    Notes: Abstract Near-isogenic lines in conjunction with bulked segregant analysis were used to identify a DNA marker in wheat (Triticum aestivum L.) associated with the H21 gene conferring resistance to biotype L of Hessian fly [Mayetiola destructor (Say)] larvae. Near-isogenic lines were developed by backcross introgression BC3F3:4 (‘Coker 797’ * 4 / ‘Hamlet’) and differed by the presence or absence of H21 (on 2RL) derived from ‘Chaupon’ rye (Secale cereale L.). Bulked DNA samples were prepared from near-isogenic lines and BC3F2 population individuals segregating for reaction to Hessian fly biotype L and screened for random amplified polymorphic DNA markers using 46 10mer primers. Random-amplified polymorphic DNA markers from resistant and susceptible individuals and parental lines were scored and these data were used to identify a 3 kb DNA fragment that was related to the occurrence of H21. This fragment was amplified from DNA isolated from Hamlet, a near-isogenic line carrying 2RL, and bulked DNA from resistant BC3F2 individuals, but not from the recurrent parent Coker 797 or DNA bulks from susceptible BC3F2 plants. Analysis of 111 BC3F2 segregating individuals and BC3F2:3 segregants confirmed the co-segregation of the 3 kb DNA marker with the H21 resistance gene to Hessian fly. Use of this marker could facilitate more rapid screening of plant populations for Hessian fly resistance and monitoring the introgression of H21.

    Notes: Abstract Shoot length (cm), shoot fresh weight (g/pot), root length (cm), and root fresh weight (g/pot) were measured on six cultivars of wheat (Triticum aestivum L. cv Saluda, C9733, Gore, Stacy, FL301, and FL302) grown at pH 6.0, 5.5, 5.0, 4.5, or 4.0 for 14 days in ‘white quartz flintshot’ sand. Plants were watered on alternate days with pH-adjusted buffer solutions. All measured plant parameters decreased as H+ concentration increased from pH 6.0 to 4.0. Decreased lengths of shoots and roots were similar among the cultivars as the pH decreased. This indicated a uniform response of wheat cultivars to excess H+ concentration in the soil solution; however, the decrease in shoot and root length was only about 50% as large as was previously reported for sorghum [Sorghum bicolor (L.) Moench.].

    Notes: Abstract This investigation was performed to study the effect on plant water relations and growth when some of roots grow into dry soil. Common spring water (Triticum aestivum) plants were grown from seed in soil in 1.2 m long PVC (polyvinyl chloride) tubes. Some of the tubes had a PVC partition along their center so that plants developed a split root system (SPR). Part of the roots grew in fully irrigated soil on one side of the partition while the rest of the roots grew into a very dry (-4.1 MPa) soil on the other side of the partition. Split root plants were compared with plants grown from emergence on stored soil moisture (STOR) and with plants that were fully irrigated as needed (IRR). The experiment was duplicated over two temperature regimes (10°/20°C and 15°/25°C, night/day temperatures) in growth chambers. Data were collected on root dry matter distribution, soil moisture status, midday leaf water potential (LWP), leaf relative water content (RWC) and parameters of plant growth and yield. Some roots were found in the dry side of SPR already at 21 DAE (days after emergence) at a soil depth of 15 to 25 cm. Soil water potential around these roots was -0.7 to -1.0 MPa at midday, as compared with the initial value of -4.1 MPa. Therefore, water apparently flowed from the plant into the dry soil, probably during the night. Despite having most of their roots (around 2/3 of the total) in wet soil, SPR plants developed severe plant water stress, even in comparison with STOR plants. Already at 21 DAE, SPR plants had a LWP of -1.5 to -2.0 MPa, while IRR and STOR had a LWP of -0.5 MPa or higher. As a consequence of their greater plant water stress, SPR as compared with IRR plants were lower in tiller number, ear number, shoot dry matter, root dry matter, total biomass, plant height and grain yield and had more epicuticular wax on their leaves. It was concluded that the exposure of a relatively small part of a plant root system to a dry soil may result in a plant-to-soil water potential gradient which may cause severe plant water stress, leading to reduced plant growth and yield.

    Notes: A unique pathway that utilizes endoplasmic reticulum (ER) networks is proposed for screening pigment granule formation in the retina of adult Eumesosoma roeweri. Smooth endoplasmic reticulum (SER) functions to transport pigment particles to the formative site. Each site is composed of concentric, interconnected rings of SER that are filled with dense-cored, spherical pigment particles. Formation of the screening pigment granule begins by the release of particles from the innermost rings of carrier SER. Continued release followed by fusion and condensation of the pigment particles results in the formation of a mature pigment granule.

    Notes: In order to understand the evolutionary significance of sperm-pairing in American marsupials, an ultrastructural investigation was made of this process in the South American grey short-tailed opossum, Monodelphis domestica. One epididymis from each animal (5) was fixed for light and electron microscopy and divided into 18 segments. The contralateral tract was divided into similar segments and assessments made of the total number of spermatozoa and the proportion of sperm-pairs. The mean total sperm number was 4.20 ± 0.62 × 106/epididymis. Sperm-pairing commenced around segment 9 in the proximal corpus epididymidis and reached a maximum of 80% in the caudal sperm storage region of the duct. The sperm-pairing process was characterised by four stages. Spermatozoa exhibited parallel alignment as indicated by the positioning of identical cross-sections of sperm heads. This was followed by close apposition with acrosomal faces parallel rather than opposite. Rotation of the sperm heads around each other then apparently occurred as indicated by the morphological alignment of sections of paired sperm heads. Sperm-pairing was complete when the acrosomal faces were precisely aligned and joined. Misalignment and failure to pair was observed in about 20% of spermatozoa in the cauda epididymis. Such a complex sperm-pairing process may ensure that conjugated spermatozoa are precisely aligned so that flagella movement can be accurately coordinated for maximal progressive motility. © 1993 Wiley-Liss, Inc.

    Notes: A study was undertaken to examine, by scanning electron microscopy (SEM), the kidney glomeruli of control mice 1 mo, 10 mo, and 24 mo of age, as well as dietarily restricted mice 10 mo and 24 mo of age. One month old female C57BL/6J mice were offered one of the following: (1) a control diet containing 24% protein fed ad lib; (2) the control diet fed on alternate days (intermittently fed); or (3) a diet containing 4% protein fed ad lib. Animals were sacrificed, by aldehyde perfusion at 1 mo, 10 mo, and 24 mo of age. The kidneys were sliced and prepared for SEM.There was a significant age-related increase in glomerular diameter and amount of microvilli on the podocyte surface (microvillus index). Although the diameters of the podocytes increased approximately 20% with age, these differences were not statistically significant. Feeding a 4% protein diet resulted in smaller diameters of glomeruli and podocytes as well as smaller microvilli indices as compared to those of control animals. Although similar differences were observed in the kidneys of intermittently fed animals, only the microvillus index was statistically significant. Therefore, dietary manipulations, which have been shown to increase life span, result in marked morphological differences when compared to control animals.

    Notes: There is an increasing body of evidence showing that, for a variety of copolymers, there are significant changes in the copolymer composition over the molecular weight distribution of the polymer. In this work, we have polymerized the copolymer poly(styrene-methyl methacrylate) using ethylaluminum sesquichloride as the initiator. The copolymers produced were fractionated using a semiprep gel permeation chromatograph. The composition of the fractions was determined using infrared spectroscopy. Results show that the percent methyl methacrylate of the copolymers was higher at both the low- and high-molecular-weight regions of the polymers.
