    Notes: Abstract Agrobacterium tumefaciens strain 1D1609 is reported here as the first field isolate from alfalfa (Medicago sativa L.). Unlike well-characterized A. tumefaciens strains such as C58 and Ach5, strain 1D1609 is highly virulent on alfalfa and has a distinctive host range. Interestingly, strain 1D1609 is naturally resistant to kanamycin and spectinomycin. The Ti plasmid in strain 1D1609 is an octopine-type; thus, tumors formed by strain 1D1609 synthesize octopine, which is utilized by the bacterium as a sole carbon source. Reciprocal exchange of Ti plasmids between strains 1D1609 and C58 showed that both chromosomal and Ti plasmid genes in strain 1D1609 contribute specifically to tumor formation on alfalfa. In addition, the nondormant CUF101 alfalfa cultivar from which strain 1D1609 was isolated was significantly more susceptible to all Agrobacterium strains tested than was the dormant Agate cultivar.

    Notes: A rapid method for determining the reactivity of murine monoclonal antibodies with apolipoprotein A-I (Apo A-I) isoforms has been developed. The method consists of a rapid (1-2 h) prescreening of fusion well culture fluids by using an automated immunofluorescent assay (IF A) to identify fusion well cultures producing monoclonal antibody against Apo A-I and high-density lipoprotein (HDL). Immediately following the IFA procedure, immunodetection of the reactivity pattern of selected fusion well culture fluids is determined by using transblotted isoforms of Apo A-I previously separated by isoelectric focusing. Strips are prepared and stored at 4 °C in protein containing blocking solution before use. Preselected culture fluids could be tested within 6 h for qualitative reactivity with Apo A-I isoforms. Using this procedure we selected several monoclonal antibody-producing cultures for further studies on HDL metabolism based on their differing reactivities with Apo A-I isoforms. This technique should be easily transferable to other laboratories for assessing proteins having multiple isoforms since the crucial steps of separation and blotting can be optimized and performed before actual testing for antibody reactivity.

    Notes: Two methods have been investigated for the measurement of dissolved oxygen in fermentations: the tubing method and the probe method. The sensing elements for both methods are steam-sterilizable and measure only the oxygen tension in the liquid phase. The tubing method is probably the most accurate and reliable method available for measurement of dissolved oxygen in fermentations but it requires extensive ancillary apparatus.

    Notes: Symptoms of oxygen deficiency and variations in dissolved-oxygen level have been described in some typical aerobic fermentations. In bacterial fermentations, dissolved-oxygen levels were almost the same in different parts of the fermentor. As a result, good correlations between apparent and true critical oxygen concentrations were obtained. Poor correlations were obtained in mould fermentations.
