    Notizen: Abstract— —The levels of the depressant amino acids found in appreciable amounts in cord extracts—α-alanine, cystathionine, GABA, glycine and serine—were not significantly influenced by tetanus toxin. This supports the view that the antagonism of spinal inhibition by the toxin is the result of an interference with transmitter release rather than a reduction in the amount of transmitter available for release.The marked increase in aspartic acid levels found in the spinal cord after treatment with tetanus toxin may reflect the association of aspartic acid with the increased activity of spinal excitatory interneurones or the involvement of aspartic acid as a glycine precursor in spinal tissue.

    Notizen: Abstract Electrophoretically administered morphine and codeine, but not meperidine, blocked the inhibitory effect of glycine on spinal neurones, without reducing the effectiveness of GABA. This action of morphine was similar to, but much weaker than, that of strychnine. When administered intravenously in divided doses to circumvent marked circulatory disturbances, morphine reduced the direct and recurrent inhibition of spinal motoneurones in a parallel fashion. Again morphine was a very weak strychnine-like substance, doses in excess of 50 mg/kg being required to block these disynaptic spinal inhibitions. The results are not incompatible with the function of a glycine-like amino acid as the inhibitory transmitter for both the direct and recurrent spinal inhibitory pathway.

    Notizen: Summary An analysis has been made in anaesthetised cats of the depression by glycine and related amino acids of the firing of spinal dorsal horn interneurones, Renshaw cells and cortical neurones. In general, electrophoretically administered glycine was a more potent depressant of interneurones than GABA. The reverse was true for cortical neurones, whereas these two amino acids were approximately equally effective upon Renshaw cells. Strychnine blocked the depressant action of α- and β-amino acids, but not that of γ- and higher ω-amino acids. Only convulsants having a strychnine-like effect on spinal post-synaptic inhibition blocked the action of glycine. The depression of spinal neurones produced by glycine or GABA was not affected by structural analogues of glycine and GABA that were not depressants, or by substances influencing amino acid transport systems. Some evidence was obtained for the enzymic inactivation of electrophoretically administered glycine in spinal tissue. The results are discussed in terms of the involvement of a glycine-like amino acid as a major spinal inhibitory transmitter.
