    Notes: Abstract By means of crossed immunoelectrophoresis of the cotyledonary storage proteins of Pisum sativum L. it was shown that reduced accumulation of the legumin fraction, resulting from severe sulphur deficiency during growth, is accompanied by relative suppression of a quantitatively minor storage protein (Peak 3) shown previously by subunit analysis to be related to the vicilin series of holoproteins. The pattern of isotopic labelling of the storage proteins after injection of [35S]methionine into the pedicel during seed development under normal nutritional conditions indicated that Peak-3 protein, like legumin, has a relatively high content of sulphur amino-acids. Like certain of the vicilin molecules carrying the determinants responsible for Peak-4, Peak-3 protein binds selectively to concanavalin A.

    Notes: Abstract A major poly(A)-containing RNA fraction of the approximate size expected of a monocistronic mRNA for the storage protein calliphorin has been isolated from the larval fat bodies of Calliphora vicina during early instar 3. This 20 S RNA fraction programs the synthesis by cell-free wheat embryo extracts of polypeptides of 86,000 daltons identified by tryptic peptide fingerprinting as precursors of the authentic calliphorin subunits of 83,000 daltons. Complementary DNA synthesized by AMV reverse transcriptase using the same 20 S RNA as template hybridized in situ to a single segment of one or two bands in the salivary polytene chromosomes of C. vicina.

    Notes: Summary Seven trained male cyclists ( $$\dot V_{O_{2max} } $$ =4.42±0.23 l·min−1; weight 71.7±2.7 kg, mean ± SE) completed two incremental cycling tests on the cycle ergometer for the estimation of the “individual anaerobic threshold” (IAT). The cyclists completed three more exercises in which the work rate incremented by the same protocol, but upon reaching selected work rates of approximately 40, 60 and 80% $$\dot V_{O_{2max} } $$ , the subjects cycled for 60 min or until exhaustion. In these constant load studies, blood lactate concentration was determined on arterialized venous ([La−]av) and deep venous blood ([La−]v) of the resting forearm. The av-v lactate gradient across the inactive forearm muscle was −0.08 mmol·l−1 at rest. After 3 min at each of the constant load work rates, the gradients were +0.05, +0.65* and +1.60* mmol·l−1 (*P〈0.05). The gradients after 10 min at these same work rates were −0.09, +0.24 and +1.03* mmol·l−1. For the two highest work rates taken together, the lactate gradient was less at 10 min than 3 min constant load exercise (P〈0.05). The [La−]av was consistently higher during prolonged exercise at both 60 and 80% $$\dot V_{O_{2max} } $$ than that observed at the same work rate during progressive exercise. At the highest work rate (at or above the IAT), time to exhaustion ranged from 3 to 36 min in the different subjects. These data showed that [La−] uptake across resting muscle continued to increase to work rates above the IAT. Further, the greater av-v lactate gradient at 3 min than 10 min constant load exercise supports the concept that inactive muscle might act as a passive sink for lactate in addition to a metabolic site.

    Notes: Summary In order to determine the influence of an extensive history of participation in high intensity activity on muscle fiber type, fiber size, and metabolic profile, elite ice hockey players were selected for investigation from three different leagues. Biopsy samples from the vastus lateralis muscle were obtained from different groups of players prior to and following the season and compared with control subjects. No significant differences were found in the percentage (49.6 vs. 43.8%) or the size of the ST fibers between the elite athletes and the control group, nor was there any significant alteration following the season of play in these variables. For the FT fiber subgroups, a reduction in the FTb (12.2 vs. 3.9%) and an increase in FTa (38.0 to 45.2%) fiber populations occurred over the season. Similarly, increases in fiber area were observed for both FT subgroups pre to post season. Of the enzymes studied only 3-hydroxyacyl CoA dehydrogenase was elevated in the post season measures, while total phosphorylase and phosphofructokinase were significantly lower. The metabolic pattern exhibited does not appear to be substantially different from what would be expected from an untrained group of similar fiber distribution.
