    Description / Table of Contents: Zusammenfassung Die spezifische gegen eine akute Leukämie gerichtete Wirksamkeit einer hochdosierten Prednisolon-Behandlung ist 24–48 Std nach Therapiebeginn an einem Aktivitätsabfall der bei der DNS-Synthese beteiligten Enzyme Thymidin-Kinase und DNS-Polymerase sowie der Einbauraten von DNS-Vorstufen — Thymidin und Desoxyuridin — nachweisbar. Entsprechende Stoffwechseluntersuchungen an leukämischen Blasten bei akuten Leukämien im Erwachsenen- und Kindesalter zeigen, daß sich die antileukämische Wirksamkeit der Corticosteroide auf die akuten Lymphoblasten- und undifferenzierten Leukämien beschränkt. Diese Ergebnisse werden durch klinische Verlaufsbeobachtungen und Angaben in der Literatur bestätigt. Es ergibt sich daraus, daß nur bei der akuten Lymphoblasten- und undifferenzierten Leukämie eine Indikation zur Behandlung mit Corticosteroiden in therapeutisch wirksamen Dosen besteht.
    Notes: Summary The specific action of high dose prednisolone treatment against acute leukemia is detected 24–48 hours after the initiation of treatment by decrease of enzyme activities involved in DNA-synthesis—thymidine-kinase and DNA-poly-merase—and of the incorporation rates of DNA-precursors—thymidine and deoxyuridine. Biochemical investigations of leukemic blast cells of acute leukemias in adults and children show, that the antileukemic effects of corticosteroids are limited to the acute lymphoblastic and undifferentiated leukemia. These results are confirmed by clinical observations and by different reports in the literature. Therefore high doses of corticosteroids are only valuable for the treatment of acute lymphoblastic and undifferentiated leukemia.

    Description / Table of Contents: Zusammenfassung In der vorliegenden Arbeit werden die an 395 Proben von Blut und Knochenmark von 119 Patienten gewonnenen durchflußphotometrischen Nucleinsäurebestimmungen vorgelegt. Der Vergleich mit feulgenphotometrischen DNS-Untersuchungen zeigte nur in 3 von 35 Fällen einen allein methodisch erklärbaren Unterschied in den Ergebnissen. Bei 8 der Kranken mit Hämoblastose, bei denen erhöhte DNS-Werte Folge einer Hyperdiploidie sein könnten, wurde zweimal zusätzlich eine Stammlinie bei tetraploiden Werten gefunden, aber in keinem Fall Aneuploidien, die zwischen 2c und 4c liegende DNS-Werte erklären könnten. Die gewonnenen Ergebnisse quantitativer DNS-Untersuchungen können deshalb auch bei den Kranken mit Hämoblastosen nach dem Konzept G0, G1-S-(G2) gedeutet werden. Danach lassen sich Störungen des Zellcyclus sowohl im Übergang von G1 nach S, innerhalb der S-Phase wie in der G2-Phase erfassen. Wie bisher bereits angenommen, erfolgt eine Regulation vorwiegend durch eine Blockade des G1-S-Überganges. Die in 55 Fällen gleichzeitig durchgeführten3H-Thymidinautoradiographischen Untersuchungen gestatten bei dem statistischen Vergleich mit den durchflußphotometrisch gewonnenen Nucleinsäurebestimmungen die Annahme, daß eine ineffektive Zellneubildung durch ein Ausscheren von S-Phase-Zellen aus dem Cyclus erfolgt, und sowohl bei akuten Leukämien wie bei Panmyelopathien, wie in der untersuchten Gruppe der Anämien bei Urämie eine große Bedeutung hat. Eine ineffektive Blutzellneubildung durch Austritt aus dem Cyclus nach Beginn der DNS-Reduplikation scheint damit häufiger zu sein als bisher angenommen. Die Methode der durchflußphotometrischen Nucleinsäurebestimmung ist nicht geeignet, um bei niedrigem Anteil an S- und G2-Phase-Zellen Verschiebungen innerhalb der S-Phase auch unter Behandlung mit Cytostatika mit ausreichender Genauigkeit zu erkennen, da die Befunde durch die breite Streuung der DNS-Werte der G0/G1-Zellen überdeckt werden können.
    Notes: Summary The authors discuss the results of the pulse-cytophotometric nucleic acid analysis obtained in 395 tests of blood and bone marrow from 119 patients. Compared with Feulgen-photometric DNA investigations, a difference was found in only 3 of 35 cases. This difference may be attributed to the various methods applied. In the study of the chromosomes, an additional stem-line with tetraploid values was seen twice in eight cases of haemoblastosis. In these cases the increased DNA values might be a consequence of hyperdiploid cells. In no case however were any aneuploid cells observed which could be responsible for DNA values between 2c and 4c. Therefore, the results of quantitative DNA analysis can be adscribed also to patients with haemoblastosis according to the concept G0, G1-S-(G2). Disturbances of the cell cycle can by these means be registered in the transitional period from G1 to S, as well as within the S-phase and the G2-phase. We found that a regulation mainly occurs by blocking the transitional period from G1 to S. 553H-thymidin-autoradiographic investigations were made concomitantly on the same material and the results statistically compared with nucleic acid analysis carried out by means of cytophotometry. According to our results we assume that an ineffective cell- renewal results from S-phase cells out of the cycle. This fact has been observed with acute leukaemia as well as with panmyelophthisis and with the tested groups of anaemia caused by uraemia. We suppose that an ineffective renewal of blood cells by out-of-cycle cells after the beginning of the DNA reduplication is probably more frequent than assumed until now. It has to be said however that the pulse-cytophotometric nucleic acid analysis is not a method suitable for the very exact diagnosis of the shifting within the S-phase in cases with a low part of S- and G2-phase cells, because the diagnosis could be veiled by a large dispersion of the DNA-values of G0/G1 cells.

