    Notes: An ion doublet chemical ionization mass spectrometry technique was utilized for the rapid detection and elucidation of the structure of a new hydroxylated metabolite of the oral anticoagulant warfarin. This technique involves the use of a 50:50 mixture of stable isotope labeled and unlabeled drug. With this procedure ions found in the mass spectrum which are associated with the administered drug can be identified unambiguously. The unknown metabolite from rat liver microsomal preparations was identified as benzylic hydroxywarfarin by reincubating the microsomes with warfarin specifically labeled in the benzylic position and observing the subsequent loss of label in the product. The ion doublet technique was also employed in a single human study, and benzylic hydroxywarfarin was detected and identified.

    Notes: Our previously published gas chromatography chemical ionization mass spectrometry stable isotope dilution assay for clonazepam and its 7-amino metabolite in plasma has been modified to obtain more consistent recoveries of the amine metabolite and to permit the determination of the 7-acetamido metabolite of clonazepam. In addition, the manuscript describes the details of a method for a least squares analysis of the nonlinear standard curves associated with the use of certain stable isotope labeled analogs as internal standard.

    Notes: A gas chromatographic chemical ionization mass spectrometric assay has been developed to measure carprofen in blood. The method features the addition of either a structural or stable isotope analog internal standard to plasma prior to a simple benzene extraction at pH 4.5. The residue, after removal of the benzene, is methylated with ethereal diazomethane. Following evaporation of the methylating solvents, a portion of the reconstituted residue is analyzed by gas chromatography mass spectrometry. The mass spectrometer is set to monitor in the gas chromatographic effluent the [MH]+ions of carprofen methyl ester and the methyl ester of internal standard generated by isobutane chemical ionization. Assay sensitivity is 5 pmol ml-1. When 200 pmol ml-1 samples are analyzed using a stable isotope analog as the internal standard, the precision and accuracy are both 4%. Using a structural analog as the internal standard, the assay was neither as precise nor as accurate.
