Notes: Summary Livers of guinea pigs with chronic hepatitis after chronic intoxication with thioacetamide reveal in electron microscopy a reduction of the endoplasmatic reticulum. Gastric mucosa and kidneys are histologically unchanged. The activities of glucuronyl transferase as measured with o-aminophenol andβ-methylumbelliferone are reduced in liver and in gastric mucosa and increased, however, in kidneys. There is a rise in the conjugation of anilin with glucuronic acid in the liver, and there is no conjugation of anilin detectable in gastric mucosa. The conjugation of p-nitrophenol and bilirubin is reduced in the liver and unchanged in the kidneys. These results lead to the suggestions that there exists an individual glucuronyl transferase synthesizing bilirubin glucuronide and a separate enzyme forming N-glycosidic glucuronide linkages and that there are at least two distinct glucuronyl transferases synthesizing o-glycosidic lincages. The concentration of UDPGA in the affected livers is reduced whereas it is unchanged in the kidneys. The activity of UDPG-dehydrogenase remains constant in both organs.

Notes: Summary Zygotes of the marine brown alga,Fucus serratus, have been subjected to the different modes of electric fields. 1) The result of a former study with conductive d.c. fields has been confirmed using electrostatic d.c. fields of 0.5 to 4 V/cm: the zygotes develop the cell polarity axis parallel to the imposed field with the rhizoid pole toward the cathode. 2) The frequency response to both, conductive and electrostatic, a.c. fields represents an optimum curve. The response,i.e. rhizoid formation at either or, in rare cases, both cell poles, peaks at square pulse durations,t E, of 70 to 120 ms. 3) The same frequency response appears if the pulse number is kept constant at 8s−1 by variation of the interval between the pulses,t o. Only fort oo 〉 200 ms,i.e. a pulse number of 3s−1 the response declines markedly. The data support our hypothesis that imposed electric fields induce cell polarityvia differential shift of the membrane potential rather than transcellular current flow. Furthermore, the given dose-response curves strikingly resemble those due to the other morphogenetically active signals: percent response consistently approximates the per cent signal intensity gradient which evokes it.

Notes: Summary Polar regeneration of anucleate posterior stalk segments (PSS) of Acetabularia mediterranea has been studied electrophysiologically. 1. During regeneration the PSS develops a longitudinal, steady potential gradient, U, which is shown to reflect a spatial difference in the transmembrane potential, ΔV m,of 5 to 10 mV, i.e. about 5% of V m.While regeneration becomes visible at about t r=45 h after its light-triggered onset, its sign can be predicted from U not later than at t=29 h, because regeneration occurs at that cell pole where V mis highest, i.e. where the cytoplasm is more negative (Fig. 3, Table 1). 2. Clamping U to comparable values by external circuitry causes the regeneration again to occur where V mis highest, although in this case the flow of the current, I, which controls U is opposite to the direction of the self-generated current, or even changes its sign (Fig. 11, Table 2). 3. Clamping U to zero, i.e. eliminating any transcellular potential gradient, inhibits polar regeneration at least up to t=60 h (Table 3). 4. Spontaneous spikes of U (Fig. 4) and of I (Fig. 12), occur as early as at t=8 h (Fig. 5). These spikes arise at the presumptive regenerating pole with refractory intervals of 10–25 min. They obviously reflect the propagating action potential (Figs. 7–9) although, under the conditions of our present experiments, they spread along the PSS only electrotonically (Fig. 6). 5. Both signals, U and the spikes, are presumably due to changes in the electrogenic Cl-dependent component of V m,rather than its K+-controlled, diffusive component. 6. The data allow to substantiate our working-hypothesis (p. 242) that the mechanism of spatial differentiation involves voltage-dependent plasmalemma properties including excitation.
