    Notes: Summary The influence of the fixation method on the distribution of fluorescent Evans blue (EB) was studied in the cervical grey matter of rats treated with 6-aminonicotinamide (6-AN). 6-AN leads to selective injury to the neuroglial cells and blood brain barrier without primary neuronal damage. In this model hyperchromasia and selective fluorescence of neurons were closely related but only found in tissue fixed by immersion, indicating a supravital shift of EB to shrinking neurons. Adequate perfusion fixation with Bouin solution prevents this artifactitious neuronal staining and demonstrates that EB is primarily bound by the vessel wall and the perivascular neuropil. The supravital diffusion of EB in the CNS and its dependence from the fixation method represents relevant methodological factors in morphological studies on the blood brain barrier.

    Notes: Summary Focal vasogenic brain edema was induced by ultraviolet irradiation of the exposed cerebral cortex of 16 cats. In 5 animals horseradish peroxidase was intravenously injected at times varying from 30 min to 24 h following irradiation and allowed to circulate for 45 min. Fixation was carried out by perfusion and immersion with glutaraldehyde. The tissue, part of it incubated for peroxidatic activity, was treated for electron microscopy. The UV-irradiation leads to a shallow coagulation of the superficial cortex from which a wide zone of edematous tissue spreads to the deep white matter within 24 h. Arterioles, capillaries and venules of this zone show enhanced pinocytotic activity and a con-current rise in permeability for horseradish peroxidase which is found in micro- and macropinocytotic vesicles, in tubular vesicle-like structures, in endothelial wall invaginations and apparently not membrane-bound. These changes are most pronounced in venules which after 48 h allow penetration of reaction product though the base membrane into the surrounding neuropil. There is no evidence for the penetration of tight junctions which appear intact.

    Notes: Summary Demineralized bovine bone powder was reduced with NaB3H4 to label the collagen crosslinks with tritium. The powder was enclosed in small nylon mesh pouches and implanted subcutaneously into rats for 3 weeks. Histological examinations revealed that multinuclear giant cells accumulated around the bone matrix, some in Howship's lacunae. Collagenous peptides containing intermolecular crosslinks were detected in the urea-soluble fraction extracted from the implant. Two crosslink-containing peptides were isolated from a dialyzable fraction: one contained dihydroxylysinonorleucine and the other hydroxylysinonorleucine. Both peptides had molecular weights of approximately 1000 estimated from the elution positions of gel filtration chromatography; and both had similar quantitative compositions of amino acids. There were no homologous peptides detected in a control experiment of the reduced bone matrix which was incubated in vitro with buffered saline for 1 week at 37°C.

    Notes: In the absence of emulsifying agents, vinyl acetate polymerization in aqueous media was carried out at 50°C over a wide range of initial initiator and monomer concentrations to clarify the effect of reaction conditions on the kinetic behavior of the polymerization system. It was shown that the rate of polymerization was proportional to reaction time and initiator concentration and independent of the number of polymer particles present. The rate could also be successfully explained by the Smith and Ewart theory for emulsion polymerization when the dissolved monomer in water and the Trommsdorff effect were taken into consideration. A set of equations which could account for the effect of dissolved monomer in water on the rate of polymerization is proposed.
