    Notes: The enzymatic hydrolysis of lactose has been studied using β-galactosidase (Aspergillus niger) immobilized on collagen. The immobilization process employs simple and inexpensive techniques to bind the enzyme to collagen through noncovalent bond formation. The stability and reusability of collagen bound lactase membranes was evaluated in a biocatalytic modular reactor. Little or no loss in reactor activity was observed after storage for a period of 90 days. Following an initial decrease in activity, stable limit activity was maintained over a number of reactor volume replacements. The activity of the fungal lactase in both the soluble and bound states was determined in ultrafiltered acid whey. The catalytic potency of the enzyme, in both states, showed inhibited enzyme activity in acid whey, as compared to aqueous lactose. In the bound state, the enzyme experienced a partial irreversible inhibition.

    Notes: Abstract Particulate dispersion into and within a 10- to 13-m tall pine forest was studied experimentally at Brookhaven National Laboratory using stained ragweed pollen and other tracers ranging from 14 to 54 Μm in diam. Seventy-two continuous point source releases lasting 20 to 40 min were made at various distances from within the forest edge to 60 m upwind and at heights of 1.75 to 14.0 m. In most experiments, differently colored ragweed pollen was released simultaneously from three locations. Thirty-six longer tests were made using pollen from area sources of ragweed and three with pollen from distant sources. All tests were made during the day with steady winds and unstable lapse rates outside the forest. The sampling network consisted of 119 rotoslide samplers mounted at heights from 0.5 to 21.0 m at 57 positions extending 100 m into the forest. Deposition was sampled by greased microscope slides at each sampling position. Meteorological measurements were taken in and near the forest. Data were classified by particle characteristics; by source type, distance and height; and by meteorological parameters. Isopleths were drawn on scale diagrams of the sampling grid to illustrate concentration patterns. Changes in centerline concentration, crosswind integrated concentration, mass flux, plume width, plume height, deposition, and deposition velocity were related to distance within the forest and other variables. Results were compared to those of similar releases over open terrain and those of previous forest dispersion studies elsewhere. The plume approaching the forest is broadened both vertically and horizontally by increased turbulence at the forest edge and flows mainly into the trunk space and above the forest. Lateral spread is slow within the forest, but vertical spreading beyond the entrance region is greater than in the open. Particles become mixed uniformly below the canopy while appreciable interchange takes place through this layer. Concentration within the forest decreases at a faster rate than in the open, but change in total mass flux within and above the forest is not significantly different. Loss of material takes place by impaction near the forest edge and in the tree tops and by deposition within the forest. Most loss takes place to the foliage rather than the ground, and larger particles are lost faster than smaller ones.

    Notes: Abstract Electrophoretic variation of both the cytoplasmic and mitochondrial forms of the malic enzyme is described in Macaca nemestrina. Pedigree analysis of the observed phenotypes demonstrates that the two subcellular forms of the malic enzyme are genetically independent. The identity of the electrophoretic phenotypes in brain, heart muscle, liver, kidney, adrenal, and spleen from any given individual shows that each subcellular form is determined by the same genetic locus in a wide variety of tissues. After separation by ion exchange chromatography, the cytoplasmic and mitochondrial malic enzymes were shown to be distinct in their heat stability and K m for malate, but no significant differences were found among the variants of the cytoplasmic enzyme or among the variants of the mitochondrial enzyme. It is possible that the polymorphism of the mitochondrial malic enzyme is selectively neutral.

    Notes: Abstract Polymorphic variation of the human mitochondrial malic enzyme was detected in Caucasians and Negroes. The frequencies of the two alleles determining the three mitochondrial malic enzyme phenotypes were 0.69 and 0.31 in the Caucasian sample and 0.82 and 0.18 in the much smaller Negro sample. The distribution of phenotype frequencies among the 85 Caucasian males shows a significant deviation from Hardy-Weinberg equilibrium proportions. Two interconvertible forms of the cytoplasmic malic enzyme are described in human brain tissue, together with one variant in 132 Caucasian individuals.

    Notes: A treatment involving formaldehyde, a reducing agent, and a boron-containing compound of a wool shag carpet imparted to it a flame-retardancy effect that was effective after the seventh but not the eighth shampoo. This treatment also decreased the concentration of carbon monoxide and hydrocyanic acid generated in the products of combustion by at least a factor of 2. Analogous treatment of a wool flannel fabric produced a 15-22% sample area shrinkage and imparted a flame-retardancy effect that was still effective after a 2-hr cold water rinse.

