    Notes: Heparin binds to bovine sperm and stimulates capacitation in vitro. Seminal plasma alters the ability of epididymal sperm to bind heparin, and several heparin-binding proteins (HBPs) have been identified in bull seminal plasma. This study had three objectives: (1) to identify production sites of seminal plasma HBPs, (2) to determine which HBPs bound to cauda epididymal sperm, and (3) to determine whether presence of HBPs was testosterone dependent. Proteins from bull or rat seminal vesicles, prostates, and bulbourethral glands were separated by heparin affinity high-performance liquid chromatography. HBPs were found in all accessory glands of rats and bulls, but the major source of bovine seminal plasma HBPs appeared to be seminal vesicles. Between 25% and 50% of the protein from each gland bound to the heparin column, and NaCl concentrations required to elute proteins ranged from 0.15 to 1.4 M. One-dimensional sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) showed that major HBPs were relatively small, with molecular weights between 13 and 31 kDa, but some HBPs also exhibited higher molecular weights, between 40 and 100 kDa. Radioiodinated HBPs from each bovine gland were incubated with epididymal sperm. Labeled HBPs binding to sperm exhibited molecular weights of 14, 16, 24, and 30 kDa as determined by SDS-PAGE and autoradiography. The HBP content of the accessory sex glands decreased significantly in castrated rats and was restored to levels of sham-operated controls by testosterone replacement. Heparin-binding proteins may play a role in fertilization by attaching to sperm surfaces, enabling heparin-like glycosaminoglycans in the female reproductive tract to induce capacitation.

    Notes: Rat pheochromocytoma PC12 cells respond to the binding of nerve growth factor (NGF) and basic fibroblast growth factor (bFGF) by extending neurites in a manner resembling sympathetic neurons. This response requires cell attachment to an appropriate substratum (Fujii et al., J. Neurosci., 2:1157, 1982); attachment factors which function in this capacity include the adhesive proteins fibronectin and laminin. Incubating PC12 cells with a ployclonal antiserum directed against a putative 140-kDa fibroblast cell surface fibronectin receptor (anti-gp140) perturbed spreading but not attachment of the cells to fibronectin and laminin substrates. However, in the presence of anti-gp 140 or its Fab fragments, NGF-stimulated neurite outgrowth was dramatically reduced. The antibody also caused a retraction of previously extended neurites. SDS-PAGE analysis of immunoprecipitates of PC12 cells surface labeled with 125I identified a prominent 120-140-kDa band, suggesting that the site of anti-gp 140 action in PC12 cells is also through a fibronectin receptor.

    Notes: Terminal deoxynucleotidyl transferase (TdT) is a template-independent DNA polymerase that is transiently expressed during the normal development of T and B lymphocytes. Phorbol 12-myristate 13-acetate (PMA) has been reported to induce maturation-like changes, including the loss of TdT, in many leukemic cell lines. We investigated the mechanism of TdT repression by PMA in an early thymocyte-like cell line, RPMI 8402. At a concentration of 8 nM, PMA caused both repression of TdT synthesis and arrest of proliferation. At greater concentrations of PMA, these same changes initially occurred, but then cell proliferation resumed, and TdT was reexpressed. At both 8 and 160 nM PMA, TdT biosynthesis and TdT mRNA became undetectable within 8 hours, while cell proliferation and DNA synthesis were not significantly reduced until 16 hours. Growth arrest induced by serum starvation did not result in a similar reduction of TdT RNA even after 48 hours. With 160 nM PMA, TdT mRNA could be detected again by 24 hours, and proliferation resumed. Transcription run-off assays indicated that TdT RNA synthesis ceased within 1 hour after exposure to both 8 and 160 nM PMA. T cell receptor α (TcR α) RNA was induced when TdT RNA was repressed. TcR β RNA levels were unchanged, and TcR γ RNA was up-regulated. TdT gene repression and modulation of cell proliferation as well as induction of TcR gene expression are normal events during intrathymic T cell maturation. This cell model provides a system for analyzing the molecular regulation of these significant developmental events.

    Description / Table of Contents: Study of Jarosite FormationThe reaction of solutions of ferric sulphate with potassium hydroxide yields Jarosite KFe3(OH)6(SO4)2 as the only compound detectable by X-rays, if the mole ratio b = KOH : Fe3+ is in the range 0.5 ≤ b ≤ 2.0 independent of the temperature of reaction. At first amorphous ferric oxide hydrate is precipitated in solutions with 1 ≤ b ≤ 2. This precipitate reacts with aquo-iron complexes of the solution in a way of a acid-base comproportionation and yields Jarosite. In solutions with b ≥ 2.5 the formation of Jarosite is impossible. By raising the temperature of reaction from 20 to 95°C the degree of reaction is increased, because the equilibria of the hydrolysis are shifted to the side of the hydroxo complexes.
    Notes: Beim Umsetzen von Eisen(III)-sulfatlösung mit Kalilauge erhält man beim Einhalten eines Molverhältnisses b = KOH:Fe3+ im Bereich 0,5 ≤ b ≤ 2,0 unabhängig von der Reaktionstemperatur die Verbindung Jarosit KFe3(OH)6(SO4)2 als einziges röntgenographisch nachweisbares Reaktionsprodukt. In Lösungen mit 1 ≤ b ≤ 2 fällt beim Zugeben der Kalilauge zunächst amorphes Eisenoxidhydrat aus, das im Verlaufe der weiteren Reaktion mit noch in Lösung befindlichen Eisen(III)-aquoionen im Sinne einer Säure-Basen-Komproportionierung zu Jarosit umgesetzt wird. Bei b ≥ 2,5 wird das Eisen quantitativ als Oxidhydrat gefällt und Jarosit nicht gebildet. Beim Verändern der Temperatur von 20 auf 95°C vergrößert sich der Umsetzungsgrad durch Verschieben der Hydrolysegleichgewichte auf die Seite der Hydroxokomplexe.

