    Description / Table of Contents: Die gastrointestinale Absorption von Poly(biguanid-1,5-diylhexamethylen-hydrochlorid) (1) in Ratten beträgt nur 5,6% einer einzigen oralen Dosis, und auch nach Langzeitbehandlung liegen die Konzentrationen im Gewebe höchstens bei 0,3 μg g-1. Durch cinfache chromatographische und spektroskopische Methoden konnte gezeigt werden, daß die im Harn ausgeschiedenen Substanzen in kleinen Mengen aus 1-Oligomeren mit zwer Cyanguanidino-Endgruppen bestehen, sowie aus Spuren von 3,3′-Dicyano-1,1′-hexamethylendiguanidin (8a) und einer Verbindung, die für 1-(6-Aminohexyl)-3-cyanoguanidin (2) gehalten wird, und die bei der Synthese von 1 entsteht. Die Bestandteile von 1 werden demnach in Ratten nicht metabolisiert.
    Notes: Gastro-intestinal absorption in rats of poly[biguanide-1,5-diylhexamethylene hydrochloride] (1) amounts to only 5,6% of a single oral dose, and after chronic administration, tissue concentrations do not exceed 0,3 μg g-1. It has been shown by simple chromatographic and spectroscopic methods that urinary polymer-related material consists of small amounts of 1-oligomers, with two cyanoguanidino end groups, as well as the trace constituents, 3,3′-dicyano-1,1′-hexamethylenediguanidine (8a) and a compound which is considered to be 1-(6-aminohexyl)-3-cyanoguanidine (2) that is formed during the synthesis of 1. Hence, the constituents of 1 are unmetabolized in rats.

    Notes: Experimental molecular weights in the range 200 to 100,000 Daltons have been determined by vapor pressure osmometry using three different solvents. For all values in excess of 10,000, molecular weights were also determined by membrane osmometry. The general agreement found for molecular weights determined under a variety of conditions, leads to the conclusion that values as high as 100,000 Daltons can be determined by at least one type of vapor pressure osmometer when it is calibrated with a material of 200 Daltons. Thermistor self-heating and diffusion in the liquid phase are shown to be unimportant for this particular instrument.

    Notes: Surgical replacement of the Achilles and patellar tendons using Dacron mesh silicone prostheses was performed in 15 mature beagle dogs. This part of the study includes clinical evaluation, gross inspection at autopsy of regrown tendons, and histological determination of biocompatibility of prosthetic implants.Functional continuity and integrity of prosthetic patellar tendons have been established by evaluating the biologic-prosthetic interface and the mechanical properties of regrown tendon tissue around and through the Dacron silicone replacements. Results of prosthetic Achilles tendons were less satisfactory because of difficulties in suturing to a muscle and its fascia. Although the prosthetic tendon did not regrow through the tube, it provided a structure for regrowth around it. The regrown tendons became nearly ten times the normal crosssectional area after three to four months and decreased to two to three times after 13 months.Histological studies indicate that in the absence of infection, the Dacron mesh silicone tendon was well tolerated for periods up to 13 months. Overall results are encouraging and warrant further investigation although the regenerative capacity of human patellar and Achilles tendons is unknown.

    Notes: The results of extended Huckel calculations are reported for an isolated (SN)x chain in which some of the S2N2 units of the extended one-dimensional chain are replaced by (SCH2)2 units. It is found that, inter alia, the metallic behavior of the unsubstituted parent is retained but that the conductivity is lowered to an extent governed by the nature and degree of substitution.

    Notes: It was found that diacyl peroxides can be formed in situ in a polymerization medium by the reaction of an acid anhydride with hydrogen peroxide. For the specific application to aqueous vinyl chloride polymerization, an initiator system based on the base-catalyzed reaction of isobutyric anhydride with hydrogen peroxide to produce diisobutyryl peroxide gave very good results. In contrast, the acid chloride was completely ineffective as a peroxide precursor in this reaction. Studies pointing to diisobutyryl peroxide as the initiating species; investigations of reactant stoichiometry; and comparison of the in situ system with preformed diisobutyryl peroxide were conducted. It was shown that this system makes possible the polymerization of vinyl chloride at 30°C at rates comparable to those obtained with dialkyl peroxydicarbonates at 50°C, thus demonstrating the ability of this system to initiate vinyl chloride polymerization at low temperature. The rates of vinyl chloride polymerization with the use of different concentrations of in situ diisobutyryl peroxide at 30, 40, and 50°C were determined. Similarly, polymerization rates with the use of combinations of in situ diisobutyryl peroxide and n-propyl peroxydicarbonate were determined. The data obtained demonstrate rapid initiation of the polymerization reaction and a reduction in polymerization time made possible by this dual initiator system. These results were verified in pilot-plant and commercial-scale PVC polymerizations.
