    Notes: Monosodium glutamate was administered daily on days 5 through 10 postnatally at a dose of 2.5 mg/gm body weight. Counts of remaining perikarya in the arcuate nucleus of adult mice indicated approximately an 80% decrease in the number of perikarya. The arcuate lesion resulted in endocrine deficits; reproductive capacity was reduced, animals were smaller in stature and obese, and the weights of the anterior pituitary, ovaries and testes were significantly decreased while adrenals were unaffected. Light microscopic studies revealed no significant changes in thickness or general histological appearance of the median eminence. At the electron microscope level, there were no alterations in the number of nerve terminals or dense core vesicles per unit area in the contact zone. These observations suggest that afferents to the median eminence from the arcuate nucleus may form a relatively small portion of its total nerve terminal population.

    Notes: The morphology of a sex pheromone-producing gland found in the abdomen of Drosophila grimshawi males was studied by light and electron microscopy. This gland, consisting of two intra-anal lobes, contains cells that resemble those of other insect pheromone glands. However, in contrast to many other insect pheromone glands that release pheromone through the cuticle, cells of the intra-anal lobes secrete into a canaliculi-duct system that empties into the anal region. The liquid secretory product flows along the surface of the intra-anal lobes and is brushed onto the substrate by fingerlike projections on the lobes' surfaces.

    Notes: Newly metamorphosed Kenyan reed frogs, Hyperolius viridiflavus ferniquei, are able to regenerate amputated digits. The terminal digital pad is also completely reformed. Differentiation of the regenerating digital pad was studied by scanning electron microscopy. External differentiation of the digital pad began late in the second postamputational week with the appearance of small patches of specialized epidermal cells on the ventral surface of the regenerating digit. The differentiation of the pad spread out radially until late in the fourth week, when its overall shape approximated that of the normal digital pad. The appearance of patches of digital pad epidermis on the ends of spike regenerates arising from the forearm was also confirmed.

    Notes: The cells of the follicular epithelium of the ovarioles of the mosquito, Aedes aegypti (L.), and of the stable fly, Stomoxys calcitrans (L.), have been found to be interconnected by cytoplasmic bridges. Cytoplasmic bridges have been reported between sibling gamete cells of both male and female insects, but these bridges have not been previously reported to exist between somatic cells of insects.

    Notes: Newly metamorphosed individuals of the Kenyan reed frog, Hyperolius viridiflavus ferniquei, are able completely to regenerate amputated digits, including the morphologically complex digital pad. The sequence of morphological events is very similar to that seen in the typical epimorphic regeneration of amphibian extremities. Amputated forearms typically produce hypomorphic spike regenerates. Despite the lack of underlying hand and digital structures, an apical “digital” pad commonly differentiates.

    Notes: A survey of 69 of the 74 currently recognized chelonian genera revealed that 21 genera in three families (Emydidae, Platysternidae and Testudinidae) possess paired integumentary glands or gland vestiges in the anterior throat skin. These glands are here termed mental glands; they are holocrine and may be classified morphologically as follows: Class I mental glands are large, complex, multilobed, have specialized ducts, and are found only in the genus Gopherus: Class II mental glands are small, simple sacklike invaginations containing secretory cells or keratinizing cells. The structure of Class II glands varies from distinctive and saccular to shallow keratinized invaginations having no glandular tissue; they are found only in the families Platysternidae and Emydidae.Mental glands occur in 17 of the 22 genera in the subfamily Batagurinae (sensu McDowell, 64); only 2 of 9 genera in the subfamily Emydinae have these glands. The taxonomic occurrence of mental glands suggests that they are primitive structures. The loss of mental glands in most emydines is interpreted as a subfamilial trend toward integumentary simplification.

    Notes: We sought a cultured cell line with Proline Oxidase activity to study the regulation and physiologic role of the enzyme in mammalian tissues. Among the cell lines tested, only LLC-RK1 cells, derived from rabbit kidney, had significant Proline Oxidase activity; the Km for proline of the enzyme from these cells was similar to that for the liver enzyme. LLC cells, Proline Oxidase positive, were able to convert proline to CO2. In contrast, CHL cells, Proline Oxidase negative, did not have this capability. The presence of Proline Oxidase in LLC cells and the absence of the enzyme in fibroblasts suggest that Proline Oxidase may serve as a marker enzyme for distinguishing parenchymal kidney cells from fibroblasts in culture. Cells transformed by SV40 virus and cells transformed by methylcholanthrene had activities higher than the parent cell line, but this effect of transformation could not be generalized to all transformed cells. Finally, L-hydroxy proline at 100-fold greater concentration than substrate L-proline failed to decrease proline oxidation. This finding suggests distinct degradative enzymes for these two amino acids.

    Notes: A large collection (105) of mouse L cell mutants lacking hypoxanthine-guanine phosphoribosyl transferase activity (HGPRT; E. C. 2.4.2.8) were analyzed for the presence of serologically cross reacting material (CRM). Antibody directed against highly purified mouse liver HGPRT was used for detecting CRM activity by two methods: (1) the standard precipitation-inhibition assay; and (2) a radioimmune-precipitation assay. The latter assay proved to have far greater sensitivity for the detection of altered forms of HGPRT. Approximately 40% of the HGPRT- cell lines contain CRM activity (i.e., were CRM+). This indicates that a minimum of 40% of the HGPRT- clones arose as a result of mutations in the HGPRT structural gene. The CRM+ cell lines were shown to contain different levels of CRM activity. Measurements of the heat sensitivity of CRM in the different HGPRT- cell lines showed a broad spectrum of CRM heat inactivation kinetics. These latter two observations provide strong evidence that the mutations giving rise to the HGPRT-CRM+ phenotype occurred at different sites in the HGPRT structural gene.

    Notes: Adsorption of Sendai virus at high multiplicity (500-1,000 HAU/106 cells) to HeLa cells grown in monolayers causes immediate changes in the ion barrier of the cell membrane, as well as changes in the morphology of the virus-treated cells. Within minutes of adsorption the cells begin to lose potassium and an extensive influx of ions into the cells occurs. Concomitantly with these changes, the cell membrane becomes depolarized, and the resting potential across its membrane decreases. Twenty to sixty minutes post adsorption the damage to the cell membrane is repaired, and both the potassium uptake and the resting potential return to their pre-exposure values. Scanning electron-micrographs of Sendai infected cells incubated at 37°C show formation of bridging microvilli in a zipper-like fashion within two to five minutes post-adsorption; 30 to 60 minutes thereafter the majority of cells in the monolayer are fused. Biochemical changes induced by virus adsorption and the role of Ca++ ions in the observed effects are discussed.

    Notes: In 3T3 Swiss mouse fibroblasts, incorporation of phosphate into cells and phosphorylation of small organic compounds were increased by shaking dense cultures. This response was not obtained with SV40 transformed Swiss 3T3 cells (SV-3T3).It appeared likely that these results could be accounted for by an inhibitor released from 3T3 cells but not from SV-3T3 cells.Our new method of co-incubation of sparse and dense cultures allowed us to demonstrate inhibition of growth and phosphate metabolism in sparse 3T3 cultures which were shaken in the presence of dense cultures. The inhibition was much less when the cultures were co-cultivated but not shaken.The inhibition of phosphate incorporation in acid-soluble and acid-insoluble fractions of sparse cultures was observed as early as 20 minutes of co-incubation in the presence of dense cultures, so this inhibition is not the result of depletion of growth factors in the medium.Our experiments suggest that an inhibitor(s) was released from dense cultures of 3T3 cells.
