Zusammenfassung Es wird über eine erhöhte Tumorhäufigkeit bei Ratten nach Inhalation der hochpolymeren wasserlöslichen synthetischen Substanz Poly-2-vinylpyridin-N-oxid (PVNO) berichtet. Die Tiere der PVNO-Gruppe, konventionelle Sprague-Dawley-Ratten, inhalierten ein PVNO-Aerosol in einer Konzentration von 75 mg/m3 täglich an 5 Wochentagen entweder im Anschluß an die Bestaubung 20 min bzw. 180 min oder ohne vorherige Bestaubung 180 min. Der Versuch dauerte insgesamt 22 Monate; nach 18 Monaten Versuchszeit wurde die Staub- und Aerosolinhalation beendet. Die Tumorhäufigkeit ist in der Gesamt-PVNO-Gruppe mit 13,1% statistisch hochsignifikant unterschiedlich gegenüber der Nicht-PVNO-Gruppe mit 2,7%. Die Tumortiere der Gesamt-PVNO-Gruppe haben durchschnittlich mehr als einen Tumor je Tier. Es ist ein hoher Anteil von Tumoren mit Lokalisation in der Lunge vorhanden sowie ein breiteres Spektrum hinsichtlich der betroffenen Organe. Auf Grund der Ergebnisse der vorliegenden Untersuchung kann zunächst nur von einer beobachteten Erhöhung der Tumorhäufigkeit und nicht von Polymertumoren gesprochen werden, da zu dieser Frage noch weitere Untersuchungen notwendig sind.

Summary An increased incidence-rate of tumors in rats is reported after the inhalation of the high-polymeric, water-soluble, synthetic substance poly-vinylpyridine-N-oxid (PVNO). The animals of the PVNO group, conventional Sprague-Dawley rats, inhaled daily, on 5 weekdays, a PVNO aerosol in a concentration of 75 mg/m3 for 20 min and 180 min respectively following dusting or for 180 min without previous dusting. The experiment lasted for 22 months in the whole, dust and aerosol inhalations were finished after an experimental time of 18 months. The tumor incidence-rate — 13.1% in the total PVNO group — is, statistically seen, significantly different as compared to the non-PVNO group with 2.7%. The animals of the total group of PVNO afflicted with tumors have, in the average, more than one tumor per animal. A high portion of tumors localized to the lungs is present as well as a broader spectrum with regard to the afflicted organs. These findings are most marked within the group inhaling dust followed by PVNO for 20 min. On account of the results of the present investigation one can only speak up to now of an elevated tumor incidence-rate and not of polymer tumors. To give this interpretation, further investigations would still be necessary.
