    Notes: Abstract Sympathetic and parasympathetic activity was evaluated in ten healthy controls, nine asymptomatic, untreated asthmatic subjects and ten asthmatic patients during treatment for acute asthma, by measurement of the variation in resting heart rate using frequency spectrum analysis. Heart rate was recorded by ECG and respiratory rate by impedance plethysmography. Spectral density of the beat-to-beat heart rate was measured within the low frequency band 0.04 to 0.10 Hz (low frequency power) modulated by sympathetic and parasympathetic activity, and within a 0.12 Hz band width at the respiratory frequency mode (respiratory frequency power) modulated by parasympathetic activity. Acute asthmatics had higher heart rates than either of the other two groups; this was probably related to the effects of beta-adrenoceptor agonist medication. Sympathetically mediated heart rate variability (normalized low frequency power) was significantly lower in both asymptomatic (p 〈 0.002) and acute (p 〈 0.02) asthma subjects compared to controls. This is consistent with altered sympathetic/parasympathetic regulation of heart rate in subjects with bronchial asthma.

    Notes: Abstract Sympathetic and parasympathetic activity was evaluated on 39 occasions in 17 patients with the sepsis syndrome, by measurement of the variation in resting heart rate using frequency spectrum analysis. Heart rate was recorded by electrocardiography and respiratory rate by impedance plethysmography. The sepsis syndrome was established on the basis of established clinical and physiological criteria. Subjects were studied, whenever possible, during the period of sepsis and during recovery. Spectral density of the beat-to-beat heart rate was measured within the low frequency band 0.04 to 0.10 Hz (low frequency power, LFP) modulated by sympathetic and parasympathetic activity, and within a 0.12 Hz band width at the respiratory frequency mode (respiratory frequency power, RFP) modulated by parasympathetic activity. Results were expressed as the total variability (total area beneath the power spectrum), as the spectral components normalized to the total power (LFPn, RFPn) or as the ratio of LFP/RFP. During the sepsis syndrome, total heart rate variability and the sympathetically mediated component, LFPn were significantly lower than during the following recovery phase (ANOVA,p 〈 0.0001,p 〈 0.01 respectively). Both APACHE II (Acute Physiological and Chronic Health Evaluation) and TISS (Therapeutic Intervention Scoring System) scores showed an inverse correlation with total heart rate variability, logLFP, LFPn and the LFP/RFP ratio (p 〈 0.002 to 0.0001). Sympathetically mediated heart rate variability was significantly lower during the sepsis syndrome and was inversely proportional to disease severity.

    Notes: The addition of a lytic layer to a preformed linear sucrose gradient induces a temporary (up to half a day) initial gradient of considerable steepness which retards the sedimentation of large (〉T4) DNAs. Centrifugation at sufficiently slow angular velocities permits the temporary initial gradient to disappear and therefore the sedimentation distance increases, yielding a rotor speed dependent effect on sedimentation distance. Gradients which are free of this effect are described and shown to permit mouse leukemia cell DNA to sediment independently of rotor speed (5-30 krpm).

    Notes: In 1974, Zimm described a theory which predicts that the sedimentation coefficient of high-molecular-weight DNA will decrease as the rotor speed of measurement increases. In 1979, this theory was revised, and the new formula predicts speed-dependence effects that are substantially smaller than the predictions of the original version. This report describes the results of subjecting both the original and the revised versions of the theory to quantitative tests using a well-defined sucrose-gradient system and a DNA of known molecular weight (T4c DNA). T4c bacteriophage is a mutant, whose DNA contains the unmodified base cytosine, instead of the glucosylated hydroxymethylcytosine characteristic of the T-even bacteriophages, and has a molecular weight of 115 ± 3 × 106. The DNA of the wild-type phage (T4D+) was also used in some experiments.In addition to the quantitative tests, the experiments test for an effect first observed by Rubenstein and Leighton, which showed that the sedimentation coefficient measured for T2 DNA depended on the composition of the centrifuge tube used for the measurement (tube composition effect). It can be inferred from this observation that an interaction occurs between particle and tube wall during sedimentation, and this leads to a reduction in sedimentation velocity independent of the reduction in S described by Zimm's theory.The results show that in the range of 25,000-50,000 rpm, the original but theoretically incorrect form of the theory quite accurately describes the sedimentation behavior of both T4c and T4D+ DNA, although T4D+ was a special case in some respects. The revised (corrected) form of the theory predicts much less of a speed-dependence effect than that actually observed. The discrepancy between corrected theory and observation suggests that other factors (perhaps arising from the use of the swinging bucket rotor geometry) are causing the additional observed reduction in S20,w. However, the experiments show that the tube composition effect does not seem to be one of these.

