This paper reports autoradiographic studies of protein synthesis related to epidermal cornification in several different species. A high concentration of injected histidine appeared in granular cells of man, the monkey, guinea pig, hairless mouse, and newborn rat, indicating that synthesis of relatively histidine-rich protein is involved in formation of keratohyalin granules. Two steps in the cornification process: synthesis of this histidine-rich protein, in addition to protein synthesized in the lower layers of the epidermis are postulated in epidermis containing keratohyalin granules. In the epidermis of the turtle, which does not contain keratohyalin granules, concentration of histidine is not observed, suggesting that protein of the cornified layer in this species seems to be synthesized as a 1-stage process.
