Notes: Abstract A method for the detection of precipitating antibodies to aqueous extracts of flour in coeliac disease has been previously described by one of the authors (Rossipal). This study was done to elucidate which and how many fractions of gliadin can act as antigens in coeliac disease. A study of the aqueous extracts of flour by paper electrophoresis showed the presence of 3 gliadin fractions in all extracts. By disc electrophoresis (polyacrylamid gel, TRIS-glycine buffer pH 8.3, ionic strength 0.05), the presence of 12 fractions in wheat and barley extract, 12 fractions in extracts of oats and of 14 fractions in rye extract could be shown. Following the fractionation of the gliadin of the aqueous extracts in polyacrylamid gel, the gels were incubated with undiluted sera from infants with coeliac disease. Within 30–45 min precipitation lines had formed. It could be shown by this method that 3 gliadin fractions of wheat extract, and 2 fractions of barley oats, and rye extract act as antigens. These fractions were located close to the cathodic side of the gels.
