    Notes: A high performance liquid chromatographic (HPLC) system is described for determination of the unsaturated disaccharide (ΔDi-HA) derived from hyaluronic acid (HA) in human urine by digestion with hyaluronidase SD. The effects of eluents on the separation of ΔDi-HA and ΔDi-OS, which is derived from the reaction of chondroitin with the enzyme, have been studied. The established chromatographic conditions were as follows-column: a stainless steel tube (4 mm i.d. × 250 mm) packed with TSKgel NH2-60; eluent: a mixture of acetonitrile and 0.1 M Tris-HCI buffer containing 0.1 M boric acid and 10 mM sodium sulphate, pH 7.0 (64:36, v/v). The strong fluorescence of unsaturated disaccharide after the reaction with 2-cyanoacetamide in alkaline medium was used for post-column detection. The calibration curve for ΔDi-HA was linear in the range 5 pmol-5 nmol with a practical detection limit of 2 pmol. The assay coefficients of variation (n = 5) at 200 pmol for ΔDi-HA and ΔDi-OS were 1.7 and 1.5%, respectively. This HPLC system has been applied to the determination of HA in human urine.

    Notes: The reactivities between saccharides and primary amino compounds were studied by liquid chromatography on an amino-bonded column and by measurement of the reaction rates of aromatic amines with saccharides. The recoveries from the column and the reaction rates were correlated with their physicochemical properties calculated by the CACheTM program. The reactivities between amines and saccharides correlated well with their hydrogen bonding energies calculated by molecular mechanics.

    Notes: A sensitive liquid chromatographic analysis for myo-inositol was developed using glycocyamine as the postlabelling reagent. The sensitivty was 500 pmol/injection. The system was applied to analyse myo-inositol in sera from eight patients with chronic renal failure. The average concentration of serum myo-inositol was 498.6 ± 257.0 μmol/L before haemodialysis, and 244.0 ± 131.1 μmol/L after haemodialysis. These results indicated that the kidney is the main site of myo-inositol metabolism.

    Notes: Samples of pregnant urine were treated with three diatomaceous earth columns to fractionate organic solvent-soluble components. The first column was pre-injected with the urine sample. The second and the third columns were pre-injected with sodium bicarbonate and sodium hydroxide solutions, respectively; these played an important role in capturing acidic components in the urine extract from the first column. Neutral components could be eluted out from the columns with the organic solvent, while the basic components were retained in the first column. Once strongly and weakly acidic components were captured on the alkaline columns, they were separately treated with an acidic mobile phase and recovered. The digoxin-like activities of each fraction were examined for cross-reactivity to antidigoxin antibody and inhibitory effects on dog kidney Na+, K+-ATPase. Both types of reactivity were found in all three fractions and decreased in the following order: strongly acidic, neutral and weakly acidic. The neutral fraction of pregnant urine showed significantly greater cross-reactivity than that of the non-pregnant urine. Following hydrolysis of the pregnant urine with β-D-glucuronidase, the cross-reactivity of the neutral fraction increased significantly. High performance liquid chromatographic analysis showed that several components in the neutral fraction had apparently increased after the hydrolysis, showing the possible presence of unknown digoxin-like components in the pregnant urine.

    Notes: Field desorption mass spectrometry was applied to a series of underivatized anthracycline, cosmomycins, to obtain fragment ions which were mass analysed using the linked scan technique without using collision activated dissociation. The daughter spectra of the various protonated compounds contain sugar sequence information which is in the mass spectra. The mass spectrometric data make it clear that field desorption tandem mass spectrometry can become a valuable additional technique for the structural analysis of anthracyclines.

    Notes: The retention time of 24 alkanes (Al-Al), 11 olefins (OI), 17 alkyl alcohols (Al-OH), 30 fatty acid methyl esters (FA) and 8 polyaromatic hydrocarbons (PAH) was measured on non-polar CP-Sil-5 and DB-1 and polar CP-WAX-52 and DB-WAX capillary columns at different temperatures.The correlation coefficients of the capacity ratios of each group of compounds and that of all the compounds together was excellent; however, there was some selectivity of different types of compounds when their capacity ratios were correlated with their Van der Waals volumes. The elution order of these compounds was Al-Al 〈 OI 〈 FA 〈 Al-OH 〈 PAH on both non-polar columns, while the retention of PAH was greater on the DB-1 column and those of other compounds were greater on the CP-Sil-5-column. Such selectivity was significant on polar columns.Further, the energy effect, which was used to make a retention index system in liquid chromatography, was obtained from their capacity ratios to make a retention index system for capillary column gas chromatography. The values of the energy effect of each group of compounds were quite constant compared to the values obtained in liquid chromatography. The selectivity of these capillary columns was further discussed from their capacity ratios, their Kováts retention indices, and their energy effect to make a prediction of retention time.

    Notes: The retention times of 24 alkanes, 11 olefins, 17 alkyl alcohols, 30 fatty acid methyl esters, and 8 polyaromatic hydrocarbons were measured on two non-polar and two polar capillary columns at different temperatures. The enthalpy of these compounds was calculated from the Van't Hoff plot.The log k′s and enthalpies of each group of compounds showed individual linear relations. The relation of the enthalpy and Van der Waals volume was similar to that of log k′ and Van der Waals volume, and differed from that of their log k′ and boiling point. Enthalpy thus indicates selectivity of columns.

    Notes: A simple HPLC method for determination of chondroitin sulphates as thier unsaturated disaccharides by chondroitinases has been applied to measure the glycosaminoglycans (GAGs) in human whole blood, plasma and blood cells. This method may be useful for the investigations on the role of blood GAGs and the metabolic fates of the exogenous GAG.
