Notes: Abstract Eleven benzodiazepines were evaluated in the staircase test in mice. The behavioural parameters measured were the number of steps climbed and the number of rears during a 3-min test. Climbing and to a lesser extent rears were enhanced at low doses, whereas both parameters, particularly rearing, were reduced at higher doses. The differential effects of the drugs on the two parameters were used to determine indices of anxiolytic efficacy for each drug where increases in climbing were taken to indicate the onset of anxiolytic activity and decreases in rearing the onset of sedative activity. The compounds could be ranked according to these indices in a manner which appears to reflect their therapeutic profile in man.

Notes: Abstract The “staircase” is a simple and rapid test and was used to study two components of exploratory behaviour in the rat. The scores of rearing and the number of steps climbed during three minutes were recorded. Various psychotropic drugs were tested, which modified these two parameters. Neuroleptic induced a parallel decrease of both, while benzodiazepines, meprobamate, amobarbitone and ethanol decreased the rearing at doses which left the steps climbed unchanged. At high doses, there was a parallel decrease of both parameters. Amphetamine, at lower doses, increased the rearing score alone. The comparison of the studied psychotropic drug effects with those of two muscle relaxants (by a comparison of the slopes of regression lines) suggests that either the observed benzodiazepine effects were only partly due to their myorelaxant action, or, that both myorelaxants have some anxiolytic action. The effect of amphetamine at low doses can be viewed as a demonstration of increased anxiety.

Notes: Abstract  A 109-point linkage map consisting of three phenotypic loci (P 1, Y 2, and Rs), six restriction fragment length polymorphisms (RFLPs), two random amplified polymorphic DNAs (RAPDs), 96 amplified fragment length polymorphisms (AFLPs), and two selective amplification of microsatellite polymorphic loci (SAMPL) was constructed for carrot (Daucus carota L. ssp. sativus; 2n=2x=18). The incidence of polymorphism was 36% for RFLP probes, 20% for RAPD primers, and 42% for AFLP primers. The overall incidence of disturbed segregation was 18%. Linkage relationships at a LOD score of 4.0 and θ=0.25 indicated 11 linkage groups. The total map length was 534.4 cM and the map was clearly unsaturated with markers spaced at 4.9 cM. AFLP P6B15 was 1.7 cM from P 1, AFLP P1B34 was 2.2 cM from Y 2, and AFLP P3B30XA was 8.1 cM from Rs.

Notes: Abstract  The incidence and inheritance of a chloroplast DNA (cpDNA) mutation/marker, BP10U, was studied in crosses among cultivated carrots (Daucus carota ssp. sativus). BP10U is about 400 bp larger than the more common BP10L allele. The occurrence of BP10U among carrot inbreds was widespread. Individual plants exhibited only one form of BP10, and cpDNA inheritance was strictly maternal. BP10U only occurred in male-fertile plants. Some male-fertile inbreds and all cytoplasmically male-sterile (petaloid) carrots had the BP10L allele. Alloplasmic cpDNA variation has been reported previously in Daucus, but this is the first report of variation and inheritance of cpDNA within cultivated carrot.

Notes: Summary After a brief review of several methods described in the literature this paper discusses air-sampling on 2,4-dinitrophenylhydrazine-coated silica gel and identification requirements for aldehydes and ketones most commonly found in industrial pollution. Quantitative analysis of formaldehyde and acetaldehyde either by GC or by HPLC techniques is established using a dynamic system producing test atmospheres and compared with the usual colorimetric determinations.

Notes: Summary A useful method of sampling and measurement of toluene diisocyanate concentration in atmosphere is described. The sampler consists of glass-fibre filters impregnated with the reagent 1-(2-methoxyphenyl)piperazine1 so that the 2,4 and 2,6 isomers of TDI react to form urea derivatives which are analysed by high performance liquid chromatography in isocratic mode on cyan-amino and C18 bonded phases. A dynamic system was used to generate standard atmospheres of TDI and to validate the sampling method within the humidity range of 0% to 75%. A coated filter, a bubbling solution and an impregnated silica gel were compared as samplers requiring the piperazine reagent in performance experiments.
