    Notes: Summary Herbicide-resistant lines of birdsfoot trefoil (Lotus corniculatus L. cv ‘Leo’) were isolated after sequential selection at the callus, shoot, and whole plant levels to the sulfonylurea (SU) herbicide Harmony {DPX-M6316; 3-[[[(4-methoxy-6methyl-1,3,5, triazine-2-yl) amino] carbonyl] amino] sulfonyl-2-thiophenecarboxylate}. In field and growth chamber tests the Harmony regenerant lines displayed an increased tolerance as compared to control plants from tissue culture and controls grown from seed. Results of evaluation of callus cultures of regenerated mutant lines signify stability of the resistance. Outcrossed seeds collected from field trials, and tested in vitro for herbicide resistance, indicate that the trait is heritable and that resistance may be due to reduced sensitivity of acetolactate synthase to SU inhibition. Genetically stable herbicide-resistant lines of birdsfoot trefoil were successfully isolated using in vitro selection.

    Notes: Summary The objectives of this study were to determine if biotin-labelled total genomic DNA of rye (Secale cereale L.) could be used to (i) preferentially label rye meiotic chromosomes in triticale and (ii) detect translocation stocks at interphase and/or early prophase by in situ hybridization. Welsh triticale, a wheat-rye segmental amphiploid, and Kavkaz wheat, a wheat-rye translocation were used. The results indicated that labelled chromosomes of rye and unlabelled chromosomes of wheat could be observed throughout all meiotic stages in the triticale. For Kavkaz wheat, the presence of the translocated 1RS chromosome arm of rye was detected at the interphase or very early prophase stage. Rapid assessment of feasibility of gene transfers and detection of alien DNA in somatic cells at the interphase stage by in situ hybridization allows for rapid decision-making and saves time and expense in plant breeding programs.

    Notes: tert-Butyl α,β-dioxobutyrate (hydrate; 1d) undergoes, at medium or high pH, the benzilic-acid rearrangement with exclusive 1,2-shift of the COO(t-Bu) group; the same is true for the corresponding isopropyl ester 1c and ethyl ester 1b at high pH, whereas at lower pH, the overall picture of these reactions is complicated by concurrent hydrolysis of the ester, followed by a 1,2-shift of the COO- group. Consequently, the shift of these electron-attracting groups cannot be considered to be systematically disfavoured (compared, e.g., with alkyl-group shifts). Kinetic measurements of the rearrangement show for both esters (as well as for the analogous ethyl ester 1b, and also for ethyl 3-cyclopropyl-α,β-dioxopropionate (4)) a characteristic rate profile: at relatively low pH, k is proportional to [HO-], approaching saturation with increasing [HO-] (interpreted as complete transformation of the substrate into the hydrate monoanion), which is followed at higher pH by another rate increase with k proportional to [HO-] (probably due to the reaction of the hydrate dianion). The similarity of k values for 1b-d shows that in the shift of COOR steric hindrance caused by R is negligible.

    Notes: In alkaline medium C3H5—CO—C (OH)2—COOEt (1b) is transformed into C3H5—C (OH)(COOH)2 (2a). Labelling experiments show that the cyclopropyl group is not shifted, but only ROOC and/or -O2C groups. GC./MS. and NMR. analysis after incomplete reaction show that both ROOC- and -O2C-groups Migrate; at higher pH (ca. 14) the ester group rearrangement seems to be more important than at pH ca.9-10.

    Notes: Molecular motions in mixtures of the side chain polymer - poly(vinyl acetate) and dibuthyl phthalate were studied as a functionof polymer concentration and temperature using the technique of paramagnetic resonance (EPR). When the small spherical probe tempol (TPL) was used, we were able to approximate the observed EPR spectrum with a simulation using a single rotational correlation time τ. The peviously developed Grest-Cohen all-temperature model matched the Arrhenius polts. The EPR spectra from a cigar-shaped cholestane (COL) probe could not be adequately matched by single τ simulation when the polymer was at temperatures somewhat above the glass to rubber transition temperature (Tg). Points corresponding to these temperatures were left of the Arrhenius plot and a discontinuity was observed where the gap in the data occurred. As the concentration of plasticizer was increased, we found that the discontinuity became less steep, but the τ at which the gap occurs was always ≈ 10-8. The spectra observed at the temperature region of the gap were approximately 50-50 composites of experimental spectra observed at ± K. In both the TPL and COL cases, there was evidence of the existence of multiple correlation times. Preliminary studies of other polymers, both with and without side chains, also indicated the existence of the gap when COL is used as the probe. © 1993 John Wiley & Sons, Inc.
