    Description / Table of Contents: Abstract Monomeric and “dimeric” malononitrile condense in nonpolar solvents with dimethylamino phenyl propenone to form linear dienes and trienes, resp. 2-Cyano-5-dimethylamino-3-phenyl-2,4-pentadienamide (4) represents an intermediate in the formation of pyridones from enamine ketones and cyanoacetamide. It is cyclised in acidic medium to 2-oxo-4-phenyl-1,2-dihydropyridine-3-carbonitrile (8) (or-3-carboxamide,9).
    Notes: Zusammenfassung In unpolaren Lösungsmitteln kondensieren sich monomeres und “dimeres” Malonitril mit Dimethylamino-phenyl-propenon zu linearen Dienen bzw. Trienen. 2-Cyan-5-dimethylamino-3-phenyl-2,4-pentadienamid (4) kann als Zwischenprodukt für die Bildung von Pyridonen aus Enaminoketonen und Cyanacetamid aufgefaßt werden, da in saurem Medium die Cyclisierung zum 2-Oxo-4-phenyl-1,2-dihydro-pyridin-3-carbonitril bzw. dem Carbonsäureamid gelingt.

    Description / Table of Contents: Zusammenfassung Bei 50 normotonen und 7 hypertonen Schwangeren wurde die Plasmareninaktivität (PRA) mit der Methode nach Boucher bestimmt. Vergleiche mit einem Kontrollkollektiv von 36 Nichtschwangeren führten zu folgenden Ergebnissen: 1. Die PRA ist in der Schwangerschaft etwa um das Dreifache erhöht, ohne daß eine parallele Blutdrucksteigerung feststellbar ist. 2. Zum Ende der Schwangerschaft hin ändert sich die PRA nicht signifikant. 3. Der Hochdruck bei Frauen mit Spätgestose geht nicht mit einer Veränderung des PRA einher. Die Bedeutung reninähnlicher Substanzen für die Pathogenese der Spätgestose wird diskutiert und die Stellung der Befunde im komplizierten Regelkreis des Renin-Angiotensin-Aldosteron-Systems unter Berücksichtigung der Befunde anderer Autoren kritisch diskutiert.
    Notes: Summary Plasma renin activity (PRA) was determined in 50 normal and 7 hypertonic pregnant women according to the method of Boucher. Statistical evaluation was performed by comparing a control group of 36 nonpregnant women. The results were as following: 1. In normal pregnancy PRA is increased threefold without a concomitant rise in blood pressure. 2. There is no significant change in PRA at the end of pregnancy. 3. The rise of blood pressure in late Toxaemic women is hot concomitant with an elevation of PRA. The role of renin like substances in the pathogenesis of late toxaemia is discussed. Our results are also discussed with respect to the renin-angiotensin-aldosteron-system and they are compared to the findings of other investigators.

    Description / Table of Contents: Zusammenfassung Bei 25 normotonen Schwangeren wurden in der Spätschwangerschaft Parallelbestimmungen von Renin und Angiotensinogen (Reninsubstrat) im Plasma durchgeführt. Zum Vergleich diente eine Gruppe von 10 normotonen, gesunden, nichtschwangeren Frauen gleichen Alters. Die Bestimmung von Renin und Reninsubstrat wurden im gleichen Ansatz unter Zusatz des jeweils beteiligten anderen Wirkstoffs im Überschuß durchgeführt. Die Aufarbeitung erfolgte mit einer Modifikation der von Boucher et al. angegebenen Mikromethode unter Verwendung von angereichertem Substrat aus dem Plasma nephrektomierter Schafe. Die Untersuchungen zeigen, daß das zirkulierende Angiotensinogen in der Spätschwangerschaft um mehr als das Dreifache erhöht ist. Das Verhältnis von Reninaktivität zu Angiotensinogen (Reninsubstrat) in der Schwangerschaft ebenfalls signifikant erhöht. Die Befunde werden in Verbindung mit den Theorien über die Entstehung des Bluthochdrucks bei Schwangerschaftstoxikosen kritisch diskutiert.
    Notes: Summary Simultaneous determinations of renin and renin substrate (angiotensinogen) were carried out in 25 normotensive pregnant women during late stages of gestation. For comparison a group of 10 normotensive healthy non pregnant women of comparable age were investigated.—All determinations of renin and renin substrate were done at the same time. To each sample renin or renin substrate, respectively, was added in excess. The procedure was then followed through in a modified manner according to the micro-method of Boucher et al. for determination of renin; purification of renin substrate was done on plasma from nephrectomized sheep. The results show that circulating angiotensinogen is increased more than threefold late in pregnancy. Furthermore the ratio of renin activity/renin substrate (angiotensinogen) is significantly elevated during pregnancy.—The results are reviewed in the light of current concepts pertinent to the etiology of high blood pressure in toxemia of pregnancy.
