Summary In a case of generalized glycogenosis (type II) it has been examined whether the brain also stores other polysaccharides besides glycogen. In circumscribed areas of the cortex and especially of the white matter of the telencephalon intracytoplasmatic, fine granular, acid, mucopolysaccharid-like substances have been proofed by histochemical methods. Together with the glycogen they preferably are found in astrocytes of the white matter, in smaller amounts in the oligodendroand Hortega-glia of the white matter and cortex, in endothelia of capillaries and also in cortical nerve cells. Their histochemical reactions agree with those of the “basophilic substances” of skeletal muscles and correspond to inhomogeneous complex substances. They are partially water-soluble, chromotropic, stainable with acid astrablue solution and react negative to medium positive with PJS. The biochemical analysis byEdgar (Heemstede) brought forward a considerable increase of non-lipid-soluble hexosamine. The discussion emphasises the possible relations between the histochemical and the biochemical findings and the derivation of the mucopolysaccharides associated with the glycogen storage disease.

Zusammenfassung Bei einem Fall von generalisierter Glykogenose (Typ. II) wurde die Frage untersucht, ob das Gehirn neben dem Glykogen noch andere Polysaccharide speichert. In umschriebenen Gebieten der Rinden-und besonders der Marksubstanz des Telencephalon gelang der histochemische Nachweis von intracytoplasmatischen, feingranulären,sauren mucopolysccharidartigen. Substanzen. Sie sind zusammen mit dem Glykogen vorzugsweise in den Astrocyten des Markes und geringfügig in der Oligodendro-und Hortegaglia der Mark-und Rindensubstanz, den Capillarendothelien sowie den corticalen Nervenzellen eingelagert. Ihr histochemisches Verhalten läßt übereinstimmend mit den “basophilen Substanzen” der Skeletmuskulatur auf uneinheitlich zusammengesetzte Stoffe schließen. Sie sind wasserempfindlich., chromotropiefähig, färbbar mit sauren Astrablaulösungen und geben eine negative bis mittelgradig positive PJS-Reaktion. Die biochemische Untersuchung durchEdgar (Heemstede) erbrachte im Großhirnmark eine beträchtliche Erhöhung des nicht-lipidlöslichen Hexosamins. In der Diskussion wird u. a. auf die möglichen Beziehungen zwischen den histochemischen und biochemischen Befunden und auf die Entstehung der Mucopolysaccharide bei der Glykogenspeicherkrankheit eingegangen.
