Summary It is shown that 100 I.U. of ACTH (β-1-25-Corticotropin, synthetic pentacosapeptide DW 75 “Sandoz”), given once daily by intravenous injection over longer periods of time, does not impair the activity of the hypoglycemia-stress-activated adrenotropic center. The time-rhythm of plasma-corticoid concentration is again normalized on the second day after termination of ACTH-therapy. A slight reduction of the morning concentration of plasma corticoids during therapy and an increase of the morning concentration of corticoids in the blood after termination of therapy indicates a slight inhibition of the function of the adrenotropic center followed by a rebound-phenomenon. The effect of ACTH-therapy on adrenotropic function within the system of hypothalamic-pituitary regulation is discussed.

Zusammenfassung Es wird gezeigt, daß eine Dauertherapie mit 100 IE ACTH (β-1-25-Corticotropin, synthetisches Pentacosapeptid DW 75 „Sandoz“), täglich als i.v. Einmalinjektion verabreicht, zu keiner Einschränkung des im Hypoglykämie-Stress aktivierbaren adrenotropen Zentrums führt. Der Chronorrhythmus der Corticoid-Plasma-Spiegel ist am 2. Tag nach Absetzen der Therapie normal. Am leichten Absinken der morgendlichen Corticoid-Plasma-Spiegel während der Therapie mit einer Überhöhung der morgendlichen Corticoid-Konzentrationen im Blut nach Absetzen der Therapie wird eine leichte Dämpfung der Funktion des adrenotropen Zentrums mit einem folgenden Rebound-Phänomen sichtbar. Die Wirkung einer ACTH-Therapie auf die adrenotrope Partialfunktion des hypothalamisch-hypophysären Regelkreises wird diskutiert.