    Notes: Raman spectroscopic studies have been carried out on polymers of L-valine ranging in degree of polymerization (DP) from 2 to 930. The spectrum of the hexapeptide (DP = 6) is closely similar over the entire range 40-1750 cm-1 to those of polymers with much higher DP, and the structure is clearly shown to be that of the antiparallel pleated sheet (β-structure) by the amide I and III frequencies. The formation of a little α-helical structure occurs in polymers with DP above 500, although the amount does not appear to be a linear function of DP. The α-helical structure is unstable and readily destroyed in samples cast from trifluoroacetic acid solution. It is stabilized by the incorporation of L-alanine, a strong helix-former; polymers of the latter may in turn be forced into a α-structure in copolymers sufficiently rich in L-valine.

    Notes: The model presented here describes the DNA viscosity and viscoelastometric responses to ionizing radiation-induced molecular breakdown. Four DNA conformations are considered: (1) linear chain, (2) relaxed circle, (3) supercoiled circle, and (4) supercoiled linear. The model includes the systems of differential equations for calculating the size distributions of DNA molecules as functions of dose. Thedistributions are used in the usual hydrodynamic formulae for heterogeneouspopulations of polymer molecules. The sensitivity of viscoelastometry for the estimation of D37 is better than that of viscometry. The model predicts a considerable difference between the radiation responsesof linear and circular DNAs. The prediction is confirmed by experimental data for T4 DNA [C. S. Lange et al. (1984) Rad. Res. 100, 1-15] and E. coli DNA [S. E. Bresler et al. (1984) Biophys. J. 45, 749-754]. Thus, ionizing radiation can be used to determine the original conformation of DNA molecules.

    Notes: Crystals of the monomer and polymer of the bis-ethylurethane of 5,7-dodecadiyne-l, 12-diol (1 a, ETCD), systematic name: 5,7-dodecadiynylene bis(N-ethylcarbamate) have been studied by X-ray powder and single-crystal photographic techniques. Lattice constants at 20°C were obtained from both single-crystal and powder data and are in satisfactory agreement with a previous report. Lattice constants at 130-135°C were obtained from powder data and reveal that the lattice expands in volume by 4% on heating from 20°C, in contrast to a previous report which indicated a volume contraction on heating. Rotation photographs of poly(ETCD) crystals before and after extraction of unpolymerized monomer with boiling methanol reveal that solvent extraction renders many reflections more diffuse. Weissenberg h0l-photographs of the polymer reveal sufficient disorder so as to preclude a high-precision structural determination. Rotation and Weissenberg photographs of monomeric ETCD at 20°C manifest the major reflections of the polymer.

    Notes: ESCA and contact angle measurements were used to characterize the surfaces of Polyethylene and polypropylene films exposed to SF6, CF4, and C2F6 plasmas. None of these gases polymerized in the plasma. However, all plasma treatments grafted fluorinated functionalities directly to the polymer surfaces. SF6 plasmas graft fluorine atoms to a polyolefin surface. CF4 plasmas also react by a mechanism dominated by fluorine atoms, but with some contribution from CFx-radical reactions. Although C2F6 does not polymerize, the mechanism of grafting is still dominated by the reactions of CFx radicals. For all gases studied, the lack of polymerization is attributed to competitive ablation and polymerization reactions occurring under conditions of ion bombardment.

SF6 plasmas cause loss of aromaticity in the polystyrene surface region via saturation of the phenyl ring and/or carbon-bond breakage and subsequent fluorination. C2F6 plasmas graft CFx radicals directly to the polystyrene surface without necessarily destroying the aromaticity of the polymer. CF4 plasmas appear to be intermediate in character between SF6 and C2F6 plasmas.

Affinities and mechanisms for PO-PRO-3 binding to superhelical and linearized pUC 18 plasmid DNA were examined through measurement of binding isotherms, continuous-variation analysis, and DNA titrations. In addition, the effects of DNA conformation, protein and RNA contaminants, solvent polarity, and ionic strength are examined with the aim of optimizing binding and elution conditions and of assisgning limits to the range of applicability of the affinity purification. © 1995 John Wiley & Sons, Inc.
