    Notes: Abstract Different NAA plus kinetin or BA combinations were tested on Francia Pernod fennel seedlings for callus induction and plant regeneration. Callogenesis from hypocotyls was obtained in all auxin/cytokinin-containing media. The organogenic response was observed especially in presence of NAA plus kinetin. The highest frequency of shoot regeneration was found when the auxin and kinetin were used at a 1:1 ratio. Moreover, a prolonged culture period increased shoot formation. Somatic embryogenesis was tested on several fennel populations. The results gave evidence of the genotypic importance. Two different protocols were used for somatic embryo induction. Using the first protocol among the different fennel genotypes tested, only Francia Pernod showed embryogenic capacity. In this case, from a primary non-embryogenic callus cultured for 12 months in presence of 2,4-D, an embryogenic secondary callus was produced. When transferred to the medium without 2,4-D (agarized or liquid), this gave embryogenic plants in high frequency. As far as the second embryogenic method is concerned, secondary embryogenic callus developed only in the presence of 2,4-D plus kinetin in Francia Pernod genotype. Thereafter, the replacement of those growth regulators by GA3 into the medium greatly increased the somatic embryo development, especially in `Francia Pernod', but also in `Aboca erbe' callus, a population with a very poor embryogenic capacity. In Francia Pernod, the primary and secondary (embryogenic) calli showed different morphological and histological responses, either when the secondary callus was induced by 2,4-D alone or by 2,4-D plus kinetin. Ontogenetic processes leading to somatic embryo formation are described in this context.

    Notes: Abstract Callus cultures were initiated from micropropagated Artemisia absinthium plantlets on MS basal medium supplemented with different concentrations of BA, Kn, NAA, IAA and 2,4-d in combination or singly. Supplementing the medium with low doses of both BA in combination with NAA, and Kn in combination with NAA enhanced the growth rate of callus cultures. However, cultures grew slowly following the second subculture and the majority turned brown and died within the next month. Initiation of root and shoot primordia occured directly from leaf explants cultured on 1.81 μM 2,4-d, while adventitious shoot formation from callus was observed occasionally when BA was added to the medium in combination with IAA. Furthermore, medium containing 2.22 μM BA and 2.69 μM NAA stimulated both callus growth and organogenesis on some callus cultures derived from leaves and stems of young stock material. The best results were obtained with leaf explants. Cytological analysis of root meristems revealed that all regenerants were diploid (2n=18), as expected.

    Notes: Abstract A study has been carried out on several Amaranthus L species and breeding lines (A. caudatus, A. cruentus, A. hybridus, A. hypochondriacus), some of which have previously shown poor or no plant regeneration from callusing primary explants. Callus formation occurred from explants from almost all the lines tested. Significant differences in callus growth were observed among the lines depending on the growth regulator combinations used. Cultural conditions such as type and growth regulator combination, as well as age-dependent competence of the explant tissues, also influenced the frequency of shoot (and/or root) regeneration in Amaranthus species that had been shown difficult to regenerate in the past. The results obtained demonstrate that in Amaranthus the genotype, growth regulator dose and combination, the type and the physiological stage of explant are factors of great importance for in vitro plant regeneration.

    Description / Table of Contents: The detection of thin contaminated layers on contact surfaces.Today electrical contacts must satisfy requirements which are growing steadily. The detection of very thin contaminated layers at contact surfaces therefore becomes very important. Clad gold contacts were contaminated with very small amounts of Zink. Contact resistance measurements and surface analysis measurements by AES and SIMS were shown to be adequate for the detection of this surface contamination.
    Notes: Der Nachweis dünner Verunreinigungsschichten auf Kontaktoberflächen wird bei den steigenden Anforderungen an die Qualität elektrischer Kontakte immer wichtiger. Am Beispiel mit Zink verunreinigter Kontaktoberflächen wird gezeigt, daß durch die Kombination von Kontaktwiderstandsmessungen und der Analyse der Oberfläche durch moderne Methoden auch sehr dünne anorganische Verunreinigungsschichten erfaßt und identifiziert werden können. Die spezifischen Oberflächenanalysenmethoden AES und SIMS sind natürlich besser geeignet als die Elektronenstrahlmikroanalyse.

    Description / Table of Contents: Proportioning of noxious gases through hole type diaphragms - A method for testing the corrosion resistance of nobel metal materials for electrical engineeringCorrosion testing of nobel metal base materials for weak current engineering requires laboratory proportioning methods enabling the usually occurring noxious gases (such as hydrogen sulfide, sulfurdioxide or nitrogen oxides) to be added to a carrier gas stream in concentrations between appr. 10 ppb and 10 ppm. The authors present a method suitable for corrosion applications; this method is based on hole diaphragms used in the so called critical flow range. This method utilizes the phenomenon that - if the pressure difference across the diaphragm is sufficiently high - the gas flow rate in the narrowest cross section of the diaphragm hole will approach the velocity of sound, which, however, will not be exceeded even if the pressure difference is further increases. The diaphragms used were gold foils having hole diameters between 10 and 100 microns so that with a two-step dilution concentrations between 0.1 ppm and 〉 100 ppm could be obtained. This proportioning mecanism is of simple design and, because of the absence of moving or fragile parts, rather robust. The concentrations of the noxious gases may be held constant for a long time; the method prooves also successful when compared to other known metering methods. It appears therefore perfectly sutied to incorporation in the planned testing standards DIN 40 046, parts 36 and 37.
    Notes: Für die Korrosionsprüfung von Edelmetallwerkstoffen der Schwachstromtechnik im Labor werden Dosiermethoden benötigt, mit denen die häufigsten Schadgase, wie z. B. H2S, SO2 oder NOx in Konzentrationen zwischen etwa 10 ppb und 10 ppm in einen Trägergasstrom dosiert werden können. Es wird eine für Korrosionsanwendungen neue Dosiermethode vorgestellt. Diese Methode arbeitet mit Lochblenden in sog. Kritischen Bereich der Strömung. Dabei wird ausgenutzt, daß bei hinreichend großem Druckunterschied zwischen dem Bereich vor und hinter der Blende die Gasströmung im engsten Querschnitt der Blende Schallgeschwindigkeit erreicht und auch bei weiterer Vergrößerung der Druckdifferenz nicht überschreiten kann.Es wurden blenden aus Goldfolie mit Lochgrößen zwischen 10 und 100 μm benutzt. Damit ließen sich bei Verwendung einer zweistufigen Verdünnung Konzentrationen in Bereich zwischen 0,1 ppm und 〉 100 ppm erreichen. Die Dosierung ist im Aufbau einfach, und da sie keine beweglichen oder zerbrechlichen Teile enthält, auch sehr robust. Die Langzeitkonstanz der Schadgas-konzentrationen ist mit diesem Verfahren sehr gut. Im Vergleich zu bekannten Disierverfahren schneidet die Methode ebenfalls gut ab. Sie erscheint hervorragend für die Realistierung der angestrebten Testnormen DIN 40046, Teil 36 und Teil 37 geeignet zu sein.
