Zusammenfassung Mit gereinigtem Kontaktaktivierungsprodukt (KAP) lassen sich unter Stasebedingungen Thromben erzeugen; hierbei treten Gerinnungsveränderungen auf, die einer Verbrauchscoagulopathie entsprechen. Es wurde daher an 92 Kaninchen unter verschiedenen Versuchsbedingungen die Frage geprüft, ob das KAP nicht nur typisch lokalisierte Thromben in einem Stasegebiet, sondern auch Gerinnungsthromben bzw. eine disseminierte intravasale Gerinnung (DIC) wie bei einer Verbrauchscoagulopathie auslösen kann. In einer ersten Versuchsgruppe erhielten die Tiere KAP als einmalige Injektion (0,8 ml/kg Körpergewicht), wobei eine V. jugularis unterbunden wurde, in einer zweiten Gruppe wurde KAP an mehreren aufeinanderfolgenden Tagen wiederholt injiziert. In einer dritten Gruppe erfolgte nach einer vorherigen Injektion von Endotoxin eine KAP-Gabe, wobei die Wessler-Technik (Ligatur einer Vena jugularis) angewandt wurde, in der vierten Versuchsgruppe wurde KAP nach Endotoxin gegeben ohne zusätzliche V. jugularis-Ligatur. Nach KAP-Injektion entstanden sowohl bei einmaliger als auch bei wiederholter Injektion Abscheidungsthromben außerhalb des Stasebereiches. Auch nach vorheriger Endotoxininjektion ließen sich Abscheidungsthromben in Lungenschlagadern und in der Herzkammer nachweisen. Disseminierte intravasale Präcipitate wie bei einer Verbrauchscoagulopathie waren nach KAP-Gabe trotz typischer Gerinnungsveränderungen nicht zu finden.

Summary By use of purified contact activation product (CAP) it is possible to produce stasis thrombi. The changes in parameters of the clotting system are similar to those observed in consumptive coagulopathy or disseminated intravascular clotting (DIC). It was the aim of the present study to examine whether by use of CAP it would be possible to not only produce typical localized stasis thrombi but also mixed thrombi or DIC which are characteristic for consumptive coagulopathy. 92 rabbits were divided into four groups. The animals of the first group received 0.8 ml per kilogram body weight as a single intravenous injection. In the second group, CAP was injected repeatedly on 4 consecutive days. In the third group CAP was injected following preteatment with endotoxin using the Wessler technic (ligation of the jugular vein) and in the fourth group CAP was injected following endotoxin administration but without ligation of the jugular vein. In the animals which had received single or multiple injections of CAP there were agglutinative thrombi beyond the areas of stasis. Similar agglutinative thrombi were also observed in the pulmonary arteries and in the left ventricle of the heart following pretreatment with endotoxin. In spite of typical changes of coagulation parameters in all experimental animals, DIC occurred only in the endotoxin-treated animals but not following treatment with CAP alone.
