Notes: Summary 1. The contractions of the isolated, electrically stimulated guinea-pig ileum during ganglionic blockade by Hexamethonium are inhibited by BAY 1470 [2-(2,6-Dimethylphenylamino)-4H-5,6-dihydro-1,3-thiazine], Morphine, Naphazoline, Isoprenaline, Noradrenaline and Adrenaline. These drugs are approximately equipotent in producing inhibition. 2. The inhibitory effects of Noradrenaline and Isoprenaline are diminished only by Propranolol and those of BAY 1470 and Naphazoline are abolished only by Phentolamine. The inhibitory effect of Adrenaline is diminished both by Propranolol and by Phentolamine. Neither Propranolol nor Phentolamine influence the Morphine induced effect. 3. Noradrenaline and Isoprenaline inhibit the electrically stimulated contractions and the effects of added Acetylcholine to the same extent. BAY 1470, Morphine and Naphazoline diminish only the effect of electrical stimulation and not that of Acetylcholine. Adrenaline inhibits both, but against the electrically induced contractions it is effective in lower concentrations than against the action of added Acetylcholine. 4. The results of these and of preceding investigations indicate that therelease of Acetylcholine from the parasympathetic nerve can be inhibited by stimulatinga-adrenergic, and theaction of Acetylcholine by stimulatingβ-adrenergic mechanisms.

Notes: Summary A new method is described which permits the determination of the biometrically defined relative combined activity of drug combinations. Combinations of substances with antihypertensive action were investigated on rats with experimental renal hypertension. The effects of the combinations α-methyldopa-guanacline and reserpine-guanacline were significantly more than additive, the effect of the combination α-methyldopa-reserpine was at most, additive.
