    Notes: The high resolution mass spectra (500 eV) of some α,β-unsaturated steroidal ketones have been studied and compared with the spectra of the corresponding α-chloromercuri ketones. In the latter, the carbon-mercury bond frequently remains intact at the expense of the fission of two carbon-carbon bonds. The abundance of mercury-containing ions allows the use of the mercury atom fingerprint in confirming ring B fragmentation of the steroid nucleus at C(6)-C(7) and C(9)-C(10) for 5α-androst-1-ene-3,17-dione, 1,4-androstadiene-3,17-dione and their 2-chloromercuri derivatives; and at C(7)-C(8) and C(9)-C(10) for 1,4,6-androstatriene-3,17-dione, 1,4,6-androstarien-17 β-ol-3-one and their 2-chloromercuri derivatives. 2-Chloromercuri-1,4,6-androstatriene-3,17-dione and 2-chloromercuri-1,4,6-androstatrien-17 β-ol-3-one also give an abundant ion as the result of ring C fragmentation at C(8)-C(14) and C(11)-C(12), the chloromercuri group being replaced by a hydrogen atom. This ring C cleavage gives the only recognizable distinctive fragmentation ion for 1,4,6-pregnatriene-3,20-dione and 2-chloromercuri-1,4,6-pregnatriene-3,20-dione. For most of the mercurated steroids, the low resolution mass spectra (70 eV) are reported. In these spectra, the fragmentation patterns are similar to those obtained using the higher ionization energy employed for the high resolution spectra.

    Notes: Analysis of small amounts of glycoproteins by high performance liquid chromatography (HPLC) and liquid secondary ion mass spectrometry (LSIMS) together with enzyme digestion has been investigated using fetuin as a model. Preliminary data indicates that 71% of the expected peptides were detected by LSIMS analysis of 200 pmol total digest. HPLC profiles of peptides and glycopeptides were obtained from 2 nmol of digest using a reversed phase (C18) column eluted in a solvent system containing TFA, water and acetonitrile. This has provided glycopeptides for subsequent oligosaccharide analysis. Strategies are reviewed for the chromatographic characterization of oligosaccharides following their release from glycopeptides by chemical and enzymatic procedures.

    Notes: In certain pathophysiological conditions, such as rheumatoid arthritis, there are alterations in the glycosylation pattern of the acute phase protein, alpha-1-acid glycoprotein (AGP). These changes are likely to be functionally significant, however, verification of the latter role requires a system which reflectsin vivo glycosylation changes in AGP and also produces sufficient quantities of the protein for further study. The human hepatoma cell line HepG2 is documented as displaying a shift in the glycosylation pattern of glycoproteins from normal state to acute phase after stimulation with inflammatory mediators. We have isolated AGP from the culture medium of HepG2 cells both before and after stimulation with a cytokine preparation and analysed the glycosylation pattern of each preparation, after enzymatic release, by high pH anion-exchange chromatography. Before stimulation, the glycosylated population was similar to a profile which was of AGP isolated from normal plasma; however, cytokine stimulation resulted in a shift to a profile which was consistent with that of AGP from a rheumatoid arthritis sufferer. Thus a HepG2 cell culture system is capable of being a crude model of the changes in glycosylation of acute phase proteins although it has a tendency to produce oligosaccharide chains which are not fully sialylated.

    Notes: An isocratic reversed-phase high performance liquid chromatographic method for the determination of tamoxifen and its metabolites in plasma and tissues is described. Plasma or tissue homogenate was extracted with methanol/dimethyl sulphoxide (4:1 v/v). The supernatant after centrifugation was separated on a BDS-Hypersil column with methanol/0.5 M ammonium acetate (75:25 v/v) as the mobile phase. The recoveries of tamoxifen added to plasma and liver tissue homogenate by the extraction procedure were 102± 1.6 and 98±2.4% (mean±SD, n = 6), respectively. The solutes were detected at 280 nm with a detection limit of 0.25 μg/mL for tamoxifen.

    Notes: Over the past 15 years the linear learning machine has been applied to a large number of chemical problems. The learning machine approach is conceptually simple and does not require knowledge about the statistical distribution of the data. However, there are problems associated with this approach. One problem which has not been investigated is the influence of mislabeled samples on the positioning of the hyperplane in feature space. If a few samples in a data set are incorrectly tagged prior to training (i.e. the samples are labeled as members of class 2 even though they are actually members of class 1), it is still possible using the linear learning machine to achieve a classification success rate of 100% for the training set. However, unfavorable results will be obtained for the prediction set. The magnitude of this effect and its potential implications regarding the proper use of the linear learning machine are discussed.

    Notes: The object of this paper is to develop a suitable statistical procedure to evaluate clean-up standards at hazardous waste sites. Under the assumptions that contaminant masses at a site follow a gamma distribution and that the data from the pre-remediation baseline sample as well as from the interim or final sample taken after a certain period of operation are both distributed as gamma with the same shape parameter but different scale parameters, we derive a uniformly most powerful unbiased test of the hypothesis that a specified percentage of contaminant mass has been reduced. A large-sample approximation of the exact test procedure and a comparison with the likelihood ratio test are provided.

    Notes: The determination of serum cortisol by thermospray liquid chromatography/mass spectrometry (LC/MS) has been assessed. The method incorporates stable isotope dilution and innumoadsorption extraction. [M + H]+ ions are monitored during LC/MS. The within-assay reproducibility is satisfactory (coefficient of variation 7% at a concentration of 190 ng ml-1), though inferior to that achieved with the (more lengthy) procedure of gas chromatography/mass spectrometry (GC/MS). Satisfactory agreement between LC/MS (y) and GC/MS (x) data was observed (y = 0.934x + 12.4 ng ml-1; r = 0.968; n = 14). It is concluded that the generation of precise reference data for the assessment of routine cortisol assays is at present better achieved by GC/MS. LC/MS, however, provides satisfactory quantitative data via a more simple and rapid method.

    Notes: Thermospray spectra on a magnetic sector mass spectrometer, by definition measured at high accelerating voltages, are seen to be highly fragmented. This is rationalised in terms of the necessity to accelerate ions through a region of high pressure in the vicinity of the ion-collecting orifice at the spray volume, where ions may be energized by ion molecule collisions. In this region, a high percentage of the parent ion current is lost through the dispersion of the ion beam and by the collisional activation of molecular ions resulting in extensive fragmentation. The investigation of means by which these effects may be minimized has led to the construction of an improved sector thermospray source. Comparative results for gramicidin S demonstrate that its fragmentation is reduced and that the intensity of the protonated molecular ion is greatly enhanced.

    Notes: A series of 17 cysteine, N-acetyl cysteine, glutathione, and N-trifluoroacetyl glutathione conjugates have been prepared, and their thermospray (TSP) spectra have been recorded in the positive and negative ion modes. The compounds undergo extensive fragmentation, which primarily occurs at the carbon-sulfur bonds. For most of the compounds, positive ion TSP is more sensitive than negative ion thermospray. Probably due to the thermal lability of these adducts, the quality of the spectra obtained are dependent on source conditions, requiring fine control of the vaporization/desolvation process.
