    Notes: In a search for H- or L-chain-related cross-idiotypic specificity among human anti-I and anti-i cold agglutinins, two idiotypic antisera raised against the IgMχ cold agglutinin Da were tested for their binding activity to isolated cold agglutinin H and L chains. Negligible H-chain binding activity was found, but there was high-titre L-chain binding activity in one of the antisera. This was an unsuspected VχIII subgroup activity which enabled the classification of VχIII proteins into three subgroups. The χ chains of five out of six anti-I and anti-i cold agglutinins belonged to the antigenically most active VχIII subgroup. Absorption of the idiotypic antiserum with a Bence Jones protein of this latter subgroup did Dot appreciably alter the precipitating cross-idiotypic activity of the antiserum when tested with intact cold agglutinins. However, these studies do not rule out the possible existence of a VχIII subgroup-associated conformational antigen in an intact Fab region, which is seen as a ‘cross idiotypic’ antigen by heterologous (rabbit anti-human) antisera.

    Notes: The region on the light chain molecule responsible for expression of the χ and λ antigenic determinants was determined by comparative immunochemical analyses of intact Bence Jones proteins and naturally occurring or enzymatically derived fragments of Bence Jones proteins that lacked extensive portions of the V region or part of the C region. The reactivity of these fragments with numerous antisera having specificity for light-chain antigenic determinants indicated the essentiality of the intact light polypeptide chain for expression of the χ and λ antigenic determinants. The conformational dependency of the χ and λ antigenic determinants was also evidenced by denaturation-renaturation studies on χ and λ chains. The V domain, C domain, and interdomain ‘switch’ region contribute to the expression of χ or λ antigenicity and to certain isotypic and allotypic specificities.

    Notes: Abstract Five babies, aged between one to three months presented with clinical and serological evidence of syphilis. In addition to the usual periosteal and metaphyseal bone lesions of lues, areas of focal lucencies and sequestra were present. The association of sequestra in focal diaphyseal erosions may represent a non-dystrophic manifestation of the disease. The lesions were attributed to the syphilitic process. Trivial trauma to disorganised metaphyses, dystrophic changes and possibly focal osteomyelitis account for the spectrum of bone lesions in congenital syphilis. No tubular bone appears immune to these processes.

    Notes: Summary Some properties of nuclear RNA taken up by isolated mitochondria during aerobic incubation were studied. Nuclear RNA fraction re-isolated from incubated mitochondria was characterized by homogeneous sedimentation distribution in sucrose gradient (7–9S) and by absence of extended secondary structure. In this respect the RNA fraction differed from the nuclear RNA added to mitochondria and was similar toin vivo rapidly labelled mitochondrial RNA. The hybridization pattern of re-isolated RNA with nuclear DNA is consistent with the presence of transcripts from unique DNA sequences in this RNA fraction.

    Notes: Summary The synthesis of virus-specific macromolecules was studied in the reconstituted system containing inner membrane-matrix fraction from rat liver mitochondria and infectious RNA of Venezuelian equine encephalomyelitis (VEE) virus. In a series of preliminary experiments it was shown that isolated submitochondrial fraction was completely free of interfering cytoplasmic contaminations and particularly, of cytoplasmic 80S ribosomes. VEE RNA when added to submitochondrial system caused significant stimulation of RNA and protein synthesis. These processes were resistant to actinomycin D which inhibited profoundly the synthesis of proper mitochondrial macromolecules. The stimulating effect of VEE RNA in experiments with submitochondrial system was about three times higher than that with intact mitochondria. The stimulation of14C-amino acid incorporation increased as a function of incubation time; a certain lag-period being observed. The newly formed virus-specific RNA's and ribonucleoproteins were identified with the aid of sedimentation analysis. In particular, radioactive RNA's with sedimentation coefficients 40S and 26-18S were isolated from the incubated system. These RNA's are similar respectively to VEE genome RNA and doublestranded VEE replicative RNA. In double labelling experiments with3H-uridine and14Camino acids it was shown that VEE RNA induced synthesis of ribonucleoproteins containing newly formed RNA and protein. These RNP possessed sedimentation coefficients 60-80S, 140S and 300S in sucrose gradient and buoyant densities 1.32 and 1.50 g/cm3 in cesium chloride gradients. These properties of ribonucleoproteins synthesized de novo in submitochondrial system are close to those of RNP intermediates of VEE virus reproduction in the infected cells. We concluded that viral RNA could program virus-specific synthesis in the submitochondrial system under conditions that eliminated the contribution of cytoplasmic ribosomes.

    Notes: Summary The mathematical equations for the transport of nonelectrolytes across cell membranes are critically examined and cast in forms suitable for solution which involve fewer approximations than has heretofore been commonly done. For the case of red cells, the equations are developed to include the effect of the variation in apparent nonosmotic water owing to the variation in hemoglobin concentration as the cell swells or shrinks. Two methods of solution of the equations are developed and studied and sample calculations are provided. It is shown that the solutions to the linearized equations commonly found in the literature are insufficiently accurate for some purposes and this inaccuracy is avoided by the methods given here. The importance of retaining the effects of variations in apparent nonosmotic water and in solute volume in the cell is demonstrated.

    Notes: Summary Inside out and right side out vesicles were used to study the sidedness of Ca binding to the human red cell membrane. It was shown that these vesicles exhibited only a limited permeability to Ca, enabling the independent characterization of Ca binding to the extracellular and cytoplasmic membrane surfaces. Ca binding was studied in 10 mM Tris HCl at pH 7.4, 22±2°C and was shown to be complete in under 5 min. Scatchard plots were made from Ca binding data obtained at free Ca concentrations in the range of 10−6 to 10−3M. Under these conditions inside out vesicles exhibit two independent binding sites for Ca with association constants of 1×105 and 6×103 M−1, and right side out vesicles exhibit three independent binding sites with association constants of 2×105, 1.4×104 and 3×102M−1. Upon the addition of 0.1M KCl a third, high affinity site was found on inside out vesicles with an association constant of 3×105, (in 0.1 M KCl). Ca binding to inside out vesicles increased nearly linearly with pH in the, range of pH 4 to pH 11, while binding to right side out vesicles remained practically unchanged in the range of pH 7 to pH 9. Progressive increase of the ionic strength of the medium by the addition of K, Mg or Tris decreased Ca binding to inside out vesicles as did the addition of ATP. Comparison of a series of cation competitors for Ca binding sites on inside out vesicles at 0.003 mM Ca showed that La was the most effective competitor of all while Cd was the most effective divalent cation competitor of those tested. Our findings suggest that the effects of low concentrations of Ca at the inner surface of the red cell membrane are mediated primarily through Ca binding to site 1 (and, possibly site 2) of inside out vesicles of which there are approximately 1.6×105 per equivalent cell.
