    Notes: Abstract Neon-like niobium X-ray lasers have been studied using both slab and thin-foil target geometries. Niobium foils of various lengths were irradiated from both sides with two beams of the frequency-doubled Nova laser system using a line focus. We looked for gain by measuring spectrally integrated line intensities at different plasma lengths. Gain was observed in four neon-like niobium lines corresponding to 3s−3p transitions. The line profile of theJ = 0−1 line (λ = 145.9 Å) shows splitting due to the hyperfine effect. Improved contrast in the hyperfine structure is observed as the plasma length is increased. Hyperfine splitting may be relevant to other 3s−3p transitions in neon-like niobium as well as other neon-like X-ray laser systems.

    Notes: Summary This study compares two simplified techniques, which use only 2 and 4 sutures respectively, with a conventional technique with 8 sutures for fallopian tube anastomosis. Experimentally these techniques were performed on the uterine horns of fifteen female rats. A 100% patency rate was obtained with all three techniques. No difference in the mucosal, muscular and serosal regeneration was observed in the three groups at 10, 20 and 60 days. Two months after surgery, the serosa, muscularis and mucosal layers were completely continuous in all groups. The operating with 2 sutures (5′ 30″±1′ 10″) was significantly less than with 4 (9′09″±0′55″,P〈0.05 ANOVA) and 8 sutures (15′12″ ± 1′41″,P〈0.05 ANOVA). A minimum inflammatory reaction to sutures was observed in all three groups at 60 days after surgery. The results suggest that with 2 sutures are all that is needed for tissue repair.

    Notes: Summary A one centimeter length of the left uterine horn in two groups of Wistar rats (n=10) was replaced with normal (VAS group) or with denervated and devascularizated (NVAS group) colonic grafts. All animals maintained pregnancy in the right control horns but not in the grafted horns. At 40, 60 and 90 days after surgery, the light microscopic appearance of the autografts was studied. In the VAS group, and with respect to the last period, the number (5.5±0.7) and height (1.0±0.1 mm,P〈0.05 ANOVA) of the folds, the intestinal glands height (160.2±21.2μ,P〈0.05 ANOVA) and the number of globet cells per gland (26.6±4.2,P〈0.05 ANOVA) had decreased in relation to the colon control (6.0±0.7, 1.4±0.1, 251.7±31.8, 42.6±5.2 respectively). A similar intestinal structure to that described above was observed in the anastomosis areas of the NVAS group, and therefore a decreased mucus production was maintained in this areas. No folds or intestinal glands were observed, but a monoestratified cubic epithelial cells type was observed along 63.7±4.1% of the NVAS colonic graft center. Under this epithelium a connective tissue, like a non-glandular submucosa, was obtained.

    Notes: Abtract  Foetal calf serum (FCS) and platelet-derived growth factor (PDGF) induced arachidonate release in subconfluent murine 3T6 fibroblast cultures. However, the magnitude of this effect decreased significantly as the cell culture became confluent. Wound-induced injury of the fibroblast monolayer initiated a repair process that was potentiated by FCS or PDGF and which restored the integrity of cell monolayer. Under these experimental conditions, FCS and PDGF induced phospholipase A2 activity and subsequent arachidonic acid mobilization and production of eicosanoids in wounded fibroblast cultures. Finally, it is demonstrated that prostanoids, specifically prostaglandin E2, play an important role in cell proliferation induced by FCS and PDGF during wound repair.

    Notes: Abstract When we stimulated 3T6 fibroblasts, we observed a mobilization of arachidonic acid (AA) from phospholipids. The magnitude of this response decreased as the cells became confluent and the change coincided with a decrease in the percentage of cells in growth phases (G2 + M); this was not a consequence of the time in culture or a factor in the growth medium. Preconfluent fibroblasts incubated with the calcium ionophore A23187 (1 μM), 4β-phorbol-12-myristate acetate (PMA, 10 μM), bradykinin (10 μM) or fetal calf serum (20%) released 38.8%, 62.5%, 11.2% and 45.6% [3H]AA, respectively. Confluent cells stimulated under the same conditions released only 16.9%, 1.5%, 0.5% and 18.5% [3H]AA, respectively. This decreasing mobilization of AA was demonstrated using metabolic labelling and measurement of prostaglandin PGE2. The decrease was not due to a changing pool of AA. [3H]AA release from each phospholipid decreased with confluence. Conversion of confluent cells to the proliferative phenotype by mechanical wounding of the monolayer increased the release of [3H]AA. This effect is consistent with regulated, growth-dependent changes in the activity of phospholipase A2, a process regulated by changes in phosphorylation of the enzyme. The increased release of [3H]AA from preconfluent, actively dividing cells may have important physiological implications and may help elucidate mechanisms regulating release of AA.
