    Notes: Abstract: β-Phenylethylamine (PEA) was characterized as a substrate for type A and type B monoamine oxidase (MAO) in brain and liver mitochondria of eight species at different substrate concentrations. In all species, at 10.0 μM, PEA was almost specific for type B MAO. At 1000 μM, however, the amine was common for both types of MAO in rat brain and liver, human brain and liver, mouse brain, guinea pig brain and liver, and bovine brain, while it was specific for type B MAO in mouse liver, rabbit brain and liver, bovine liver, pig brain and liver, and chicken brain and liver. From the present study, when PEA is used as a type B substrate, it is recommended that the substrate concentration should be sufficiently low to avoid the effects of species and tissue differences.

    Notes: Summary 4-Methoxyphenylethylamine was found to be a specific substrate for type B monoamine oxidase (MAO) in rat brain mitochondria, whereas 3,4-dimethoxyphenylethylamine was common for both types of MAO. These results suggest that O-methylation in the para-position increases the preference of the substrate for type B MAO, while a methoxy-group in the meta-position contributes to the substance being a type A substrate.

    Description / Table of Contents: Zusammenfassung Es wird eine einfache, qualitative Methode zur Identifizierung von Spermaspuren beschrieben, die auf der hohen γ-Glutamyltransferase-Aktivität (γ-GTP) des menschlichen Sperma basiert. Dabei wird die Abspaltung von α-Naphthylamin von N-γ-Glutamyl-α-naphthylamid durch das Enzym benutzt, wobei α-Naphthylamin mit Fast Garnet GBC-Salz gekoppelt eine tief braunrote Farbe bildet. Die Ergebnisse der Untersuchung bestätigen die Einfachheit, Spezifität und Stabilität dieser Methode als Vortest zur forensischen Untersuchung von Spermaspuren.
    Notes: Summary A simple qualitative method for identification of seminal stains based on a high activity of γ-glutamyltransferase (γ-GTP) in human semen is described. It employs the release of α-naphthylamine from N-γ-glutamyl-α-naphthylamide by the γ-GTP action; α-naphthylamine couples with Fast Garnet GBC salt to produce a strong brownish-red color. The data on its simplicity, specificity, and stability show that the present method is suitable for medicolegal examination of seminal stains as a preliminary test.

    Description / Table of Contents: Zusammenfassung Es wird eine einfache und genaue Methode zur Bestimmung von Eisen im Blut unter Verwendung von Säureaufschluß im Heizblock beschrieben. Der Aufschluß benötigt wenig Mühe und erlaubt die Verarbeitung einer großen Zahl von Proben. Der verdünnte Aufschluß wird zur kolorimetrischen Eisenbestimmung mittels o-Phenanthrolin benutzt und die entsprechende Hämoglobinkonzentration im Blut berechnet. Es zeigte sich eine gute Korrelation der Ergebnisse mit der beschriebenen und der Cyanmethämoglobin-Methode anhand von 21 Standardproben, die 4,9 bis 26,8 g/dl Hämoglobin enthielten.
    Notes: Summary A simple and accurate determination of iron in blood involving acid digestion in a heating block is described. The digestion requires little attention and permits the handling of large numbers of samples. The diluted digest is used for the colorimetric determination of iron using o-phenanthroline, and the equivalent hemoglobin in blood is calculated. There was a good correlation between the results of the present method and the cyanmethemoglobin method with 21 standard blood samples containing 4.9–26.8 g/dl of hemoglobin.

    Description / Table of Contents: Zusammenfassung Es wird eine neue enzymatische Methode zur quantitativen Bestimmung von Spermin im menschlichen Samen beschrieben, die auf einer spezifischen Reaktion von Polyamin-Oxidase aus Gerstensämlingen mit Spermin beruht. Geringe Mengen menschlichen Samens wurden mit der Polyamin-Oxidase inkubiert und das durch die Oxidase-Reaktion gebildete Wasserstoffperoxid photometrisch nach Kupplung von 4-Aminoantipyrin mit N-Äthyl-N-(3-methylphenyl)-N′-acetylethylendiamin in Gegenwart von Peroxidase gemessen. Die Nachweisgrenze von Spermin in der Probe liegt bei etwa 10 nmol. Die mittlere Konzentration von Spermin im menschlichen Samen beträgt 2.41 μmol/ml; die Werte für Vaginalflüssigkeit, Speichel, Serum und Urin liegen unter der Nachweisgrenze der Methode.
    Notes: Summary A new enzymatic method for the quantitation of spermine in human semen, based on the specific reaction of barley seedling polyamine oxidase with spermine, is described. Small amounts of human semen were incubated with the polyamine oxidase; hydrogen peroxide formed in the oxidase reaction was measured photometrically by coupling 4-aminoantipyrine with N-ethyl-N-(3-methylphenyl)-N′-acetylethylenediamine in the presence of peroxidase. The detection limit of spermine of this method was about 10 nmol per tube. The mean level of spermine in human semen was 2.41 μmol/ml; the levels in vaginal fluid, saliva, serum, and urine were below the detectable limit by this procedure.

    Notes: Summary A detailed procedure of a new photometric assay for total diamines and polyamines in human urine using soybean seedling amine oxidase (SSAO) as an enzyme reagent is described. It is based on the unique substrate specificity of SSAO that the enzyme is active toward all diamines and polyamines. The amines were purified from urine by cation-exchange chromatography and incubated with SSAO. Hydrogen peroxide formed in the oxidase reaction was measured photometrically by coupling 4-aminoantipyrine with phenol in the presence of peroxidase. For its simplicity and sensitivity, our method seems useful for routine clinical investigation. The data obtained from normal subjects and patients of various cancers are presented to validate the present method.
