    Notes: Several biological distances based on cranial and mandibular variation among breeding groups of white-tailed deer were calculated and compared with geographic distances among the groups. Distances based on epigenetic variation among ten groups were calculated using 16 non-metric variants of the cranium and mandible. Penrose's size and shape distances and Mahalanobis' D2 distance were calculated for 11 groups; the calculations were based on seven skeletal and seven dental metric variables of the mandible.The biological distances were correlated with geographic distance as follows: the epigenetic distance, 0.74; Penrose's shape distance, 0.71; Penrose's size distance, 0.45; and Mahalanobis' distance, 0.37. All correlations were significant at the 0.01 level. The epigenetic and Penrose shape correlations were significantly higher than the Mahalanobis correlation.Because of the conditions under which the breeding groups were selected, it was assumed that genetic affinites among the groups would be a function of geographic distance. The results suggest that the epigenetic distance and Penrose's shape distance reflect genetic affinities among groups better than do the Penrose size and Mahalanobis distances.

    Notes: A principal component analysis revealed that the two major components of mandibular shape variation among individuals within breeding groups of white-tailed deer in Canada and the United States involve contrasts between the mandible and the dentition and between the premolars and the molars. Size variation appeared to account for 34% of the total variation within the groups, and the two major shape variations accounted for 23% and 8% respectively.A canonical analysis was used to identify the major components of mandibular variation among the breeding groups and to provide measures of the proportion of the total variation accounted for by each component. Among male groups, size variation was associated with latitude, and the major shape variation was closely associated with longitude, so a bivariate plot of the first two canonical variates reflected the general geographic orientation of the populations.The mandibular size in a Tennessee population that descended from Wisconsin and Michigan ancestors appears to have not decreased appreciably in the more southerly habitat after introduction more than 20 years ago. Changes in range conditions in eastern Upper Michigan over the past 30 years have not influenced local mandibular morphology as reflected by the first two canonical variates. Regardless of general smallness of individuals, the mandibular morphology of the deer from the Cross Timbers area in Kansas appears to approximate more closely that of northern populations than that of the more southerly populations from Oklahoma and Texas.Sexual dimorphism on the first two canonical axes was observed.

    Notes: 9+0 cilia associated with diplosomes were observed in myoepithelial cells of the ductal system in the undiseased, resting, adult female breast. Ciliary diameter was 250 nm, and the longest found was 830 nm. The centrioles comprising the diplosome were 500 nm in length and 160 to 200 nm in diameter. Satellite bodies were associated with the diplosome but there was no rootlet system. Evidence suggested that all myoepithelial cells bear a cilium. The possible functions of the cilia in a sensory role as chemo- or mechanoreceptors related to myoepithelial and epithelial activities are discussed.

    Notes: In an attempt to facilitate quantitative analysis of neural peri-karya, an inexpensive system of gridded coverslips has been developed to permanently map sections of tissue on microscopic slides. Utilizing common photographic copying techniques with Kodalith film, a mylar coverslip is produced which is imprinted with a grid of known dimensions. The exact scale of the grid can be varied precisely for compatibility with a particular field of magnification. The versatility and economy of this technique make it useful in many microscopic studies requiring superimposition of discrete landmarks.

    Notes: Initial cytodifferentiation of neuroblasts in the retina of chick embryos was investigated by means of silver impregnation, H3-thymidine autoradiography, and electron microscopy. Utilization of various combined procedures with the same initial fixative demonstrated that the results of neurofibrillar staining are complementary, not contradictory, to information obtained from both autoradiography and electron microscopy.The question of a homogeneous versus a non-homogeneous cell population within the neural epithelium was clarified by the closely related appearance of neurofibrils to the terminal mitotic cycle of some presumptive neuroblasts. Neurofibrils were observed in certain apolar pre-division cells as well as in pairs of transitional apolar and bipolar daughter cells which apparently do not remain in the neural epithelium. At the ultrastructural level, aggregates of 60 Å neurofilaments were observed in configurations that corresponded to neurofibrillar networks. As the bipolar neuroblast moves into the ganglion cell layer, it would appear that much of the neurofibrillar substance is transported within the elongating peripheral process which becomes the optic nerve fiber. Further investigation of the relationship of neurofilaments to neurotubules in the neuroblast undergoing cytodifferentiation is now in progress.

