    Notes: Abstract  This study investigated regulatory volume increase (RVI) in rat pancreatic β-cells. Volume changes in isolated β-cells were measured by a video-imaging method. Cell shrinkage was induced by exposure to solutions made hypertonic by the addition of 100 mM mannitol. In HEPES-buffered solutions, β-cells exhibited an RVI which was almost completely abolished by 10 μM bumetanide. These data indicate that Na+-2Cl–-K+ cotransporters make a major contribution to RVI in β-cells. In HCO3 –-buffered solutions, however, an RVI was observed in the presence of 10 μM bumetanide. This bumetanide-insensitive component of RVI was inhibited by 100 μM amiloride or 100 μM 4,4’-diisothiocyanatostilbene-2,2’-disulphonic acid (DIDS). These data suggest that, in addition to the Na+-2Cl–-K+ cotransporter, functionally coupled Na+-H+ exchangers and Cl–-HCO3 – exchangers may also contribute to RVI in pancreatic β-cells.

    Notes: Abstract The volumes of acinar cells isolated from rat lacrimal gland were measured on computer by video-imaging. Cells were found to swell on exposure to hypotonic solutions; they subsequently exhibited a regulatory volume decrease (RVD). RVD was inhibited in the absence of extracellular Ca2+, and by the K+ channel blocker tetraethylammonium chloride (2 mm TEA+). The possible involvement of K+ channels in RVD was further investigated in cell-attached patches. Exposing the cells to a hypotonic solution activated channels with a conductance of 141±6 pS (n=11). These channels were partially blocked by 0.5 mm TEA+, and channel activation was not observed in the absence of extracellular Ca2+. Experiments in the inside-out patch configuration demonstrated that the channels activated by hypotonie stress were “maxi” Ca2+-activated K+ channels. It is concluded that the opening of these channels plays an important role in RVD, by facilitating K+ loss from the cell.

    Notes: A series of polymers, bisphenol A-based poly(phosphoesters), were evaluated as degradable biomaterials. Degradation was observed for the four polymers studied under both in vitro and in vivo conditions. The rate of degradation was affected by polymer side-chain structure and correlated with the swelling behavior. The ethyl side-chain polymers absorbed more water than their phenyl counterparts. Among the sterilization methods, UV irradiation followed by antibiotic treatment was the most suitable, as steam autoclave and ethylene oxide treatments altered the properties of several of the poly(phosphoesters). Tissue response to the poly(phosphoesters) in rabbits was characterized by minor encapsulation and slight or no lymphocyte, giant cell, or macrophage activity. No evidence of edema or necrosis was found. The elastic moduli of these materials varied from 488 MPa for poly(bisphenol A-ethylphosphate) (BPA/EOP) to 627 MPa for the more rigid poly(bisphenol A-phenylphosphonate) (BPA/PP). The ultimate strength, modulus, and energy to failure of BPA/PP were lower than those of similarly compression molded high-molecular-weight poly(L-lactic acid) (PLLA).

    Notes: Although there is interest in forming synthetic analogs of hard tissues at physiologic temperature, significant gaps in knowledge exist with respect to the mechanisms by which precursor solids convert to apatites and also with respect to the apatite compositions that may be formed. In this study calcium-deficient HAp [Ca9(HPO4)(PO4)5OH] was prepared by hydrolysis of tricalcium phosphate (TCP), α-Ca3(PO4)2. The kinetics of HAp formation were studied as a function of temperature by isothermal calorimetry. TCP hydrolyzed completely within about 12 h, and the hydrolysis reaction evolved 133 kJ/mol of HAp formed. Although the kinetics of hydrolysis exhibited a strong temperature dependence, the mechanistic path taken appeared independent of temperature. The fluoridation of hydroxyapatite compositions having Ca/P ratios higher than 1.59 previously has been investigated. However, little work has been done on the fluoridation of more calcium-deficient hydroxyapatite. Ca9(HPO4)(PO4)5OH was formed at temperatures between 37.4° and 55°C to vary its morphology. These preparations then were reacted in NaF solution and the kinetics of fluoride incorporation studied. Solution chemical analyses were used to determine the amounts of fluoride incorporated. The extent of hydroxyl replacement by fluoride ranged from 17 to 72% and correlated with the surface area of the parent HAp. © 1998 John Wiley & Sons, Inc. J Biomed Mater Res, 42, 458-464, 1998.

    Notes: Hydrogen migrations accompany many mass spectrometric fragmentation processes and a detailed study of these migrations, largely by means of deuterium labeling, has shed much light on electron-impact induced bond fissions. More recently, it has been found that groups other than hydrogen, such as alkyl, aryl, hydroxyl, etc., also can migrate after electron bombardment and the present article is concerned with a discussion of such molecular rearrangements among organic compounds. These observations are not only of intrinsic mechanistic interest, but a detailed knowledge of the occurrence of such migration reaction is crucial to the proper interpretation of mass spectra and to the avoidance of misinterpretations in “element mapping”.
