Abstract The mechanisms by which glucocorticoids areeffective in acute liver rejection therapy are notentirely clear. The aims of this study were tocharacterize the intrahepatic immunological phenotype inacute liver rejection, as well as the effect ofglucocorticoids on cytokinestimulated hepatocyte celllines. Biopsy sections from these patients were studiedby immunohistochemistry. Cytokine-stimulated hepatocyte cell lines treated with glucocorticoids wereevaluated by flow cytometry. The intrahepatic expressionof both β2-microglobulin conformationalepitope and intercellular adhesion molecule-1 was higher in acute rejection than in resolving rejection.Interestingly, glucocorticoids were able to modulate invitro the cytokineinduced expression of these moleculeson hepatocyte cell lines. Beneficial effects of the glucocorticoid treatment appear to beassociated with a modulation of aβ2-microglobulin conformational epitopeand the intercellular adhesion molecule-1 onintrahepatic cellular targets in the acute rejection process.
