    Notizen: Abstract This paper describes a method for learning flexible concepts, by which are meant concepts that lack precise definition and are context-dependent. To describe such concepts, the method employs a two-tiered representation, in which the first tier captures explicitly basic concept properties, and the second tier characterizes allowable concept's modifications and context dependency. In the proposed method, the first tier, called Base Concept Representation (BCR), is created in two phases. In phase 1, the AQ-15 rule learning program is applied to induce a complete and consistent concept description from supplied examples. In phase 2, this description is optimized according to a domain-dependent quality criterion. The second tier, called the inferential concept interpretation (ICI), consists of a procedure for flexible matching, and a set of inference rules. The proposed method has been implemented in the POSEIDON system, and experimentally tested on two real-world problems: learning the concept of an acceptable union contract, and learning voting patterns of Republicans and Democrats in the U.S. Congress. For comparison, a few other learning methods were also applied to the same problems. These methods included simple variants of exemplar-based learning, and an ID-3-type decision tree learning, implemented in the ASSISTANT program. In the experiments, POSEIDON generated concept descriptions that were both, more accurate and also substantially simpler than those produced by the other methods.

    Notizen: Abstract Motivated by the method of Martinez and Qi (Ref. 1), we propose in this paper a globally convergent inexact generalized Newton method to solve unconstrained optimization problems in which the objective functions have Lipschitz continuous gradient functions, but are not twice differentiable. This method is implementable, globally convergent, and produces monotonically decreasing function values. We prove that the method has locally superlinear convergence or even quadratic convergence rate under some mild conditions, which do not assume the convexity of the functions.

    Notizen: A six-node plate bending element has been developed by employing mixed formulation based on a modified Hellinger-Reissner principle and the Reissner-Mindlin plate bending theory. The numerical result indicates that, among the types of assumed independent transverse shear strains considered, a combination of 2α version with either 5α version or 6α version is free of spurious kinematic modes and leads to accurate and reliable solutions even for very thin plates.

    Notizen: Optically active isotactic polychloral was prepared in which the optical activity comes exclusively from molecular asymmetry (i.e., helical conformation). Molecular asymmetry requires a high conformational energy barrier for the polymer backbone, and an asymmetric initiator to induce a predominance of one helical screw-sense. Polychloral meets the criteria. Asymmetric initiators used to obtain optically active polychloral include tetramethyl ammonium (+)- or (-)-0-acetylmandelate, tetramethylammonium (+)- or (-)-0-methylmandelate, the lithiumalkoxides of methyl (+)- or (-)-mandelate, lithium cholesten-3β-oxide, lithium cholestan-3β-oxide, and lithium (+)- or (-)-2-octanoxide. Using the above initiators at 0.5 mol %, a maximum specific rotation of [α] D25 = 5000 was obtained for polychloral. Errors in specific rotation were typically ±7%.

    Notizen: The non-linear field-boundary-element technique is applied to the analysis of snap-through phenomena in thin shallow shells. The equilibrium path is traced by using the arc-length method and the solution strategy is discussed in detail. The results show that, as compared to the approaches based on the popular symmetric-variational Galerkin finite element formulation, the current approach based on an unsymmetric variational Petrov-Galerkin field-boundary-element formulation gives a faster convergence while using fewer degrees of freedom. The illustrative numerical examples deal with post-buckling responses of several shallow shells with different geometries.

    Notizen: The baculovirus/insect cell expression system has provided a vital tool to produce a high level of active proteins for many applications. We have developed a very high-density insect cell perfusion process with an ultrasonic filter as a cell retention device. The separation efficiency of the filter was studied under various operating conditions. A cell density of over 30 million cells/mL was achieved in a controlled perfusion bioreactor and cell viability remained greater than 90%. Sf9 cells from a high-density culture and a spinner culture were infected with two recombinant baculoviruses expressing genes for the production of human chitinase and monocyte-colony inhibition factor. The protein yield on a cell basis from infecting high-density Sf9 cells was the same as or higher than that from the spinner Sf9 culture. Virus production from the high-density culture was similar to that from the spinner culture. The results show that the ultrasonic filter did not affect insect cells' ability to support protein expression and virus production following infection with baculovirus. The potential applications of the high-density perfusion culture for large-scale protein expression from Sf9 cells are also highlighted. © 1998 John Wiley & Sons, Inc.     Notizen: Long-chain unsaturated fatty acids (UFA) can easily be converted on a microgram scale to the corresponding 2-alkenyl-4,4-dimethyloxazolines by condensation with 2-amino-2-methylpropanol (AMP). These modified molecules with a ‘hidden’ carboxyl group have been proved to be a class of useful derivatives for gas chromatography/mass spectrometry (GC/MS) of UFA mixtures. While possessing very good GC characteristics, the title compounds show regular, well-recognizable diagnostic ion peaks of the double bond position in the chain. Detailed description of the method as well as electron impact (E1) mass spectra of derivatives resulting from mono-, di and polyenoic (maximally containing six double bonds) acids are presented.

    Notizen: Abstract Trust-region algorithms solve a trust-region subproblem at each iteration. Among the methods solving the subproblem, the optimal path algorithm obtains the solution to the subproblem in full-dimensional space by using the eigenvalues and eigenvectors of the system. Although the idea is attractive, the existing optimal path method seems impractical because, in addition to factorization, it requires either the calculation of the full eigensystem of a matrix or repeated factorizations of matrices at each iteration. In this paper, we propose a scaled optimal path trust-region algorithm. The algorithm finds a solution of the subproblem in full-dimensional space by just one Bunch–Parlett factorization for symmetric matrices at each iteration and by using the resulting unit lower triangular factor to scale the variables in the problem. A scaled optimal path can then be formed easily. The algorithm has good convergence properties under commonly used conditions. Computational results for small-scale and large-scale optimization problems are presented which show that the algorithm is robust and effective.

    Notizen: Abstract In Ref. 1, Nocedal and Overton proposed a two-sided projected Hessian updating technique for equality constrained optimization problems. Although local two-step Q-superlinear rate was proved, its global convergence is not assured. In this paper, we suggest a trust-region-type, two-sided, projected quasi-Newton method, which preserves the local two-step superlinear convergence of the original algorithm and also ensures global convergence. The subproblem that we propose is as simple as the one often used when solving unconstrained optimization problems by trust-region strategies and therefore is easy to implement.
