    Notes: Summary A simple model of the distribution and metabolism of insulin in vivo has been evaluated using data from insulin infusion into a group of normal subjects. The major rate-limiting step for access to degradation pathways is assumed to consist of binding of the ligand to a single population of insulin receptor sites, except that provision is made for the possibility of linear non-receptor-mediated degradation and for the phenomenon of negative cooperativity. The model has been shown to accommodate the non-linearity of insulin metabo lism, allows evaluation of receptor association and dissociation constants and provides for the first time an estimate of total accessible receptor number in the intact organism. For normal fasting man the model predicts 1.00±0.05 nmol accessible binding sites/kg (mean±SD).

    Notes: Summary In two contrasting types of S5 inbred Lolium perenne a correlation has been found between the distribution pattern of chiasmata within bivalents and that of meiotic U-type exchange. The relationship between the two processes is discussed, in terms of inbreeding effects upon chiasma redistribution, and mention is also made of some other types of chromosome breakages which arise following enforced self-pollination.

    Notes: Summary An analysis of the patterns of chiasma distribution within bivalents, in a number of inbred lines of Lolium perenne, reveals a striking potential for variation in this character, as well as a polygenic genetic basis for its control. In parent plants the chiasmata have a regular pattern of localisation which restricts their formation to the ends of the chromosome arms: this strict localisation is progressively lost with inbreeding and novel patterns of distribution are found where chiasmata occur in more interstitial and proximal regions. The variation is continuous and originates from genotypic differences between the different parent plants, rather than from segregation among families deriving from individual parents.

    Notes: Summary A number of inbred lines of Lolium perenne have been developed as far as the fifth inbred generation and are being used for basic studies on the cytogenetics of the species. An analysis on variation in the pattern of chiasma frequency and distribution, including the parent plants and all generations down to the S5, reveals that the effect of inbreeding is to reduce chiasma frequency and to increase both the cell and bivalent variances. Evidence is presented for a genetic basis of polygenic control of chiasma formation and distribution, and a model is suggested for control over the three related components of variation which involves a two-stage level of regulation.

    Notes: Abstract Single input, single output experiments can result in nonunique solutions for the rate constants of a linear compartmental model used to describe the pharmacokinetics. Where a finite number of solutions exists, a priori knowledge has to be used to distinguish between the solutions. Where there is an infinite number of solutions, assumptions have to be made about the values of some rate constants in order to obtain a unique solution for the others. This paper considers such experiments and determines whether either the addition of an extra input (simultaneously with the first input) or the taking of an extra measurement would result in a unique solution. It is found that perturbing a second input can be useful, but only if the perturbation is of different shape from the first input. Measurements of drug in urine and metabolite in plasma are generally not helpful in resolving identifiability of the drug dynamic model. If a radioactive tracer is used, though, the second measurement (for example, by externally scanning the radioactivity of the liver) can prove useful, but only if the gain of the measuring device is known.

    Notes: Human leucocytes incubated in tissue culture fluid of low-sodium concentration (2 mM; iso-osmolarity maintained with choline chloride) reached a new equlibrium within 1 hour and lost approximately 25% of intracellular potassium and 70% of intracellular sodium. The rate constant for ouabainsensitive sodium efflux fell by more than 50% and the ouabain-insensitive rate constant increased nearly threefold in the low-sodium medium. Total sodium efflux fell in proportion to internal sodium whereas ouabain-insensitive sodium efflux remained unchanged. A reduction in external sodium from 140 to 2 mM was associated with a 75% fall in sodium influx. In the low-sodium medium ouabainsensitive potassium influx exceeded ouabain-sensitive sodium efflux and no ouabain-sensitive potassium efflux could be demonstrated. Ouabain-insensitive potassium influx and that portion of potassium efflux which is dependent on external potassium fell in parallel in low-sodium cells, suggesting reduced activity of a ouabain-insensitive K:K exchange system.
