    Effect of endothelium and carbachol on α-adrenoceptor agonist stimulated uptake and efflux of 45Ca in rat isolated aorta (1987)

    Notes: Summary Preincubation with carbachol (10 μM) did not affect basal 45Ca accumulation by rat isolated aortic segments complete with endothelium, although 45Ca accumulation was enhanced by removal of endothelium. This confirms the observation that in the presence of endothelium Ca2+ influx in rat aorta is antagonized, and indicates that the basal release of an endothelial derived factor might be sufficient to maximally antagonize basal Ca2+ influx, or alternatively that EDRF released as a result of muscarinic stimulation does not have identical effects to the factor released under basal conditions. Accumulation of 45Ca stimulated by B-HT 920 but not that stimulated by phenylephrine was antagonized in the presence of endothelium. Contractions elicited by B-HT 920 were abolished in the presence of endothelium while contractions evoked by phenylephrine were reduced by about 50%. Preincubation with 10 μM carbachol antagonized both phenylephrine (1 μM) stimulated 45Ca accumulation and contractile responses in the presence of endothelium to about the same extent. Therefore, it might be concluded that the inhibitory effect of EDRF in this tissue is due to an inhibition of stimulated Ca2+ influx. However, while addition of carbachol to tissues precontracted with phenylephrine elicited an immediate relaxation in the presence of endothelium, this relaxation could not be correlated with a reduction in tissue accumulation of 45Ca. Carbachol also antagonized the phenylephrine-induced reduction of tissue 45Ca content (i.e. efflux of Ca2+), in tissues preloaded with 45Ca. This implies that the initial endothelial-mediated relaxant effect of carbachol in precontracted tissues cannot be explained either by reduced influx or by an enhanced efflux of Ca2+ from the smooth muscle cells. Therefore, it is concluded that although EDRF mediates a reduction in agonist-induced Cat2+ uptake it also exerts other effects, one of which may be an initial redistribution of intracelllar Ca2+, resulting in a reduction in free intracellular Ca2+ levels and therefore in relaxation.

    Notes: Cellulomonas fimi produces an endoglucanase and an exoglucanase which bind strongly to cellulose. Each enzyme contains three distinct regions: a short sequence of about 20 amino acids containing only proline and threonine (the ProThr box); an irregular region, rich in hydroxyamino acids, of low charge density, and which is predicted to have little secondary structure; and an ordered region of higher charge density which contains a potential active site, and which is predicted to have secondary structure; and an ordered region of higher charge density which contains a potential active site, and which is predicted to have secondary structure. The Pro-Thr box is conserved almost perfectly in the two enzymes. The irregular regions are 50% conserved, and the conserved sequences include four Asn-Xaa-Ser/Thr sites. The ordered regions appear not to be conserved, but the potential active sites both have the sequence Glu-Xaa7-Asn-Xaa6-Thr; they occur at widely separated sites in the two regions. The order of the regions is reversed in the two enzymes: irregular-Pro-Thr box-ordered in the endoglucanase; ordered-Pro-Thr box irregular in the exoglucanase. The genes for the two enzymes appear to have arisen by shuffling of two conserved sequences and either one or two other sequences.

    Notes: Liquid-vapor phase equilibria measurements were made at 112.00 K on the binary systems nitrogen-argon, nitrogen-methane, and argon-methane and the ternary system nitrogen-argon-methane. Values of gE, the excess Gibbs free energy, have been calculated from the experimental data for all the systems studied. The data and derived gE values for the binary systems were compared with the results of previous investigations with satisfactory agreement.

    Notes: Dielectric constants and Clausius-Mossotti functions (CM) have been determined for selected pure components, binary mixtures, and ternary mixtures at temperatures between 91 and 115 K and pressures near saturation. Excess Clausius-Mossotti functions (CME) have been calculated from the pure fluid and mixture values.Mixture CME values are found to be generally less than 0.1% of the mixture CM values for systems containing only nonpolar species. Thus, dielectric constant measurements can be conveniently used as substitute density measurements for simple liquid mixtures. Methods have been developed to utilize dielectric constant measurements to accurately determine densities and heating values per unit liquid volume for liquefied natural gas mixtures.
