    Description / Table of Contents: Zusammenfassung Ein 68jähriger Patient mit seit fünf Tagen bestehender Dyspnoe wurde nach nuklearmedizinischem Hinweis auf eine Lungenembolie im Stadium II–III einer Sonographie vor der geplanten Lysetherapie unterzogen. Dabei fand sich ein rechtsatrialer, flottierender Thrombus. Bei bestehender akuter vitaler Gefährdung wurde eine Ultrahochdosierte Streptokinasekurzzeitlyse (UHSK) mit 9.000.000 Einheiten Streptokinase über 6 Stunden und paralleler intravenöser Antikoagulation mit Heparin eingeleitet. Nach 3 Zyklen hatte sich der Transit-Thrombus vollständig und die Thrombose des Beines weitgehend aufgelöst. Diese Therapieform stellt eine interessante Alternative bei dem angesprochenen Krankheitsbild dar.
    Notes: Summary A 68-year-old patient with dyspnea since 5 days from pulmonary embolism documented by perfusion scintigraphy underwent examination with ultrasound before thrombolytic therapy. US showed a transit-thrombus in the right atrium. Because of threat to life a thrombolytic therapy was given with 9 million IE of streptokinase over 6 h (UHSK) and additional therapy with heparin. After three courses of UHSK the transit-thrombus and the thrombosis of the leg had nearly completely resolved. This mode of therapy is an interesting alternative in treatment for pulmonary embolism and transit-thrombus of the right heart.

    Notes: A transgenic mouse strain derived from embryonic stem (ES) cells infected with multiple copies of a retroviral vector carries a recessive insertional mutation resulting in prenatal lethality. A detailed histological analysis of developing embryos has shown that the mutation results in hyperplasia of both embryonic and extraembryonic ectoderm and failure of mesoderm formation in the egg cylinder stage embryo. The number of cells in each lineage of normal and mutant embryos was estimated using stereological analysis of serial sections taken from implantation sites. We observed a 2-fold increase in the number of embryonic ectoderm cells in mutant embryos at 7.5 days postcoitum (dpc). In addition, we found that mutant embryonic ectoderm cells are only 0.6 times as large as normal cells. The number of extraembryonic ectoderm cells in mutant embryos at 7.5 dpc is also increased, by almost 4-fold. Mutant extraembryonic ectoderm cells are also smaller than normal, being only two-thirds the size of wild-type cells. The mutant phenotype suggests that the gene identified by this insertional mutation plays an important role in the growth control of early embryonic lineages. © 1992 Wiley-Liss, Inc.

    Notes: In crude membrane fractions of rat pancreatic islets and of RIN-A2-cells, forskolin and NaF stimulated adenylate cyclase activity. Basal and stimulated enzyme activity was approximately 3 to 6 fold higher in membranes of RIN-A2-cells than in membranes of islet cells. In RIN-A2-cells GppNHp and NEM inhibited forskolin-stimulated cyclase system of RIN-A2-cells contains inhibitory and stimulatory N-proteins and that there are critical thiols related to Ni, Ns and/or the catalytic unit. Thus, membrane fractions of RIN-A2-cells may be an appropriate model for studies on the adenylate cyclase system of insulin-producing cells.
