Zusammenfassung Es wird eine einfache Technik beschrieben, mit der hydrolytische Enzymreaktionen kontinuierlich verfolgt werden können, bei denen der Chromophor erst in einer Folgereaktion entsteht. Mit Hilfe eines Pumpensystems werden dem Enzymansatz und der Reagenslösung konstante Ströme entnommen und direkt hinter dem Inkubationsgefäß gemischt. Die Konzentration des freigesetzten Farbstoffs wird in einer Durchflußküvette bestimmt. Als Beispiele werden die Spaltung von p-Nitrophenyl-N-acetyl-β-d-glucosaminid und-galaktosaminid durch die menschlichen N-Acetyl-β-d-hexosaminidasen A und B sowie von p-Nitrobrenzcatechinsulfat durch ein Arylsulfatase A-Präparat aus Schweinenieren gezeigt.

Abstract A simple technique is described for the kinetic assay of hydrolytic enzyme reactions. By a second reaction one of the reaction products is continuously transformed into a dye whose extinction can be measured. Using a special pumping system, constant streams are drawn from both incubation mixture and reagent solution and immediately mixed. The reaction kinetics of human N-acetyl-β-d-hexosaminidase A and B and of a preparation of pig kidney arylsulfatase A using p-nitrophenyl-N-acetyl-β-d-glucosaminide and -galactosaminide and p-nitrocatechol-sulphate as respective substrates are shown.
