    Notes: A special type of cell, called ‘Lichtzellen’ by Hesse, was found in the photosensitive regions of the earthworm. Wherever the epidermis is most sensitive to light these cells are most abundant. They are found in the epidermis of all segments of the body and also in nerve enlargements of the prostomium and the caudal segment, but the intersegmental and ventral regions of the different segments, except the more distal ones, contain none of them. They are supplied by nerves and each contains a characteristic inner structure, the optic organelle, composed of a large central hyaline structure, the lens, which is surrounded by a dense network of nerve fibrillae, the retinella. In hanging drops the lens was found to focus light in the region of the retinella irrespective of the direction of the rays.These cells are similar in structure and function to the visual cells in leeches. Available data indicate that these cells function as photoreceptors and that the fibrillae of the retinella are the direct receptors of light stimuli.Pigment is not associated with the photoreceptors in a way that suggests direct functional relationship, but there is a subepidermal pigment layer through which pinhole windows admit light along the path of nerves to each of the nerve enlargements containing photoreceptor cells. These windows open in such directions as to determine the direction of withdrawal of the worm.

    Notes: Epididymal sperm from the painted turtle (Chrysemys picta) possess a cytoplasmic droplet which is located eccentrically on the sperm midpiece. The droplet contains a large quantity of lipid droplets in addition to hollow vesicles and degenerate mitochondrial fragments. Lipid droplets are closely associated with mitochondrial membranes and may function in the formation or degradation of mitochondria. Cytoplasmic droplets become detached from the sperm midpiece in a coordinated manner shortly before the commencement of fall mating and are not observed on sperm recovered from the oviduct of females. © 1992 Wiley-Liss, Inc.

    Notes: The pleiotropic nature of insulin action suggests diverse mechanisms of signal transduction for the hormone. The specific protein phosphatase inhibitor, okadaic acid, is utilized to differentiate metabolic pathways that may be regulated by phosphorylation or dephosphorylation of key enzymes. In H-35 hepatoma cells, okadaic acid inhibits insulin-stimulated glycogen synthesis with an IC50 of 400 nM. In contrast, activation of lipogenesis by insulin is inhibited with an IC50 of 50 nM okadaic acid. The toxin also inhibits stimulation of lipogenesis in these cells by the insulin-sensitive inositol glycan enzyme modulator. In isolated rat adipocytes, insulin-stimulated lipogenesis is also inhibited by okadaic acid with an IC50 of approximately 1,700 nM. The antilipolytic effect of insulin in these cells is more sensitive to okadaic acid, exhibiting an IC50 of 150 nM. Maximal activation of lipogenesis by insulin is dramatically reduced by okadaic acid with no effect on the concentration required for half-maximal activation, whereas the sensitivity of insulin-induced antilipolysis is attenuated by okadaic acid, with no apparent reduction in the maximal effect of the hormone. Taken together, these data suggest that specific phosphatases may be differentially involved in some of the metabolic pathways regulated by insulin.

    Notes: Background: The testes of rats treated neonatally with propylthiouracil (PTU) grow to almost twice their normal size. The cause of testicular enlargement has been suggested to be the result of delayed maturation of Sertoli cells, allowing Sertoli cell division to occur beyond the 15th postnatal day, the commonly recognized cutoff date for Sertoli cell divisions. It has been shown that an increased population of Sertoli cells in postnatal development supports increased numbers of germ cells in adult animals. After examining developing rats treated neonatally with PTU, we hypothesized that an approximate 10-day delay in maturation was occurring and proceeded to test this hypothesis experimentally. Thus the purpose of this report was to determine if a 10-day delay in maturation could explain the increased numbers of Sertoli cells and increased testis size in PTU-treated animals.Methods: Both control animals and animals treated neonatally with PTU N = 5/group were sacrificed at 15 and 25 days of age and prepared for electron microscopy.Results: Micrographs show and morphometric ultrastructural analysis of numerous parameters demonstrated at the 95% probability level that Sertoli cells from 25-day-old PTU animals are not different in size and most constituents (volume and surface area) from 15-day-old control animals and are less mature than 25-day-old control animals. Mitosis of Sertoli cells was observed in PTU-treated animals in 25-day-old animals but not in agematched controls. The number of Sertoli cells in 25-day-old PTU-treated animals is significantly increased over age-matched controls. Micrographs show the presence of immature Sertoli cell nuclei in 25-day-old animals receiving PTU as well as increased germ cell degeneration in this group. Sertoli cell tight junction formation is also delayed in PTU-treated animals as compared with controls.Conclusions: Together, the data show that delayed maturation of Sertoli cells occurs in treated animals that corresponds to a minimum of 10 developmental days. In the immature state, Sertoli cells continue to divide. Data presented herein and published data related to PTU treatment indicate that delayed maturation of the Sertoli cell results in delayed maturation and proliferation of other testicular cell types. From this and from published data, the hypothesis is presented that the Sertoli cell is responsible for the overall control of testis development. © 1995 Wiley-Liss, Inc.
