Summary After i.p. application of 2 mg sulfanilamid to rats the rate of acetylation is 66% of the urinary sulfanilamid. It amounts to 32% after 48 mg. There is a linear decrease of acetylation in relation to the logarithmic of the dose. Above 10 mg the slope becomes steeper indicative of an inhibition of acetokinase by the substrate. Acetylation of sulfanilamid is diminished by some 25% after treatment with toxic doses of inorganic mercury, lead or arsenic for 5–7 days. The organic derivative of mercury called Mersalyl and also bismuth which was given for 23 days both were inactive in this respect. The reduced acetylation of sulfanilamid after incorporation of mercury, lead or arsenic can be explained by an impaired synthesis of coenzym A.

Zusammenfassung Nach der Applikation von 2 mg Sulfanilamid pro Ratte sind 66% des im Harn ausgeschiedenen Sulfanilamids acetyliert, bei 48 mg Sulfanilamid pro Ratte beträgt der acetylierte Anteil nur 32%. Die Abhängigkeit der Acetylierungsrate vom Logarithmus der applizierten Dosis ist gradlinig, sie hat jedoch oberhalb 10 mg eine größere Steilheit als bei kleineren Dosen, so daß eine zusätzliche Substrathemmung der Acetokinase wahrscheinlich ist. Nach mehr als 5–7 tägiger Zufuhr von anorganischem Quecksilber, Blei oder Arsen in toxischen Dosen ist die Acetylierung von Sulfanilamid um etwa 25% vermindert. Kürzere Behandlungen haben diesen Effekt nicht. In organischer Bindung z. B. als Mersalyl (Salyrgan) ist Quecksilber unwirksam. Wismut hatte auch nach 23 tägiger Applikation keinen Einfluß. Der Mechanismus des beobachteten Effektes wird diskutiert und postuliert, daß eine Störung der Synthese von Coenzym A dafür verantwortlich sein könnte.
