    Description / Table of Contents: Zusammenfassung Schallreizkorrelierte Gleichspannungsänderungen der primären Hörrinde (CGA=corticale Gleichspannungsantwort) wurden über subdurale implantierte Ag−AgCl Elektroden von der Katze abgeleitet und mit einem LINC-8-Rechner ausgewertet. Die CGA sind während der ganzen Reizdauer vorhanden, sie sind abhängig von der Reizqualität (z. B. Tonfrequenz), der Narkosetiefe und der Reizintensität. Die Intensitätsfunktion (Zusammenhang zwischen CGA-Amplitude und Reizintensität) wurde genauer untersucht und durch die Steigungskoeffizienten der Ausgleichsgeraden in Abhängigkeit von der Analysezeit (Zeitdifferenz zwischen Reizbeginn und Analysezeitpunkt) unter Berücksichtigung der Korrelation charakterisiert. Der Steigungskoeffizienn variiert mit der Analysezeit. Während er sich in der ersten Sekunde zwischen den Werten 0,7 und 0,4 bewegt, tendiert er mit wachsender Analysezeit gegen 0,3.
    Notes: Summary Sound evoked d-c changes in the primary auditory cortex (CGA= corticale Gleichspannungsantwort) of the cat were recorded with subdural implanted Ag-AgCl electrodes and analysed with a LINC-8-Computer. The CGA were maintained for the whole duration of stimulus, they depend on the stimulus quality (e.g. tone frequency), on the level of anesthesia and on the stimulus intensity. The intensity function (relation between CGA amplitude and stimulus intensity) has been investigated. It is characterised by the rising coefficient of the approximating line in dependence of the time of analysis (time of analysis starts with the stimulus beginning) with respect to correlation. The rising coefficientn varies with the time of analysis. Whereas in the first second it varies between the values 0.7 and 0.4 it tends with increasing time of analysis to 0.3.

    Description / Table of Contents: Zusammenfassung Zur Ermittlung der reizintensitätsabhängigen Aktivitätsänderung einzelner Neurone wurden die durch Reizung mit Tonbursts, Rauschbursts und Clicks ausgelösten Entladungen einzelner Elemente im colliculus inferior und im corpus geniculatum mediale mit Mikroelektroden extracellulär abgeleitet. Dabei wurde festgestellt, daß die Menge der sich im PST-Histogramm präsentierenden Entladungsmuster in wenige Klassen unterteilt werden kann, die jeweils durch einen Grundtyp charakterisiert sind. Es wurden: 1. Diese Klassen beschrieben und die Häufigkeitsverteilung von an 9 Katzen untersuchten 129 Einzelelementen hinsichtlich der Klassenzugehörigkeit angegeben. 2. Die Intensitätsfunktion der Einzelelemente untersucht und auf der Grundlage einer von Ranke (1952) aufgestellten Überlegung deren Verlauf diskutiert. 3. Ein einfaches Schema dargestellt, das den Zusammenhang zwischen den Intensitätsfunktionen der Einzelelemente und den von Elementkomplexen beschreiben soll.
    Notes: Summary To evaluate the activity variations of single neurons as a function of stimulus intensity the discharges of single elements were recorded extracellularly with microelectrodes from colliculus inferior and geniculate mediale. As stimuli served noise bursts, tone bursts and clicks. It is found that the number of response patterns in the PST-histogramm can be classified into a few groups everyone characterized by a groundtype. 1. These groups are described and the frequency distribution of 129 single elements of 9 cats is studied with regard to the classification. 2. The intensity function of single elements is studied on the basis of Ranke's adaptation theory and the curve is discussed. 3. A simple scheme is shown to describe the relation between the intensity functions of single elements and that of element complexes.

    Description / Table of Contents: Zusammenfassung Die in der Literatur für alle Teile der Hörbahn unterhalb des Geniculatum mediale beschriebenen reizkorrelierten Entladungsperiodizitäten einzelner Neurone wurden für den Colliculus inferior bestätigt. Darüber hinaus wird über eine ganz andersartige Entladungsperiodizität im Geniculatum mediale berichtet. Davon ausgehend werden Strukturmodelle für die neuronale zeitliche Koinzidenzmessung angegeben. Schließlich wird auf eine Unterscheidungsmöglichkeit von intramodalitätsspezifischen und intramodalitätsunspezifischen neuronalen Verarbeitungsmechanismen hingewiesen.
    Notes: Summary The stimulus correlated discharge periodicities of single neurons described in the literature for all parts of the acoustic channel below the geniculatum mediale were confirmed for the colliculus inferior. Furthermore a quite different kind of discharge periodicity in the geniculatum mediale is reported. With these results structure models are given for the neuronal temporal measurement of coincidence. Finally a possibility of discrimination between intra-modality-specific and intra-modality-nonspecific processing mechanism is shown.

