Summary The ultrastructure of uredia, intercellular and intracellular hyphae ofUromyces viciae-fabae onVicia faba has been studied by scanning electron microscopy of frozen-hydrated material. Frozen-hydrated leaves will readily fracture and can be viewed on the cold stage of the microscope at liquid nitrogen temperatures for several hours without visible surface changes occurring. Both fungal and host-plant cells show a high degree of morphological preservation with little or no loss of cell contents and with minimal signs of shrinkage. Intracellular hyphae may be filamentous and branched or short and lobed. They are constricted at the point of entry into the host cell. Intercellular matrix material is well preserved around developing pedicels and urediospores and may act as a lubricant. The technique described is very quick compared with conventional preparatory procedures.