    Description / Table of Contents: From calorimetric and potentiometric measurements of equilibria between 3d ions and N-(o-carboxyphenyl)-iminodiacetic acid and N-2-carboxyethyl-iminodiacetic acid, respectively, we have determined the influence on the complex formation of a benzene ring being bound to a chelate ring. On the one hand the aromatic system modifies the qualities of the donor atoms by inductive and mesomeric interaction, and thus the ligand field stabilisation energy, and on the other hand it diminishes the mobility of the free ligand. These effects become evident in the ΔG, ΔH and ΔS values.
    Notes: Durch calorimetrische und potentiometrische Messungen an Lösungsgleichgewichten zwischen 3d-Ionen und Anthranilsäure-N,N-diessigsäure bzw. β-Alanin-N,N-diessigsäure konnte der Einfluß bestimmt werden, den ein Benzolring, der an ein Metallchelat gebunden ist, auf die Komplexbildung ausübt. Das aromatische System verändert einmal durch mesomere und induktive Wechselwirkungen die Donoreigenschaften der Haftatome und dadurch auch die Ligandenfeldstabilisierungsenergie und verringert andererseits die Beweglichkeit des freien Liganden. Diese Effekte kommen in den Reaktionsenthalpien und -entropien deutlich zum Ausdruck.

On the Formation of ZnFe2O4 from ZnO and α-Fe2O3Depending on the way of preparation α-Fe2O3 powder of comparable morphologic constitution show different behaviour in the reaction with ZnO. α-Fe2O3 prepared from geothite shows a high reactivity. The time dependence of the reaction rate follows a logarithmic law. The powder mixture reaction is compared with the data of the reaction of ZnO and α-Fe2O3 in a sandwich arrangement taken from the literature.
    Notes: α-Fe2O3-Pulver verhalten sich je nach der präparativen Vorgeschichte selbst bei vergleichbarer morphologischer Konstitution in der Reaktion mit ZnO unterschiedlich. α-Fe2O3 aus Goethit weist eine hohe Reaktivität auf. Der Reaktionsverlauf kann durch ein logarithmisches Zeitgesetz beschrieben werden. Die Umsetzung in der Pulvermischung wird mit dem zeitlichen Verlauf der Reaktion aneinandergefügter ZnO- und α-Fe2O3-Sinterpreßlinge nach Literaturangaben verglichen.

    Description / Table of Contents: N-(2-Carboxyphenyl)iminodiacetic acid (H3A) and N-(2,5-dicarboxyphenyl)iminodiacetic acid (H4B) are tetradentate ligands and form complexes of the composition MA- and MB2- with MII ions. These compounds differ by the additional charge of the second carboxylic group only, which is fixed to the benzene nucleus and which is unable for coordination for steric reasons.Using an anisothermal calorimeter ΔH values for the formation of the complexes MA- and MB2- in aqueous solution have been measured at an ionic strength 0.1 m KNO3. From these data, and from the stability constants of the complexes, entropy changes ΔS have been calculated.In all cases investigated (Mm+ = H+, Mg2+, Ca2+, Sr2+, Co2+, Ni2+, Cu2+, Zn2+) the ΔH values are more negative for the complexes MAm-3 than for complexes MBm-4, whereas the ΔS values are greater for complexes MBm-4. Using a simple model for the molecules of the complexes MBm-4 and empirically determined dielectric constants of the medium between the central ions and the noncoordinated ionized carboxylic group, the electrostatic attraction between these charges was calculated. Basing on these results the influence of the noncoordinated carboxylic group on the central atom by the mesomeric and inductive effect is discussed.
    Notes: Anthranilsäure-N,N-diessigsäure (H3A) und Aminoterephthalsäure-N,N-diessigsäure (H4B) bilden mit Erdalkalimetall- und zweiwertigen 3d-Ionen Komplexe der Zusammensetzung MeA- und MeB2-. Diese Verbindungen unterscheiden sich voneinander nur dadurch, daß in den Komplexen MeB2- die Carboxylatgruppe in 4-Stellung durch ihre Ladung auf die in der Koordinationssphäre gebundenen Zentralionen einmal direkt rein elektrostatisch, andererseits aber auch indirekt über das π-Elektronen-system des Benzols einen zusätzlichen Einfluß ausübt. Durch kalorimetrische Untersuchungen und durch Gleichgewichtsmessungen wurden ΔG, ΔH und ΔS für die Bildung dieser Komplexe in wäßriger Lösung bestimmt. In allen untersuchten Fällen (Mem+ = H+, Mg2+, Ca2+, Sr2+, Co2+, Ni2+, Cu2+, Zn2+) ist ΔH für die Bildung der Komplexe MeAm-3 stärker negativ als für MeBm-4, während letztere in jedem Falle entropiestabilisiert sind. Unter Verwendung eines einfachen Molekülmodells für die Komplexe sowie empirisch ermittelter Dielektrizitätskonstanten für das Medium zwischen den Zentralionen und der ionisierten Carboxylgruppe wurde die elektrostatische Anziehung zwischen diesen Ladungsträgern berechnet. Daraus konnte der Einfluß abgeschätzt werden, den die Carboxylatgruppe durch Mesomerie und Induktion auf die koordinierten Metallionen ausübt.