    Notes: This paper describes a new contouring algorithm for isoparametric elements which can be used to map three-dimensional scalar fields. The contours are generated on arbitrary planes intersecting finite element structures. Tracing element contour lines is accomplished by an accurate predictor-corrector technique. A method of finding starting points for the algorithm on the boundary of the elements is also given.

    Notes: An analysis of the sequences of scyllatoxin and charybdotoxin suggested that it would be possible to design a core peptide sequence which would still fold to give the β-hairpin and helix seen in the toxins, but which would eliminate one disulfide and connecting residues. The core sequence was modeled, then synthesized and purified. The cysteines oxidize in air to give the same disulfide pairings as seen in the parent toxins as the major product. The three-dimensional structure of the core sequence peptide, termed Max, was determined using proton NMR spectroscopy and found to be identical in secondary structure to the toxins. However differences were found in the relative orientation of the β-hairpin and helix. The use of this structural motif, found in many insect toxins, as a disulfide framework for exploring sequence/structure/activity relationships is discussed. © 1994 John Wiley & Sons, Inc.

    Notes: Molecular dynamics simulations have been carried out on the complex formed between the tetraheme cytochrome c3 and the iron protein rubredoxin from the sulfate-reducing bacterium Desulfovibrio vulgaris. These simulations were performed both with explicit solvent water molecules included, and without solvent molecules using a distance-dependent dielectric constant to approximate the screening effects of solvent. The results of both simulations are strikingly different, indicating that the representation of environmental effects is important in such simulations. For example, a striking adaptation of the two proteins seen in the nonsolvated simulation is not seen when explicit solvent water is included; in fact, the complex appears to become weaker in the solvated simulation. Nonetheless, the iron-iron distance decreases more significantly in the solvated simulation than in the nonsolvated simulation. It was found that in both cases molecular dynamics optimized the structures further than energy minimization alone.

    Notes: The immunogenicity of a folded, all D-amino acid protein- rubredoxin, has been compared with that for the corresponding L-protein enantiomer. Following multiple administrations with alum adjuvant, the L-protein induced a strong, specific lgG antibody response, whereas the D-protein did not. This relative lack of responsiveness to the D-protein cannot be attributed to rapid excretion, since it is retained at least 4 times longer than the natural L-protein. These observations provide the first direct evidence that a folded D-amino acid protein has low immunogenicity and is long lived in vivo. Proteins with such properties may be useful as molecular platforms in a variety of chemical and pharmaco-logical applications. © 1993 Wiley-Liss, Inc.

    Notes: A significant fraction of the amino acids in proteins are alpha helical in conformation. Alpha helices in globular proteins are short, with an average length of about twelve residues, so that residues at the ends of helices make up an important fraction of all helical residues. In the middle of a helix, H-bonds connect the NH and CO groups of each residue to partners four residues along the chain. At the ends of a helix, the H-bond potential of the main chain remains unfulfilled, and helix capping interactions involving bonds from polar side chains to the NH or CO of the backbone have been proposed and detected. In a study of synthetic helical peptides, we have found that the sequence Ser-Glu-Asp-Glu stabilizes the alpha helix in a series of helical peptides with consensus sequences. Following the report by Harper and Rose, which identifies SerXaaXaaGlu as a member of a class of common motifs at the N termini of alpha helices in proteins that they refer to as “capping boxes,” we have reexamined the side chain-main chain interactions in a varient sequence using 1H NMR, and find that the postulated reciprocal side chain-backbone bonding between the first Ser and last Glu side chains and their peptide NH partners can be resolved: Deletion of two residues N terminal to the Ser-Glu-Asp-Glu sequence in these peptides has no effect on the initiation of helical structure, as defined by two-dimensional (2D) NMR experiments on this variant. Thus the capping box sequence Ser-Glu-Asp-Glu inhibits N terminal fraying of the N terminus of alpha helix in these peptides, and shows the side chain-main chain interactions proposed by Harper and Rose. It thus acts as a helix initiating signal. Since normal a helix cannot propagate beyond the N terminus of this structure, the box acts as a termination signal in this direction as well. © 1994 John Wiley & Sons, Inc.

    Notes: This article describes the implementation of a new docking approach. The method uses a Tabu search methodology to dock flexibly ligand molecules into rigid receptor structures. It uses an empirical objective function with a small number of physically based terms derived from fitting experimental binding affinities for crystallographic complexes. This means that docking energies produced by the searching algorithm provide direct estimates of the binding affinities of the ligands. The method has been tested on 50 ligand-receptor complexes for which the experimental binding affinity and binding geometry are known. All water molecules are removed from the structures and ligand molecules are minimized in vacuo before docking. The lowest energy geometry produced by the docking protocol is within 1.5 Å root-mean square of the experimental binding mode for 86% of the complexes. The lowest energies produced by the docking are in fair agreement with the known free energies of binding for the ligands. Proteins 33:367-382, 1998. © 1998 Wiley-Liss, Inc.