    Notes: Abstract The isotopes15B,18C,19,20N,34,35Al and39P have been produced as projectile-like fragments from the interaction of a86Kr beam of 45 MeV/u from GANIL with a181Ta target. Separated by the 0° magnetic analyser LISE, the nuclei are identified by means of aΔE- E semiconductor telescope. It is surrounded, in a geometry close to 4π, by a thin plastic scintillator (NE 102) and a large volume liquid scintillator (NE 213) for the detection of betas and neutrons, respectively, from the decay of the implanted nuclei. By observation of theβ-delayed neutrons, the half-livesT 1/2 and neutron emission probabilitiesP n have been measured. TheT 1/2 of18C,20N,35Al,39P and Pn of18C,19,20N,34,35Al and39P are determined for the first time. For the isotopes17C,36,37Si and38P upper limits of Pn are deduced.

    Notes: Abstract Total nuclear reaction cross-sections are determined by means of a 4π-γ method. The results cover a wide span of targets for various stable beams. The validity of the method is shown in a combined systematics including also the results of transmission-type experiments. The data are very well described by the formula developed by Kox et al. The same method is applied to secondary fragment beams produced from a 44 MeV/u22Ne beam on a 332mg/cm2 181Ta target. Using the LISE spectrometer the fragments4, 6He,6–9, 11Li,7, 9–12, 14Be,10–15, 17B11–19C,13–19N,15–21O,18– 21F and20,21 Ne are analyzed and transported to interact with a 199.4 mg/cm2 Cu target surrounded by a 4π-γ counter. The measured total reaction cross-sectionsσ R are discussed in terms of the reduced strong absorption radiusr 0 and compared with other experimental results.

    Notes: Abstract We have measured production cross-sections of the new neutron-rich isotopes58Ti,61V,63Cr,66Mn,69Fe,71Co and neighbouring isotopes that have been identified as projectile fragments from reactions between a 500 MeV/u86Kr beam and a beryllium target. The isotope identification was performed with the zero-degree magnetic spectrometer FRS at GSI, using in addition time-of-flight and energy-loss measurements. The experimental production cross-sections for the new nuclides and neighbouring isotopes are compared with an empirical parametrization. The resulting prospects for reaching even more neutron-rich isotopes, such as the doubly-magic nuclide78Ni, are discussed.

    Notes: A tandem mass spectrometric method is described which allows the assignment of stereochemistry to fragment ions comprising intact sugar subunits of larger glycosides without chemical degradation and product isolation by chromatography. The approach relies on the mass selection of the ‘sugar ion’ of interest followed by analysis of stereoselective fragmentation induced by low-energy collisional activation. The daughter ion spectra provide configurational fingerprints of the selected sugar ions which can be matched for identity with reference spectra obtained from suitable precursors of known stereochemistry. Glucose, mannose and galactose furnished the required set of the most important reference ions. By using peracetyl (and perdeuterioacetyl) derivatives, galactose was readily identified as the glycosidic sugar constituent of the (known) antibiotic papulacandin B and a further (unknown) congener.

    Notes: Various derivatives of a set of three isomeric (linear and branched) pentasaccharides (LNF-1, LNF-2 and LNF-3) were prepared and investigated by high-performance tandem mass spectrometry in order to compare their collision-induced dissociation (CID) behaviour. The fast atom bombardment tandem mass spectra of [M + H]+ ions of peracetylated derivatives mainly display fragments containing the non-reducing terminus (B-type ions) and allow a straightforward assignment of sugar sequence and branching together with the identification of some interglycosidic linkages. Permethylated derivatives, which better accommodate the mass range of tandem mass spectrometers in the case of larger oligosaccharides, yield similar results, i.e. predominance of B ions, but the necessary information has to be retrieved from incomplete Bi and Yi ion series. By contrast, CID of permethyl or peracetyl derivatives carrying a preformed charge due to prior reductive amination of the oligosaccharides with trimethyl(p-aminophenyl)ammonium chloride yields exclusively fragment ions comprising the reducing end. In this case, the four distinct series of fragments observed involve charge-remote fragmentation processes. As a consequence, the spectral patterns are not significantly affected by the nature of the sugar O-substituents additionally introduced (i.e. methyl or acetyl).
