    Notes: Abstract  Segments taken from young leaves of an orchid (Oncidium Gower Ramsey) produced clusters of somatic embryos directly from epidermal and mesophyll cells of leaf tips and wound surfaces without an intervening callus within 1 month when cultured on a GelriteTM-gelled 1/2-MS basal medium supplemented with a low dosage (0.3–1 mg/l) of thidiazuron. Subculturing of these embryo clusters produced more embryos and subsequent plantlet formation on the same medium. The high-frequency embryogenesis of these leaf cells in this orchid is strong evidence of their totipotency, and further modification of the protocol for plant formation could be useful for the mass propagation and transformation of selected elite lines.

    Notes: Summary Waste paper was pretreated with 5M sulfuric acid at 28°C to give a gel-like solution which was then hydrolyzed with cellulase, purified fromTrichoderma viride, to form carbohydrates. The determination of the carbohydrates was accomplished using an aminopropylbonded silica HPLC column and an RI detector. The detection limits were 20 ppm; xylose, glucose, and cellobiose were the major components. Salts from the buffer solution may affect the activity of cellulase. The analysis of carbohydrates can be performed under acid conditions. HPLC analysis of carbohydrates from enzymatic hydrolysis is reliable and successful.

    Notes: Abstract The paramagnetic Meissner effect (PME), in which the field-cooled-magnetization (FCM) of superconducting samples is positive below the superconducting transition temperature Tc, has been observed in certain ceramic and single-crystal samples of the high-Tc cuprate superconductors and more recently in disk-shaped Nb samples. Through systematic investigations of the conditions for observing the PME in Nb disks, various surface treatments to the Nb disks were found to change both the zero-field-cooled-magnetization (ZFCM) and the FCM, including the appearance of a positive FCM in samples previously not exhibiting the PME as well as the elimination of the PME through surface abrasion. These results suggest that the PME arises from the field distributions created by the flux pinning associated with microstructural defects on the surface layer of the disk.
