    Notes: Insulin-like growth factors (IGF-I and IGF-II) play an important regulatory role in fetal growth and development. Alterations in expression of these growth factors may result in developmental abnormalities, macrosomia, and intrauterine growth retardation, which occur with a higher incidence in diabetic pregnancies. In situ hybridization histochemistry was employed to investigate the distribution and abundance of IGF-I and IGF-II in peri-implantation and postimplantation conceptuses from normal and streptozotocin-treated diabetic mice. Animals were sacrificed on gestational days 5, 6, 7, 8, and 9. The entire uterine horn was prepared for hybridization with antisense and sense α-35S-dATP labeled oligonucleotide probes for IGF-I, IGF-II, and mouse β-actin. IGF-I transcript was apparent only in myometrium at 6 days of gestation in normal and diabetic mice. IGF-II transcripts were restricted to trophoectoderm cells within the implantation chamber on day 5. Following implantation, IGF-II transcripts were found in trophoectodermal derivatives, primitive endoderm, mesoderm, heart, walls of the foregut, and mesenchyme in normal and diabetic postimplantation conceptuses. There were no apparent differences between normal and diabetic samples in the distribution and abundance of the IGF-II transcript from gestational days 7, 8, and 9. The embryos from the diabetic mother at day 6 were growth retarded and had a significant decrease in the expression of IGF-II. These results suggest that maternal hyperglycemia may retard development of the early implanting conceptus in a narrow window around day 6 through a mechanism involving decreased IGF-II expression. Fetuses from diabetic pregnancies that escape this critical period appear to develop and express IGF-II in an equivalent manner to those of the control group. © 1994 Wiley-Liss, Inc.

    Notes: Arginine, cystine, histidine, leucine and threonine were needed for outgrowth of the mouse blastocyst in vitro. Omission of lysine, methionine, phenylalanine, tryptophane and tyrosine from the culture medium markedly reduced blastocyst outgrowth, but did not inhibit it completely; while omission of isolecine and valine reduced the extent of outgrowth only slightly. Blastocysts kept for seven days in a free-floating condition by omitting arginine and leucine from the medium, grew out when these amino acids were added. Such behavior may be analogous to delayed implantation in utero and suggests that the free amino acid content of the uterus could be an important factor in the control of implantation. Blastocysts delayed from implanting in the uterus by ovariectomy were activated to outgrowth in a complete medium, but the intracellular changes associated with outgrowth occurred more slowly than in undelayed blastocysts.

    Notes: Hamster and bovine zonae pellucidae were dissolved by heating them in buffered saline for 35 min at 65°C and 80°C, respectively. Pretreatment of noncapacitated sperm with these solutions did not affect their binding to eggs, but the binding of cumulus-capacitated hamster sperm and of bovine sperm that had undergone a capacitation-like surface change in response to hypertonic-treatment was severely curtailed. These experiments demonstrate that such zona solutions exhibit receptor activity, but only for the capacitated sperm.

    Notes: Bovine zonae were solubilized by heating to 80°C in buffered saline. On injection of female rabbits with this solution, zona-specific antisera were produced that could be assayed on bovine eggs in vitro by immunofluorescence, coating, and inhibition of sperm-binding. When the latter was complete, the rabbits were found to be infertile. Immunofluorescence assays showed that the rabbit anti-bovine zona serum reacted as strongly with rabbit and rhesus monkey zonae as with homologous zonae. Strong cross-reaction was also observed with marmoset and dog zonae, but with hamster zonae the reaction was relatively weak.

    Notes: Labeling of the zona pellucida of cow blastocysts with zona-specific anti-serum shows that antigenicity is unaffected by abnormal cleavage, in vitro culture, or frozen storage. The uniform labeling in thin sections indicates that the zona pellucida is homogeneous antigenically. Heavier labeling of the inner and outer surfaces of the zona pellucida in thick sections appers to be due to greater porosity of these regions, in which the zona material becomes highly dispersed, or even partly solubilized, thereby permitting the formation of an antigen-antibody matrix.

    Notes: Treatment of bovine ovary sections with rabbit antibovine zona serum followed by fluorescein-conjugated goat antirabbit IgG produced a specific fluorescent staining only of the zonae pellucidae. Fluorescence was greater near the inner and outer surface of the zona, suggesting that either the same antigen occurs in higher concentration in these regions or that there is more than one antigen, the most immunogenic being located peripherally. In some atretic follicles fluorescent material appeared to diffuse into the degenerating oocyte and into the intercellular spaces of the cumulus oophorus.

    Notes: Zonae pellucidae were collected from bovine ovaries by chopping, dispersing the chopped tissue with collagenase, sieving through nylon mesh screens, and pipetting. The zonae were free of corona cell processes when examined under the scanning electron microscope. Solutions of zonae obtained with collagenase exhibited the same antigenic and sperm receptor properties as those obtained without enzyme treatment.

    Notes: A procedure is described for isolating milligram quantities of bovine and porcine zonae pellucidae, uncontaminated by follicle cells or their processes. On SDS-polyacrylamide gel electrophoresis the isolated bovine zona material formed one major glycoprotein band with an estimated molecular weight of approximately 100,000 daltons and two minor lower molecular weight components. The isolated pig zonae formed only one glycoprotein band with a molecular weight of approximately 62,000 daltons. Rabbit antisera raised against the isolated zonae were zona-specific and formed only a single precipitin line against the heat-solubilized zonae on immunoelectrophoresis. An adjuvant was not required for high-antibody titers. High titers were also obtained by injection of the dog and rhesus monkey. Anti-zona antibody was detected by immunofluorescence, zona-coating, double-immunodiffusion, and the inhibition of spermbinding to eggs, including those of human origin. Antigenic and sperm receptor properties were stable at 100°C for five minutes, but some activity was lost after longer exposure. The serum antibody produced by rabbits immunized with pig zonae was predominantly IgG and cross-reacted with the zonae of a variety of other species, including primates. Pregnancy was inhibited in female rabbits immunized with pig zona preparations.

    Notes: Hamster oocytes were frozen using a 1,2-propanediol-sucrose procedure, which resulted in over 90% survival. After thawing and zona removal the oocytes were compared with non-frozen oocytes in a zona-free hamster egg test employing spermatozoa from human semen donors and suspected infertility patients. Similar data were obtained, indicating that propancdiol-sucrose frozen hamster eggs may be used in place of fresh eggs for convenience and to a void scheduling problems.
