    Notes: Rat pituitary homogenates were subjected to two phase countercurrent partition in a poly(ethylene glycol)-dextran mixture using a simple apparatus with enhanced gravity to facilitate the phase separations. Assay of the fractions for organelle marker enzymes and prolactin after 17 transfers showed similar distributions for endoplasmic reticulum, lysosomes, prolactin granules and plasma membrane at the lowest dextran concentrations. Increasing the dextran concentrations had a differential effect on the various organelles. Excellent resolution of endoplasmic reticulum from the other organelles was obtained and marked organelle heterogeneity was demonstrated. Two-phase countercurrent partition thus offers an alternative approach to the subcellular fractionation of pituitary homogenates and should prove useful in separating endoplasmic reticulum from plasma membrane and other cell components.

    Notes: A rapid, reliable, sensitive and reproducible HPLC method was developed for the assay of ferrochelatase activity in rat liver. The assay was carried out aerobically with Zn2+ and mesoporphyrin or protoporphyrin IX as substrates. Zn-porphyrins formed were extracted with dimethyl sulphoxide/methanol (30:70, v/v) containing Zn-deuteroporphyrin as the internal standard for separation and quantification by reversed-phase chromatography. The Km for mesoporphyrin was 5.9 μM, for protoporphyrin IX 8.8 μM and for zinc 6.0 μM. The specific activities were 33.1 ± 5.0 nmol Zn-mesoporphyrin or 13.4 ± 2.0 nmol Zn-protoporphyrin formed per hour per mg of protein for mitochondria and 12.3 ± 2.2 nmol Zn-mesoporphyrin or 4.6 ± 0.9 nmol Zn-protoporphyrin per hour per mg of protein for liver homogenate.

    Notes: The distribution of intestinal transglutaminase was investigated by immunofluorescence microscopy using rabbit anti-guinea pig transglutaminase immunoglobulin. Transglutaminase-related antigen was demonstrated principally in the cytoplasm of villous core interstitial cells with some activity in the brush border region of the villous epithelial cells. Implications for the pathogenesis of coeliac disease are discussed.
