    Notes: Abstract The12C+n total cross section has been studied in the region of the lowestT=3/2 states of13C. The firstT=3/2 state at (15108.2±1.2) keV excitation is observed as a weak resonance anomaly. The deduced resonance parameters agree with previous results. At higher excitation energies four sharp anomalies have been observed at (17534±3) keV, (18082±3) keV, (20057±4) keV and (21703±4) keV excitation with total widths between 12 keV and 20 keV. The results are discussed with respect to a possibleT=3/2 assignment. An upper limit of the elasticity (J+1/2) Γno/gG is deduced for thoseT=3/2 levels which do not appear as resonance anomalies.

    Notes: Quantitative hemolysis assays of acrylate and methacrylate esters provided estimates of the intrinsic hemolytic activity (Hi the slope of the concentration-response curve) and the concentrations effecting 5% (H5) and 50% (H50) hemolysis. The dependence of hemolytic activity and LD50 (mice) on physical properties (lipophilicity, molar refraction, and molecular volume) of the esters was determined by multiple regression analysis. The observed correlations were: Hi, R2 = 0.94; H5, R2 = 0.95; H50, R2 = 0.94; and LD50, R2 (all compounds) = 0.80, R2 (all compounds less the methyl esters) = 0.94. The difference of the methyl esters was associated with the smaller steric volume of the methyl ester substituent and the presence (methacrylates) or absence (acrylates) of the branched methyl group. Associative steric contributions of the branched methyl group and the ester substituents were probably responsible for greater variability in the methyacrylate series. The results were consistent with the conclusion that the mechanism of the action of the esters is membrane mediated and relatively nonspecific and that in vivo biotransformation was not a significant factor. Also, long-term toxic liability of the esters may be more closely related to intrinsic toxicity than acute toxicity.

    Notes: In order to reduce the uncertainties involved in the morphologic evaluation of the cell culture agar diffusion test, the originally recommended neutral red stain was replaced by fluoresceindiacetate (FDA). This stain proved to be nontoxic in the concentrations used in this study (0.002%). Toxicity tests with phenol, formalin, and methylmethacrylate monomer revealed results corresponding to literature evaluations by other methods. Since FDA makes cell metabolism visible and easily accessible to automated evaluation techniques, it presents certain advantages over the morphologic evaluation using the neutral red stain.

    Notes: The mutagenic potential of glutaraldehyde-containing dentin bonding agents was shown in previous studies using a bacterial gene mutation assay, the Ames test. However, current strategies of genotoxicity testing and regulatory requirements for the biological evaluation of medical devices recommend a battery of tests that indicate induced mutations in prokaryotic and eukaryotic cells. Accordingly, the mutagenicity of three glutaraldehyde-containing bonding agents (Syntac adhesive, Prisma Universal Bond 3 adhesive, and Gluma 3) was investigated using a quantitative mammalian cell gene mutation assay (V79/HPRT test) in the present investigation. The materials were extracted in dimethyl sulfoxide (0.1 g/2 mL) for 24 h and original extracts were then serially diluted in cell culture medium before exposure to V79 cells. Cytotoxic and mutagenic effects were observed with identical concentrations of extracts of the different test materials. There was a moderate decrease of the number of surviving cells immediately after the end of exposure. Mutagenicity at the hprt locus in V79 cells was found with all materials tested, and the increases in the absolute numbers of mutants were dose dependent. The mutant frequencies were about 15- (Syntac adhesive and Gluma 3) to 20-fold (Prisma UB3 adhesive) higher than solvent control values. Since other substances than glutaraldehyde may be responsible for the mutagenic effects in mammalian cells in this study, work is currently in progress to identify the individual mutagenic compounds of dentin adhesives and related composite materials. © 1997 John Wiley & Sons, Inc.     Notes: Aside from the considerable number of reports on the physical and chemical properties of dental bonding agents, information concerning their biologic effects is sparse. Three dentin bonding agents (Prisma Universal Bond, Pertac Universal Bond, and Syntac) and the ingredients methylmethacrylate, 2-hydroxyethyl-methacrylate, and glutaraldehyde were investigated in the Salmonella typhimurium mutagenicity test system using five different bacterial strains (TA97a, TA98, TA100, TA102, and TA104). The materials as well as the ingredients were eluted in both dimethyl sulfoxide and physiologic saline, and serially diluted eluates were used in the plate incorporation test. Pertac Universal Bond and Prisma Universal Bond did not elicit any mutagenic effects in any of the bacterial strains. In contrast, Syntac adhesive showed clear mutagenicity in S. typhimurium strain TA102. Dimethyl sulfoxide eluates, as well as physiologic saline eluates of the Syntac bonding agent, caused numbers of revertants that were about 6 times higher than control values. Reversion rates with other strains were moderately enhanced. Glutaraldehyde, an ingredient of Syntac adhesives, caused mutagenicity in a manner similar to Syntac adhesive eluates. Neither 2-hydroxyethyl-methacrylate nor methylmethacrylate monomer was found to be mutagenic over a broad concentration range. © 1994 John Wiley & Sons, Inc.
