_2 } $$ and ventilation volumeV W were followed for 18 h. Settled values were established after 3–5 h of submersion in normoxic water at 15°C. 3. On exposure to air at 15°C (R.H. 70–80%) for 3 h,f card,f resp and $$\dot M_{O_2 } $$ were similar to settled submerged values. Ventilation volume in airV air was only 5% of normoxicV W. 4. In air the crayfish experienced internal hypoxia with postbranchial oxygen tension ( $$Pa_{O_2 } $$ ) reduced to 33% and oxygen content ( $$Ca_{O_2 } $$ ) to 45% of the submerged normoxic level. $$\dot M_{O_2 } $$ was maintained by drawing on a venous reserve of oxygen, reducing postbranchial oxygen content ( $$Cv_{O_2 } $$ ) in air to 15% of the submerged normoxic level. 5. After 3 h in air the crayfish experienced a marked acidosis of the haemolymph which was both respiratory, due to the accumulation of CO2 and metabolic, due to the accumulation of lactate. 6. On return to water after 3 h in air,f card,f resp, $$\dot M_{O_2 } $$ andV W quickly returned to their settled normoxic values. There was evidence of an initial period of hyperventilation, but no repayment of an oxygen debt. 7. The effectiveness of respiratory gas exchange in water and in air are compared with other aquatic and terrestrial species.
