    Notes: Abstract Ten euthymic manic-depressive patients with therapeutic plasma lithium levels were each given two i.v. infusions of 30 mg of methylphenidate. The infusions were separated by at least 3 days. Before one infusion each patient was given 5 mg of haloperidol i.v. and before the other infusion each was given an identical volume of saline. A psychiatric observer was blind to the nature of the pretreatment and the order of pretreatment was randomized. Saline pretreated patients showed marked activation and euphoriant responses despite therapeutic lithium levels. Haloperidol pretreatment reduced this response in three patients and eliminated the euphoriant and activating response in the remaining seven patients. These results agree with the existence of a dopaminergic step in the induction of methylphenidate-induced activation and euphoria.

    Notes: Abstract Administration of epinephrine in man has been shown previously to lead to a rise in plasma cyclic AMP levels by activation of the β-adrenergic-stimulated adenylate cyclase. Therapeutic doses of lithium in humans block the epinephrine-induced rise in plasma cyclic AMP levels, suggesting that lithium inhibits β-adrenergic adenylate cyclase. In contrast, ten subjects receiving haloperidol, a drug also effective in the treatment of mania, show a mean rise in plasma cyclic AMP levels after epinephrine administration and the magnitude of the response is the same as for non-drug treated individuals. These findings are discussed in relation to the possible pharmacological mechanisms of action of lithium and haloperidol in the control of mania.

    Notes: Formation of the antiparallel-chain β-sheet protein conformation is induced in in vitro human stratum corneum by three homologous organic sulfoxides known to enhance skin permeability: dimethylsulfoxide (Me2SO), hexylmethylsulfoxide (HxMeSO), and decylmethylsulfoxide (DecMeSO). Me2SO and HxMeSO apparently function by displacing water molecules bound to polar protein side-chains, whereas DecMeSO probably interacts hydrophobically with the protein. The conformational transition does not result from lipid removal. The β-sheet protein, most likely formed in normally α-helical portions of the intracellular keratin filaments, is reconverted to α-helix upon rehydration of the tissue. Though neat Me2SO produces the most β-sheet of all treatments examined, the sequence of ability to promote β-sheet formation at the 1M level is HxMeSO 〉 DecMeSO 〉 Me2SO. Spectroscopic evidence is presented regarding the dependence of β-sheet formation on sulfoxide concentration, treatment duration, pH, and tissue hydration. The relationship of this conformational change to the enhancement of skin permeability is briefly discussed. The result of sulfoxide treatment is different from results of sodium dodecylsulfate and heat treatments of stratum corneum.

    Notes: Exturded filaments of unmodified polypropylene (PP) with and without antioxidant were implanted subcutaneously in hamsters in order to determine their rate of degradation. Specimens were removed periodically during a 5 month test period and analyzed by infrared spectroscopy and dynamic mechanical testing. The analyses show that degradation begins to occur after only a few days. Although the reaction sequence is not known, several factors suggest that the in vivo degradation process is similar to autoxidation which occurs in air or oxygen.The infrared data indicate that the hydroxyl content of the implants increases at a rate of 0.061 mg/g polypropylene per day during the initiation phase of the reaction. An induction time of 108 days was established. Carbonyl bonds appear after an implantation time of 50-90 days and increase thereafter. Mechanical tests indicate a decrease in the dynamic loss tangent, tan δ, during the first month of implantation for unmodified polypropylene. No change in the infrared spectra or tan δ was observed, however, for implants containing an antioxidant.Thus, it is apparent that polypropylene filaments implanted subcutaneously in hamsters degrade by an oxidation process which is retarded effectively by using an antioxidant. While the findings reported are specific to subcutaneous polypropylene implants, they suggest that degradation of other systems may involve similar processes. This notion suggests directions for further research on increasing the in vivo stability of synthetic polymers.Long-term effects of polymer implantation upon tissue were not studied in this work.

    Notes: In evaluating molecular orientation by dynamic infrared techniques, it is shown, subject to certain restrictions, that an extensional mechanism plays an important role in the measured orientation. Dichroic measurements can indicate a change in segmental orientation without any change occurring in the chain axis orientation. Extensional orientation was evaluated for a general helical molecule in terms of singular and coupled vibrational modes. Significant orientation changes can occur by the extensional mechanism, the sign and magnitude of which depend on the components of the transition moment vector.

    Notes: In the vibrational spectrum of a polymer obtained under tensile stress loading conditions, several modes of molecular deformation can lead to different mechanisms of frequency shifting and asymmetric band deformation. Theoretically and experimentally it has been observed that quasielastic deformations (reduction of force constants due to bond weakening under stress), pure elastic bond stretching and angle bending, conformational variations, and several types of chain defects can cause linear shifts in frequency and infrared band distortion. A detailed study of the deformation spectra of isotactic polypropylene and polyphenyl-p-sulfide indicated that quasielastic and elastic mechanisms are the major contributors to frequency shifting, principally affecting stretching and bending vibrational modes. Conformational mechanisms can affect torsional modes whereas the defect mechanism, when present, can cause random distortion of an infrared band. The latter mechanism is difficult to quantize. The extent to which each mechanism contributes to the total spectral deformation can be a function of morphology, macroscopic loading conditions, thermal and strain histories.

    Notes: The mechanics of “springy” or “hard elastic” row-crystallized polymers were investigated. It was found that their unique tensile and physical properties could be explained by adhesive fracture mechanics and simple beam deflection. The deformed lamellae had an apparent negligible modulus of flexural rigidity. An adhesive failure model was designed which duplicated all the tensile characteristics of springy polymers, including constant lateral contraction, critical stress, peel stress, rehealing, energetic elasticity, and cyclic strain behavior. The ideal relative density change as a function of strain was found to be the same for all springy polymers. Dynamic infrared analysis showed that the molecular orientation decreased during the deformation of springy polypropylene films. The latter is due to the deflection of lamellae. Rehealing was accompanied by a time-dependent reversal of orientation. The enthalpic elasticity is derived from the strain energy of the lamellar network.