    Notes: Twenty-five organoboron compounds were evaluated for their ability to impart flame retardancy properties to a cotton twill fabric. Only those organoboron compounds in which the carbon-to-boron atom ratio was 4 or less imparted flame retardancy at an approximate 20% add-on level; if a halogen atom was also present in the molecule this ratio could be higher than 4.     Notes: Unilateral ovariectomy of hamsters on day 1 of pregnancy resulted in an increase in weight of the remaining ovary by day 4, primarily by increasing the number of antral follicles. The ovulability of these follicles was tested by injecting 20 IU human chorionic gonadotropin (HCG) on day 12 of pregnancy. Following this treatment, the unilaterally ovariectomized animals ovulated 29.4 eggs whereas intact animals ovulated 16.6 eggs per ovary. In both instances, the number of antral follicles ovulated by HCG was between 40% and 50% of the number present. Luteal weight was unaltered by semispaying, indicating that the regulation of corpus luteum growth does not fit a negative feedback system.The ability of exogenous steroids to prevent ovarian compensation was assessed by injecting either progesterone (2 mg) or estradiol cyclopentylpropionate (1 μg) daily from days 8 to 11 of pregnancy. Either steroid injected into the semispayed hamsters prevented the expected increase in the remaining ovary in weight, follicular development and the ovulatory response to HCG. The same hormonal treatment of intact, pregnant animals slightly increased mean ovarian weight but did not affect follicular development.These results suggest that unilateral ovariectomy of the pregnant hamster, by decreasing peripheral level of ovarian steroids, partially reduces the inhibition of gonadotropin secretion and therefore leads to ovarian compensation.

    Notes: Hamsters injected at 0900 on day 1 of the cycle (metestrus) with either 0, 5 or 15 IU pregnant mare's serum (PMS) were killed at 1500 of days 1 to 4 of the cycle and the ovaries prepared for light microscopy and for a quantitative evaluation of follicular development. In the untreated cyclic hamster, the maximal number of preantral follicles with eight or more layers of granulosa cells occurred between the afternoon of day 4 (proestrus) and day 1, coinciding with the highest blood levels of FSH and LH. It is concluded that the elevated preovulatory levels of gonadotropins not only induce the ovulation of the mature antral follicles but at the same time recruit the next set of follicles for development during the new cycle.By the afternoon of day 1, treatment with either 5 or 15 IU PMS recruited more follicles into large preantral and incipient antral stages than in the untreated hamsters. However, by day 2 the pattern of follicular distribution was similar between the 5 IU PMS and untreated group whereas considerably more antral follicles had differentiated in the animals given 15 IU PMS. The ability of 15 IU PMS to elicit superovulation therefore depends on the levels being initially high enough to mature more follicles at critical stages of their development; the prolonged biological half life of PMS then sustains these follicles throughout the cycle.

    Notes: The surge in plasma luteinizing hormone (LH) in the proestrous hamster begins at 1430 (Turgeon and Greenwald, '72) and this is followed by a sharp increase in follicular and interstitially derived progesterone at 1500 (Norman and Greenwald, '71). The purpose of the present study was to relate various histologic events in the ovary of the preovulatory hamster to these physiological changes, dating from the LH increase between 1430 and 1500. The earliest maturational changes in the oocyte occurred at 1600 as the nuclear membrane began to disappear, correlating with an increase in the number of pycnotic nuclei in the surrounding cumulus cells and an abrupt reduction in mitotic activity in the membrana granulosa. It is possible that the latter event is related to increased progesterone secretion by the follicle at 1500. The most rapid increase in follicular diameter occurred between 2000 and 2200 - five to seven hours after the LH surge and was accompanied by a pronounced stromal edema especially of the medullary portion of the ovary. During this same time period, meiosis proceeded to the metaphase stage and the cumulus cells began to disperse to form the corona radiata. Of the eight hours required for the first meiotic division, four to six hours are spent in metaphase. The majority of animals (75%) ovulated by 0100 and all animals ovulated by 0200. Therefore, ovulation occurred 10 to 11 hours after the LH surge at 1500.