    Notes: Decidual tissue occurring within the human ovarian cortex was examined by light and electron microscopy. Of 21 ovarian specimens obtained at term (36-42 weeks of gestation), decidual cells were confirmed in each. Decidual cells were found within the tunica albuginea as single cells, in nodules, in polyps or in confluent sheets. Decidual cells exhibited several characteristics of cells engaged in secretory activity: abundant rough and smooth endoplasmic reticulum, numerous profiles of the Golgi complex and a large, euchromatic nucleus devoid of heterochromatin and displaying a prominent fibrous lamina. Peduncular protrusions at the periphery of the cell contained numerous dense bodies 0.4-0.9 μm in diameter. These dense bodies were bounded by a single membrane and contained granular subunits 30-60 nm in diameter. These granular subunits were observed in the process of apparent exocytosis, as well as free in the extracellular space. Secretory bodies and their granular content also were observed both in the region of the Golgi complex and partially extruded into peduncular processes. By far the greatest number of secretory bodies occurred within peduncular processes where they may be stored prior to release. Migration of a secretory body into a peduncular process and exocytosis from such a process appears to be an unusual mode of meocrine secretion, perhaps unique to decidual cells.

    Notes: Human marrow cells, suspended in methylcellulose medium containing erythropoietin, give rise to discrete colonies of hemoglobin synthesizing cells. The presumption that such colonies originate from single progenitor cells has been tested directly in females with X-chromosome inactivation mosaicism using glucose-6-phosphate dehydrogenase (G-6-PD) as a marker. When individual colonies were grown from marrow cells obtained from two black females heterozygous for G-6-PD, only one or the other isoenzyme type was observed, but not both. These results are most consistent with the interpretation that human erythroid colonies arise from single cells.

    Notes: When human myeloma cells are pulsed for one hour with 3H-uridine and chased for six hours in fresh medium containing unlabeled uridine, the processing of 45 S rRNA precursor into the stable 28 S and 18 S rRNA components can be followed. However, when the cells are chased in exogenous adenosine instead of uridine, the accumulation of 18 S rRNA is selectively inhibited. Cells pulsed with 3H-adenosine and chased in the absence of exogenous nucleosides exhibit normal rRNA precursor processing, while cells pulsed simultaneously with 3H-uridine and 3H-adenosine and chased with uridine and adenosine are deficient in labeled 18 S rRNA. Consequently, the inhibition of 18 S rRNA accumulation by adenosine is not an artifact of labeling nor is it relieved by an equal molar concentration of uridine. The wasting of 18 S rRNA in human myeloma cells is similar to that reported to occur in normal lymphocytes during the quiescent state.

    Notes: Under physiological conditions, erythrocytes of the horse metabolized 638 ± 37 (± SE) nmoles glucose/ml cells/hr at 37°C compared to 942 ± 31 for the cat, 1,329 ± 44 for the dog, and 1,485 ± 43 for man. On an absolute basis, pentose phosphate metabolism was similar between species, with species differences in erythrocyte glucose utilization attributable to differences in Embden-Meyerhof pathway metabolism. By examining pentose phosphate pathway recycling, it appears that some functional compartmentation exists within erythrocytes.

    Notes: Neoplastic cell lines exhibit RNA synthesis and process patterns which are related to phenotypic attributes more complex than merely the rate of proliferation. Mouse neuroblastoma cells of the same genotype but different differentiated states have different ribosomal RNA precursor processing patterns, while plasmacytoma cells of different genotypes but the same differentiated state have the same pre-ribosomal RNA processing pattern. In addition, our observations indicate that chromatin-associated RNA is involved in cytodifferentiation and is closely related to phenotypic variability. When neuroblastoma cells are induced to differentiate, there is a 2- to 3-fold increase in the labeling of chromatin-associated RNA. Both of the differentiated cell lines, human myeloma and mouse neuroblastoma, have slow-labeling, stable chromatin-associated RNA while this same fraction from HeLa cells is labeled rapidly and is unstable.
