Summary Fifty five purple spontaneous mutants of the haploid fission yeast Schizosaccharomyces pombe have been isolated and distributed, by genetic analyses, between two loci (ad-6 and ad-7) which control two independent steps in the purine biosynthesis. The molecular nature of these mutants has been determined by both their physiological behaviour (leakiness, temperature-sensitivity, pH-sensitivity) and their interallelic complementation ability. It is concluded that during the mitotic cycle the large majority of spontaneous mutations are base-pair substitutions.
